TPX2 knockdown suppressed hepatocellular carcinoma cell invasion via inactivating AKT signaling and inhibiting MMP2 and MMP9 expression

Objective： Targeting protein for Xenopus kinesin-like protein 2 （TPX2） is a nuclear proliferation-related protein that plays a critical role in the formation of mitotic spindle. High expression of TPX2 has been observed in several types of tumors. However, the role of TPX2 in hepatocellular carcinoma （HCC） remains unclear. Our study aimed to investigate the effect of TPX2 on HCC cell invasion. Methods： The immortalized normal human liver cell line L02 and six HCC cell lines including SMMC- 7721, BEL-7402, Huh-7, HepG2, Hep3B and SKHepl were subjected to qRT-PCR and western blot for TPX2 mRNA and protein, respectively. Furthermore, TPX2 small interfering RNA （siRNA） was used to knock down TPX2 expression in SMMC-7721 and HepG2 cells. Cell proliferation and invasion were determined by MTT and transwell assays. Otherwise, expression of p-AKT, MMP2 and MMP9 were evaluated by western blot in SMMC-7721 cells. Results： The expression of TPX2 in HCC cell lines was markedly higher than that in normal human liver cell line. TPX2 knockdown using a specific TPX2-siP, NA reduced the number of invaded cells and inhibited cell proliferation in SMMC-7721 and HepG2 cells. Furthermore, TPX2 knockdown resulted in inactivation of AKT signaling and down-regulation of MMP2 and MMP9 expression in SMMC-7721 cells. Conclusions： Our study identified that TPX2 might contribute to tumor cell invasion through activating AKT signaling and subsequently increasing MMP2 and MMP9 in HCC.

Objective: To evaluate the impacts of rosiglitazone and metformin on ovarian response, hirsutism and insulin action in women with polycystic ovary syndrome (PCOS). Methods: Ninety women resistant to clomiphene citrate with PCOS were randomized, 40 cases to rosiglitazone group, 50 cases to meformin group. Rosiglitazone and metformon were administered for 6 months in combination with clomiphene citrate on cycle day 5th to 9th, respectively. The clinical evaluations were performed monthly. Reproductive hormone, serum glucose and insulin levels were observed before and after treatment. The data were analyzed using repeated analysis of variance (ANOVA). Results: There were significant changes in reducing hirsutism score, serum testosterone level, LH/FSH ratio, and restoration of ovarian ovulation after metformin or rosiglitazone administration respectively (P0.05). Conclusion: Rosiglitazone can increase insulin sensitivity. Metformon may reduce BMI. They all restore regular menstrual cycles, increase pregnancy rate, and reduce testosterone and LH concentration in women with polycystic ovary syndrome.

We present a bidirectional reflection distribution function （BRDF） model for thermal coating surfaces based on a three-component reflection assumption, in which the specular reflection is given according to the microfacet theory and Snell＇s law, the multiple reflection is considered Nth cosine distributed, and the volume scattering is uniformly distributed in reflection angles according to the experimental results. This model describes the reflection characteristics of thermal coating surfaces more completely and reasonably. Simulation and measurement results of two thermal coating samples SR107 and S781 are given to validate that this three-component model significantly improves the modeling accuracy for thermal coating surfaces compared with the existing BRDF models.

Frequency domain dielectric spectroscopy(FDS)is an effective method reflecting the changes of ageing status of silicone oil‐immersed insulation *** better evaluating the ageing status of silicone oil‐immersed insulation paper,an evaluation method was studied by analysing the ageing kinetic model and results of ***,the method to separate the different polarization losses and conductivity loss from the results of FDS was ***,based on the idea of ageing correction translation coefficient and timevarying pre exponential factor,the zero order ageing dynamic model was modified,which combines the ageing kinetic model with the results of ***,the evaluation method for the degree of polymerisation(DP)of silicone oil‐immersed paper was proposed,and experiments were conducted to obtain the parameters in the modified model and then to verify the proposed *** results show that the proposed method can be used to evaluate the DP effectively.

A 14-bit 100-MS/s pipelined analog-to-digital converter（ADC） without dedicated front-end sampleand-hold amplifier（SHA） is presented. In addition to elaborate matching of the sampling network in the first stage,a background offset cancellation circuit is proposed in this paper to suppress the offset of the comparators in the first-stage sub-ADC, which ensures the overall offset does not exceed the correction range of the built-in redundant structure. Fabricated in a 0.18- m CMOS technology, the presented ADC occupies a chip area of 12 mm2, and consumes 237 mW from a 1.8-V power supply. Measurement results with a 30.1-MHz input sine wave under a sampling rate of 100 MS/s show that the ADC achieves a 71-d B signal-to-noise and distortion ratio（SNDR）,an 85.4-d B spurious-free dynamic range（SFDR）, a maximum differential nonlinearity（DNL） of 0.22 LSB and a maximum integral nonlinearity（INL） of 1.4 LSB.

Objective To investigate the independent risk factors of traumatic brain injury (TBI) prognosis. Methods A retrospective analysis was performed in 885 hospitalized TBI patients from January 1, 2003 to January 1, 2010 in the First Affiliated Hospital of Medical College of xi'an jiaotong university. Single-factor and logistic regression analysis were conducted to evaluate the association of different variables with TBI outcome. Results The single-factor analysis revealed significant association between several vari-ables and TBI outcome, including age (P=0.044 for the age group 40-60, P<0.001 for the age group ≥60), complications (P<0.001), cerebrospinal fluid leakage (P<0.001), Glasgow Coma Scale (GCS) (P<0.001), pupillary light reflex (P<0.001), shock (P<0.001), associated ex-tra-cranial lesions (P=0.01), subdural hematoma (P<0.001), cerebral contusion (P<0.001), diffuse axonal injury (P<0.001), and subarachnoid hemorrhage (P<0.001), suggesting the in-fluence of those factors on the prognosis of TBI. Furthermore, logistic regression analysis iden-tified age, GCS score, pupillary light reflex, subdural hematoma, and subarachnoid hemorrhage as independent risk factors of TBI prognosis. Conclusion Age, GCS score, papillary light reflex, subdural hematoma, and subarachnoid hemorrhage may be risk factors influencing the prognosis of TBI. Paying attention to those fac-tors might improve the outcome of TBI in clinical treatment.

Catalytic C−H bond activation is one of the backbones of the chemical *** metal subnanoclusters consisting of a few atoms have shown attractive properties for heterogeneous ***,the creation of such catalyst systems with high activity and excellent anti-sintering ability remains a grand ***,we report on alkali ion-promoted Pd subnanoclusters supported over defectiveγ-Al_(2)O_(3) nanosheets,which display exceptional catalytic activity for C−H bond activation in the benzene oxidation *** presence of Pd subnanoclusters is verified by aberration-corrected high-angle annular dark-field scanning transmission electron microscopy,X-ray absorption spectroscopy,and X-ray photoelectron *** catalyst shows excellent catalytic activity,with a turnover frequency of 280 h^(−1) and yield of 98%,in benzene oxidation reaction to give phenol under mild ***,the introduction of alkali ion greatly retards the diffusion and migration of metal atoms when tested under high-temperature sintering *** functional theory(DFT)calculations reveal that the addition of alkali ion to Pd nanoclusters can significantly impact the catalyst’s structure and electronic properties,and eventually promote its activity and *** work sheds light on the facile and scalable synthesis of highly active and stable catalyst systems with alkali additives for industrially important reactions.
