Prohibitin 1 inhibits cell proliferation and induces apoptosis via the p53-mediated mitochondrial pathway in vitro

BACKGROUND Prohibitin 1(PHB1)has been identified as an antiproliferative protein that is highly conserved and ubiquitously expressed,and it participates in a variety of essential cellular functions,including apoptosis,cell cycle regulation,prolifera-tion,and survival.Emerging evidence indicates that PHB1 may play an important role in the progression of hepatocellular carcinoma(HCC).However,the role of PHB1 in HCC is controversial.AIM To investigate the effects of PHB1 on the proliferation and apoptosis of human HCC cells and the relevant mechanisms in vitro.METHODS HCC patients and healthy individuals were enrolled in this study according to the inclusion and exclusion criteria;then,PHB1 levels in the sera and liver tissues of these participates were determined using ELISA,RT-PCR,and immunohistoche-mistry.Human HepG2 and SMMC-7721 cells were transfected with the pEGFP-PHB1 plasmid and PHB1-specific shRNA(shRNA-PHB1)for 24-72 h.Cell prolif-eration was analysed with an MTT assay.Cell cycle progression and apoptosis were analysed using flow cytometry(FACS).The mRNA and protein expression levels of the cell cycle-related molecules p21,Cyclin A2,Cyclin E1,and CDK2 and the cell apoptosis-related molecules cytochrome C(Cyt C),p53,Bcl-2,Bax,caspase 3,and caspase 9 were measured by real-time PCR and Western blot,respectively.RESULTS Decreased levels of PHB1 were found in the sera and liver tissues of HCC patients compared to those of healthy individuals,and decreased PHB1 was positively correlated with low differentiation,TNM stage III-IV,and alpha-fetoprotein≥400μg/L.Overexpression of PHB1 significantly inhibited human HCC cell proliferation in a time-dependent manner.FACS revealed that the overexpression of PHB1 arrested HCC cells in the G0/G1 phase of the cell cycle and induced apoptosis.The proportion of cells in the G0/G1 phase was significantly increased and the proportion of cells in the S phase was decreased in HepG2 cells that were transfected with pEGFP-PHB1 compared with untreated control and empty vector-trans

BACKGROUND The role of Sm-like 5(LSM5)in colon cancer has not been determined.In this study,we investigated the role of LSM5 in progression of colon cancer and the potential underlying mechanism involved.AIM To determine the role of LSM5 in the progression of colon cancer and the potential underlying mechanism involved.METHODS The Gene Expression Profiling Interactive Analysis database and the Human Protein Atlas website were used for LSM5 expression analysis and prognosis analysis.Real-time quantitative polymerase chain reaction and Western blotting were utilized to detect the expression of mRNAs and proteins.A lentivirus targeting LSM5 was constructed and transfected into colon cancer cells to silence LSM5 expression.Proliferation and apoptosis assays were also conducted to evaluate the growth of the colon cancer cells.Human GeneChip assay and bioinformatics analysis were performed to identify the potential underlying mechanism of LSM5 in colon cancer.RESULTS LSM5 was highly expressed in tumor tissue and colon cancer cells.A high expression level of LSM5 was related to poor prognosis in patients with colon cancer.Knockdown of LSM5 suppressed proliferation and promoted apoptosis in colon cancer cells.Silencing of LSM5 also facilitates the expression of p53,cyclin-dependent kinase inhibitor 1A(CDKN1A)and tumor necrosis factor receptor superfamily 10B(TNFRSF10B).The inhibitory effect of LSM5 knockdown on the growth of colon cancer cells was associated with the upregulation of p53,CDKN1A and TNFRSF10B.CONCLUSION LSM5 knockdown inhibited the proliferation and facilitated the apoptosis of colon cancer cells by upregulating p53,CDKN1A and TNFRSF10B.

Tribological properties of low-temperature time-dependent pretreated graphite for mechanical seal pairs in high-speed turbopump

The friction coefficient and wear rate of pretreated graphite with liquid nitrogen were obtained by using a ball-on-disk tester,and the wear of GCr15–graphite seal pairs with the low-temperature time-dependent pretreatment was discussed by comparing the wear morphology.The results show that liquid nitrogen pretreatment can affect the hardness and interlayer spacing of graphite.The range of the friction coefficients of pretreated graphite changes from 0.17 to 0.22.With the increase of liquid nitrogen pretreatment time,the wear mechanism of graphite would change from dominated three-body wear to adhesion wear.The experimental results of the mechanical seal with liquid nitrogen pretreatment show that the wear rate of stator is less than 0.00165 mm^(3)·N^(−1)·m^(−1),and the graphite shows a good low-temperature compatibility.

A dynamical neural network approach for distributionally robust chance-constrained Markov decision process

In this paper,we study the distributionally robust joint chance-constrained Markov decision process.Utilizing the logarithmic transformation technique,we derive its deterministic reformulation with bi-convex terms under the moment-based uncertainty set.To cope with the non-convexity and improve the robustness of the solution,we propose a dynamical neural network approach to solve the reformulated optimization problem.Numerical results on a machine replacement problem demonstrate the efficiency of the proposed dynamical neural network approach when compared with the sequential convex approximation approach.

Perioperative remedial antiviral therapy in hepatitis B virus-related hepatocellular carcinoma resection:How to achieve a better outcome

BACKGROUND Although the benefits of antiviral therapy for hepatitis B virus(HBV)-related hepatocellular carcinoma(HCC)have been proven,researchers have not con-firmed the differences in patient outcomes between patients who received preoperative antiviral therapy for a period of time(at least 24 wk)and patients who received remedial antiviral therapy just before radical resection for HBV-related HCC.AIM To investigate the efficacy of perioperative remedial antiviral therapy in patients with HBV-related HCC.METHODS A retrospective study of patients who underwent radical resection for HBV-related HCC at the First Affiliated Hospital of xi’an jiaotong university from January 2016 to June 2019 was conducted.Considering the history of antiviral therapy,patients were assigned to remedial antiviral therapy and preoperative antiviral therapy groups.RESULTS Kaplan–Meier analysis revealed significant differences in overall survival(P<0.0001)and disease-free survival(P=0.035)between the two groups.Multivariate analysis demonstrated that a history of preoperative antiviral treatment was independently related to improved survival(hazard ratio=0.27;95%confidence interval:0.08-0.88;P=0.030).CONCLUSION In patients with HBV-related HCC,it is ideal to receive preoperative long-term antiviral therapy,which helps patients tolerate more extensive hepatectomy;however,remedial antiviral therapy,which reduces preoperative HBV-DNA levels to less than 4 Log10 copies DNA/mL,can also result in improved outcomes.

Sandwich-type composited solid polymer electrolytes to strengthen the interfacial ionic transportation and bulk conductivity for all-solid-state lithium batteries from room temperature to 120 °C

The insurmountable charge transfer impedance at the Li metal/solid polymer electrolytes（SPEs） interface at room temperature as well as the ascending risk of short circuits at the operating temperature higher than the melting point,dominantly limits their applications in solid-state batteries（SSBs）.Although the inorganic filler such as CeO2nanoparticle content of composite solid polymer electrolytes（CSPEs） can significantly reduce the enormous charge transfer impedance at the Li metal/SPEs interface,we found that the required content of CeO2nanoparticles in SPEs varies for achieving a decent interfacial charge transfer impedance and the bulk ionic conductivity in CSPEs.In this regard,a sandwich-type composited solid polymer electrolyte with a 10% CeO2CSPEs interlayer sandwiched between two 50% CeO2CSPEs thin layers（sandwiched CSPEs） is constructed to simultaneously achieve low charge transfer impedance and superior ionic conductivity at 30℃.The sandwiched CSPEs allow for stable cycling of Li plating and stripping for 1000 h with 129 mV polarized voltage at 0.1 mA cm-2and 30℃.In addition,the LiFePO4/Sandwiched CSPEs/Li cell also exhibits exceptional cycle performance at 30℃ and even elevated120℃ without short circuits.Constructing multi-layered CSPEs with optimized contents of the inorganic fillers can be an efficient method for developing all solid-state PEO-based batteries with high performance at a wide range of temperatures.

Stress Concentration Effect in Interface Between Conductor and Epoxy Resin in Coaxial Components of Power Equipment

The spacer used in gas insulated switchgear(GIS)plays a vital role in its safe operation.The mismatch of mechanical properties between conductor and insulating material in spacers results in stress concentration at the interface between them.During a hydrostatic test,mechanical damages are often initiated from the interface.In this paper,the stress between aluminum conductors and epoxy resin is studied by using a simplified coaxial structure specimen.The measurement system and finite element simulation are used to explore the generation of stress and strain during curing and shrinkage.The real-time measurement of strain at the interface shows that the residual strain in the entire process is primarily generated during the cooling stage.Consequently,the temperature and stress/strain distribution at the interface during the cooling stage are further studied,and combined with experiments and simulations.The consistency between the finite element simulation and the experimental measurement results verifies the validity of the simulation method used.The results show that the maximum of the first principal stress generated during the cooling stage of the production process is concentrated on the upper and lower edges of the interface which is critical to the impact of the production quality of the spacer.

The high mortality of patients with lung squamous cell carcinoma(LUSC)results from metastasis rather than primary tumors,1 whereas the molecular pathogenesis of cancer metastasis remains poorly understood.PLAU encodes urokinase-type plasminogen activator(uPA)which is closely related to tumor diagnosis,treatment target,and prognosis of patients.2.3 We previously predicted that PLAU is a key gene negatively associated with overall survival by integrated multidimensional analyses of The Cancer Genome Atlas(TCGA)datasets from 504 samples of LUSC tumor tissues and 46 samples of adjacent non-tumorous lung tissues.4 However,whether the role of PLAU contributes to the metastasis of LUC remains unclear.Here,we analyzed the association between uPA levels and LUSC and found that uPA levels were associated with pathological variations in LUSC patients.

Performance Improvement of Two-Phase Steam Ejector based on CFD Techniques,Response Surface Methodology and Genetic Algorithm

The steam ejector is a crucial component in the waste heat recovery system.Its performance determines the amount of recovered heat and system efficiency.However,poor ejector performance has always been the main bottleneck for system applications.Therefore,this study proposes an optimization methodology to improve the steam ejector's performance by utilizing computational fluid dynamics(CFD) techniques,response surface methodology(RSM),and genetic algorithm(GA).Firstly,a homogeneous equilibrium model(HEM) was established to simulate the two-phase flow in the steam ejector.Then,the orthogonal test was presented to the screening of the key decision variables that have a significant impact on the entrainment ratio(ER).Next,the RSM was used to fit a response surface regression model(RSRM).Meanwhile,the effect of the interaction of geometric parameters on the performance of the steam ejector was revealed.Finally,GA was employed to solve the RSRM's global optimal ER value.The results show that the RSRM exhibits a good fit for ER(R^(2)=0.997).After RSM and GA optimization,the maximum ejector efficiency is 27.94%,which is 48.38% higher than the initial ejector of 18.83%.Furthermore,the optimized ejector efficiency is increased by 46.4% on average under off-design conditions.Overall,the results reveal that the combination of CFD,RSM,and GA presents excellent reliability and feasibility in the optimization design of a two-phase steam ejector.

Measurement,Modelling and Analysis of Residence Time Distribution Characteristics in a Continuous Hydrothermal Reactor

Understanding the residence time distribution(RTD) of a continuous hydrothermal reactor is of great significance to improve product quality and reaction efficiency.In this work,an on-line measurement system is attached to a continuous reactor to investigate the characteristics of RTD.An approach that can accurately fit and describe the experimental measured RTD curve by finding characteristic values is proposed for analysis and comparison.The RTD curves of three experiment groups are measured and the characteristic values are calculated.Results show that increasing total flow rate and extending effective reactor length have inverse effect on average residence time,but they both cause the reactor to approach a plug flow reactor and improve the materials leading.The branch flow rate fraction has no significant effect on RTD characteristics in the scope of the present work except the weak negative correlation with the average residence time.Besides,the natural convection stirring effect can also increase the average residence time,especially when the forced flow is weak.The analysis reveals that it is necessary to consider the matching of natural convection,forced flow and reactor size to control RTD when designing the hydrothermal reactor and working conditions.
