Constitutive Overexpression of Bcl-2, Survivin and ER Stress Chaperone GRP-78 Confers Intrinsic Radioresistance in Human Hepatocellular Carcinoma Cells: Insight into the Mechanistic Pathways Involved

We present evidence here that abundantly expressed b-catenin-triggered NF-kB-dependent upregulation of inducible nitric oxide synthase(iNOS) found in hepatoma Mahlavu cells (RT-resistant variant designated as RR-Mal), but not in Hep 3B cells (RT-sensitive variant designated as RS-3B) is a key element contribrting to the radioresisitance through the activation of two prominent radioprotective pathways. First, high iNOS expression found in RR-Mal, but not in RS-3B cells was found to perturb calcium homeostasis that triggered ER stress response leading to the overproduction of ER chaperone GRP-78 via robust generation of cleaved ATF-6a (50 kDa) subunits and their nuclear translocation. Meanwhile, both abundantly expressed NF-κB and COX-2 found in RR-Mal cells could also provoke an increased production of PGE2 resulting in robust production of Bcl-2. Interestingly, when RR-Mal cells were treated with PDTC (a NF-κB inhibitor) or celecoxib (a COX-2 inhibitor), a concentration-dependent downregulation of Bcl-2 could be demonstrated implying that Bcl-2 overexpression was indeed mediated through NF-κB/Cox-2/PGE2 pathway. Importantly, we also unveiled that siRNA-mediated silencing of survivin in RR-Mal cells could result in a concomitant downregulation of GRP-78 due to a severe inhibition of ATF-6a (50 kDa) expression. Taken together, our data demonstrate that constitutively overexpressed b-catenin/NF-κB/iNOS and NF-κB/COX-2/PGE2 pathways that overproducing GRP-78, survivin and Bcl-2 expressions are responsible for radioresistance acquisition in RR-Mal cells. Thus, both pathways could be served as potential targets for overcoming radioresistance.

Spatial Pattern Characteristics and Influencing Factors of National Forest Cities in China

In view of the deficiency of national forest cities in the exploration of spatial pattern and influencing factors,this paper adopted the methods of nearest neighbor analysis,kernel density analysis and unbalanced index calculation to analyze the spatial agglomeration pattern and spatial differentiation pattern of national forest *** factors affecting the selection and distribution of national forest cities were investigated by using the geographic detector and GWR *** research results show that:① National forest cities show a spatial aggregation pattern of “east and west sparse,gradient change”,whose distribution is closely related to the climatic area;② The number of national garden cities,forest coverage rate,urbanization rate and local general budget income are four factors that have significant influence on the selection and distribution of national forest cities;③ The four significant influencing factors all have different degrees of spatial non-stationarity,and their two-factor and nonlinear enhancement effects can be generated after two interactions,among which the enhancement effect is the most obvious when the local general budget income interacts with other factors.

Predicted models were built between VC and geographical factors by five different methods,then compared them and chose the best model by *** distribution map of VC can be interpolated by method of Kriging ***:It was found that the correlation of the reference value of Chinese VC and geographical factors were quite *** best models were chosen to predict the vital capacity of the different age and the different gender groups of people in *** distribution map of VC can precisely show the spatial pattern and its variations in ***:If geographical values are obtained in any city or country,VC of this area can be simulated by the model;we can also obtain reference value of VC from the Geographical distribution map in any China area.

Cement Pavement Surface Crack Detection Based on Image Processing

This article introduces the application of image recognition technology in cement pavement crack detection and put forward to method fordetermining threshold about grayscale stretching. This algorithm is designed for binarization which has a self-adaptive characteristic. After theimage is preprocessed, we apply 2D wavelet and Laplace operator to process the image. According to the characteristic of pixel of gray image, analgorithm designed on binarization for Binary image. The feasibility of this method can be verified the image processed by comparing with theresults of three algorithms: Otsu method, iteration method and fixed threshold method.
