Simultaneous Dual Selective Targeted Delivery of Two Covalent Gemcitabine Immunochemotherapeutics and Complementary Anti-Neoplastic Potency of [Se]-Methylselenocysteine

The anti-metabolite chemotherapeutic, gemcitabine is relatively effective for a spectrum of neoplastic conditions that include various forms of leukemia and adenocarcinoma/carcinoma. Rapid systemic deamination of gemcitabine accounts for a brief plasma half-life but its sustained administration is often curtailed by sequelae and chemotherapeutic-resistance. A molecular strategy that diminishes these limitations is the molecular design and synthetic production of covalent gemcitabine immunoche-motherapeutics that possess properties of selective “targeted” delivery. The simultaneous dual selective “targeted” delivery of gemcitabine at two separate sites on the external surface membrane of a single cancer cell types represents a therapeutic approach that can increase cytosol chemotherapeutic deposition;prolong chemotherapeutic plasma half-life (reduces administration frequency);minimize innocent exposure of normal tissues and healthy organ systems;and ultimately enhance more rapid and thorough resolution of neoplastic cell populations. Materials and Methods: A light-reactive gemcitabine intermediate synthesized utilizing succinimidyl 4,4-azipentanoate was covalently bound to anti-EGFR or anti-HER2/neu IgG by exposure to UV light (354-nm) resulting in the synthesis of covalent immunoche-motherapeutics, gemcitabine-(C4-amide)-[anti-EGFR] and gemcitabine-(C4-amide)-[anti-HER2/neu]. Cytotoxic anti-neoplastic potency of gemcitabine-(C4-amide)-[anti-EGFR] and gemcitabine-(C4-amide)-[anti-HER2/neu] between?gemcitabine-equivalent concentrations of 10-12 M and 10-6 M was determined utilizing chemotherapeutic-resistant mammary adenocarcinoma (SKRr-3). The organoselenium compound, [Se]-methylselenocysteine was evaluated to determine if it complemented the anti-neoplastic potency of the covalent gemcitabine immunoche-motherapeutics. Innovative Artificial Neural Networks-Based Decision Support System for Heart Diseases Diagnosis

Heart diagnosis is not always possible at every medical center, especially in the rural areas where less support and care, due to lack of advanced heart diagnosis equipment. Also, physician intuition and experience are not always sufficient to achieve high quality medical procedures results. Therefore, medical errors and undesirable results are reasons for a need for unconventional computer-based diagnosis systems, which in turns reduce medical fatal errors, increasing the patient safety and save lives. The proposed solution, which is based on an Artificial Neural Networks (ANNs), provides a decision support system to identify three main heart diseases: mitral stenosis, aortic stenosis and ventricular septal defect. Furthermore, the system deals with an encouraging opportunity to develop an operational screening and testing device for heart disease diagnosis and can deliver great assistance for clinicians to make advanced heart diagnosis. Using real medical data, series of experiments have been conducted to examine the performance and accuracy of the proposed solution. Compared results revealed that the system performance and accuracy are acceptable, with a heart diseases classification accuracy of 92%.

Macroeconomic forecasting in China is essential for the government to take proper policy decisions on government expenditure and money supply,among other *** existing literature on forecasting Chinas macroeconomic variables is unclear on the crucial issue of how to choose an optimal window to estimate parameters with rolling out-of-sample *** study fills this gap in forecasting economic growth and inflation in China,by using the rolling weighted least squares(WLS)with the practically feasible cross-validation(CV)procedure of Hong et al.(2018)to choose an optimal estimation *** undertake an empirical analysis of monthly data on up to 30 candidate indicators(mainly asset prices)for a span of 17 years(2000-2017).It is documented that the forecasting performance of rolling estimation is sensitive to the selection of rolling *** empirical analysis shows that the rolling WLS with the CV-based rolling window outperforms other rolling methods on univariate regressions in most *** possible explanation for this is that these macroeconomic variables often suffer from structural changes due to changes in institutional reforms,policies,crises,and other ***,we find that,in most cases,asset prices are key variables for forecasting macroeconomic variables,especially output growth rate.

Proteome changes of porcine follicular fluid during follicle development

Background:Ovarian follicular fluid influences follicle and oocyte growth,but the fluctuation of its protein content during folliculogenesis has not been comprehensively *** we used a shotgun approach and bioinformatics analyses to investigate and compare the proteomes of porcine follicular fluid(pFF)obtained from small(6–12 mm)***:Follicular fluid samples containing highest estrogen levels were selected as non-atretic from small(SNA:26.1±15 ng/mL),medium(MNA:162±54 ng/mL),and large(LNA:290±37 ng/mL)follicles for proteomic *** detected 1627,1699,and 1756 proteins in SNA,MNA,and LNA samples,*** 60–63%of total proteins were specific to each sample,11–13%were shared in pairwise comparisons,and 247 proteins were shared among all *** categorization indicated comparable gene ontology(GO)terms distribution per cellular component,molecular function,and biological process categories across samples;however,the ranking of highly significantly enriched GO terms per category revealed differences between *** patterns of proteinto-protein interactions varied throughout follicle development,and proteins such as serine protease inhibitor,clade E(SERPINE);plasminogen activator,urokinase(PLAU);and plasminogen activator,urokinase receptor(PLAUR)appeared stage-specific to SNA,MNA,and LNA,***“complement and coagulation cascades”was the common major ***,properdin and fibulin-1 were abundant proteins that appeared absent in LNA ***:This study provides extensive and functional analyses of the pFF proteome changes during folliculogenesis and offers the potential for novel biomarker discovery in pFF for oocyte quality assessment.

The evaluation and follow up of liver fibrosis and cirrhosis have been traditionally performed by liver biopsy. However, during the last 20 years, it has become evident that this "gold-standard" is imperfect; even according to its proponents, it is only "the best" among available methods. Attempts at uncovering non-invasive diagnostic tools have yielded multiple scores, formulae, and imaging modalities. All are better tolerated, safer, more acceptable to the patient, and can be repeated essentially as often as required. Most are much less expensive than liver biopsy. Consequently, their use is growing, and in some countries the number of biopsies performed, at least for routine evaluation of hepatitis B and C, has declined sharply. However, the accuracy and diagnostic value of most, if not all, of these methods remains controversial. In this review for the practicing physician, we analyze established and novel biomarkers and physical techniques. We may be witnessing in recent years the beginning of the end of the first phase for the development of non-invasive markers. Early evidence suggests that they might be at least as good as liver biopsy. Novel experimental markers and imaging techniques could produce a dramatic change in diagnosis in the near future.

Temperature effect on photolysis decomposing of perfluorooctanoic acid

Perfluorooctanoic acid（PFOA）is recalcitrant to degrade and ***,the effect of temperature on the photolytic decomposition of PFOA was *** decomposition of PFOA was enhanced from 34% to 99% in 60 min of exposure when the temperature was increased from 25 to 85℃ under UV light（201–600 nm）.The limited degree of decomposition at 25℃ was due to low quantum yield,which was increased by a factor of 12 at 85℃.Under the imposed conditions,the defluorination ratio increased from 8% at 25℃ to 50% at85℃ in 60 *** of perfluorinated carboxylic acids（PFCAs,C7–C5）,PFCAs（C4–C3）and TFA（trifluoroacetic acid,C2）accelerated and attained a maximum within 30 to 90 min at 85℃.However,these reactions did not occur at 25℃ despite extended irradiation to180 *** was decomposed in a step-wise process by surrendering one *** each cyclical process,increased temperature enhanced the quantum yields of irradiation and reactions between water molecules and intermediates *** energy consumption for removing eachμmol of PFOA was reduced from 82.5 k J at 25℃ to 10.9 k J at 85℃ using *** coupled with heat achieved high rates of PFOA degradation and defluorination.
