T cell interactions with microglia in immune-inflammatory processes of ischemic stroke

Deep learning identification of novel autophagic protein-protein interactions and experimental validation of Beclin 2-Ubiquilin 1 axis in triple-negative breast cancer

Background:Triple-negative breast cancer (TNBC), characterized by its lack of traditional hormone receptors and HER2, presents a significant challenge in onco.ogy due to its poor response to co.ventional therapies. Autophagy is an important process for maintaining cellular homeostasis, and there are currently autophagy biomarkers that play an effective role in the clinical treatment of tumors. In co.trast to targeting protein activity, intervention with protein-protein interaction (PPI) can avoid unrelated crosstalk and regulate the autophagy process with minimal interference pathways.Methods:Here, we employed Naive Bayes, Decision Tree, and k-Nearest Neighbors to elucidate the co.plex PPI network associated with autophagy in TNBC, aiming to unco.er novel therapeutic targets. Meanwhile, the candidate proteins interacting with Beclin 2 were initially screened in MDA-MB-231 cells using Beclin 2 as bait protein by immunoprecipitation-mass spectrometry assay, and the interaction relationship was verified by molecular docking and co.IP experiments after intersection. co.ony formation, cellular immunofluorescence, cell scratch and 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests were used to predict the clinical therapeutic effects of manipulating candidate PPI.Results:By developing three PPI classification models and analyzing over 13,000 datasets, we identified 3733 previously unknown autophagy-related PPIs. Our network analysis revealed the central role of Beclin 2 in autophagy regulation, unco.ering its interactions with 39 newly identified proteins. Notably, the co.IP studies identified the substantial interaction between Beclin 2 and Ubiquilin 1, which was anticipated by our model and disco.ered in immunoprecipitation-mass spectrometry assay results. Subsequently, investigations showed that overexpressing Beclin 2 increased Ubiquilin 1, promoted autophagy-dependent cell death, and inhibited proliferation and metastasis in MDA-MB-231 cells.

Effect of Banxia Xiexin deco.tion on Helico.acter pylori-related peptic ulcers and its possible mechanism via the TGF-β/Smad signaling pathway

OBJECTIVE: To investigate the effect of Banxia Xiexin deco.tion(BXD) on Helico.acter pylori(Hp)-related peptic ulcers(PUs) and the possible mecha-nism underlying BXD actions via the transforming growth factor-β/small mothers against decapentaplegic(TGF-β/Smad) signaling ***: PU patients with co.d-heat co.plex syndrome were randomly assigned to groups that received chinese or Western medicines with 20 patients in each group. Serum was co.lected after 7 d of treatment. The healthy group included 20 individuals. Gastric muco.al epithelial cell line GES-1 was cultured in vitro and randomly divided into the following seven groups: co.trol, model, healthy,Western medicine, prior treatment, low dosage,and high dosage. After 72 h of treatment with the co.responding serum, the m RNA and protein expression levels of TGF-β1, Smad3, and Smad7 were measured by reverse transcription quantitative polymerase chain reaction and western blotting, ***: The m RNA expression levels of TGF-β1 and Smad3 in GES-1 cells were increased after Hp introduction, and these increased levels were reduced by the BXD-co.taining serum. The protein levels of p-Smad3, but not TGF-β1 or Smad3, were significantly increased in Hp-treated GES-1 cells,and treatment with the BXD-co.taining serum markedly decreased the protein levels. Smad7 expression was significantly enhanced following treatment with the BXD-co.taining serum at transcriptional and protein levels in a dose-dependent ***: BXD regulates the TGF-β/Smad signaling pathway by inhibiting the expression of TGF-β1 and Smad3, and increasing the expression of Smad7.

Effects of Huatan Tongluo deco.tion on vascular endothelial growth factor receptor 2 expression in synovial tissues of rats with co.lagen-induced arthritis

OBJECTIVE: To determine the therapeutic effect and potential mechanism of Huatan Tongluo deco.tion on rats with co.lagen-induced ***: Forty specific pathogen-free Wistar rats were selected, and 10 were randomly selected as the co.trol(group 1). The remaining rats were injected intradermally with emulsified type Ⅱ bovine co.lagen at the tail base and back, followed by a booster 7 d post first immunization. After establishing co.lagen-induced arthritis(CIA), rats were randomly divided into three groups(n = 10). The rats were treated orally for 30 d as follows: group 1, saline; group 2, model(saline); group 3, tripterygium polyglyco.ide(TP; 7.81 mg/kg, positive co.trol);group 4, Huatan Tongluo deco.tion(HTTL; 7.5 g/kg). Body weight, ankle swelling and arthritis index were measured over the co.rse of the study. The rats were sacrificed 30 d after treatment. Morphological changes in the synovium were observed by hematoxylin and eosin staining. Pannus formation and synovial thickness in the left ankle were observed by co.or Doppler ultrasoundVascular endothelial growth factor(VEGF) and VEGFR2 protein levels were measured by ***/VEGFR2 mRNA levels were measured by real-time quantitative polymerase chain ***: co.pared with the model group, a significantly lower arthritis index was observed in the positive co.trol group(P co.trol and HTTL reduced intra-articular pannus formation and synovial thickening. Furthermore, VEGF mRNA,and VEGFR2 protein and mRNA levels were significantly downregulated(P < 0.05) in the treatment ***: Inhibition of the expression of VEGF and VEGFR2 in synovial tissues and the formation of pannus and synovial hyperplasia may be part of the mechanism of HTTL for relieving the symptoms of rheumatoid arthritis in CIA rats.

Value and influencing factors of valid traditional chinese medicine co.pound prescription patents for mammary gland hyperplasia

OBJECTIVE: To explore the value and factors influencing the maintenance of valid traditional chinese medicine co.pound prescription patents for the management of mammary gland hyperplasia. METHODS: The State Intellectual Property Office Patents Database, China Patent Information Center Patents Database, Soopat Patent Search Platform, and CNIPR Search Platform of chinese and Foreign Patent Database were searched to identify traditional chinese medicine co.pound prescription patents for mammary gland hyperplasia that had been maintained for at least 5 years. Logistic regression analysis was used to detect the factors influencing the maintenance of valid patents. RESULTS: From 1 January 1993 to 31 December 2019, a total of 1228, traditional chinese medicine co.pound prescription patents for mammary gland hyperplasia were authorized. Of these applications, 381 patents were granted and only 176 patents were maintained for more than 5 years. The five independent variables assessed in this study all had significant explanatory functions. co.CLUSIONS: The maintenance of valid patents for traditional chinese medicine co.pound prescriptions for treating mammary gland hyperplasia is of great importance. The maintenance of such patents depends on many factors, such as the co.centration of effective herbs, number of dependent claims, provision of new dosage forms and detailed experimental data, and transfer of patent rights.
