A Based on daily maximum and minimum temperature from Rongcheng station during 1981 -2010, the changing trends of extreme temperature indexes were analyzed. The results show that for the relative indexes, number of warm days （nights） in Rongcheng station showed an increasing trend, while the number of cold days （nights） showed a decreasing trend from 1981 to 2010, but the decreasing trend of number of cold days was small. For the extreme indexes, the extreme maximum temperature, the maximum of extreme minimum temperature and extreme mini- mum temperature increased, while the minimum of extreme maximum temperature decreased from 1981 to 2010. For the absolute indexes, the numbers of frozen days and frost days reduced, and frozen days were less than frost days for all year.

This paper aimed to examine the site dependence and evaluate the methods for site analysis of far-source ground motions. This was achieved through the examination of frequency content estimated by different methods based on strong ground motions recorded at twelve far-source stations in Shandong province during the Wenchuan earthquake. The stations were located in sites with soil profiles ranging from code classes Ⅰ to Ⅲ. Approaches used included the Fourier amplitude spectrum （FAS）, the earthquake response spectrum （ERS）, the spectral ratio between the horizontal and the vertical components （H/V）, the spectral ratio between the spectra at the site and at a reference site （SRRS）, and coda wave analysis （CWA）. Results showed that major periods of these ground motions obtained by FAS, ERS and H/V ratio methods were all evidently larger than site dominant periods; the periods were also different from each other and mainly reflected the frequency content of long period components. Prominent periods obtained by the SRRS approach neither illuminated the long period aspect nor efficiently determined site features of the motions. The CWA resulted in a period close to site period for stations with good quality recordings. The results obtained in this study will be useful for the evaluation of far-source effect in constructing seismic design spectra and in selecting methods for ground motion site analysis.

Lancelets (subphylum Cephalochordata) are a transitional species between invertebrates and vertebrates. They are currently listed in the Second Order of Protected Animals in China. Lancelets were first documented in the waters around the city of weihai (Shandong, China) in 2002. However, little is known about the phylogeny of this population. We analyzed the sequences of cytochrome b (Cyt b) and cytochrome oxidase c subunit I (CO I) genes from samples collected from coastal waters in the cities of weihai and Qingdao (~150 km to the south). We analyzed 176 sequences, of which 150 were novel sequences and 26 were obtained from GenBank. Our results suggest that (1) lancelets in the two cities belong to the species Branchiostoma japonicus and have a high level of genetic diversity; (2) there is a high level of gene flow and low level of genetic differentiation between lancelets from the two cities; (3) demographic expansion occurred an estimated 1.1 million years (Ma) ago (mid Pleistocene) for lancelets in weihai-Qingdao; and (4) the divergence between B. belcheri and B. japonicus was estimated at between 37.75 Ma (early Oligocene)-46.5 Ma (late Eocene).

Appropriate estimates of earthquake response spectrum are essential for design of new structures, or seismic safety evaluation of existing structures. This paper presents an alternative procedure to construct design spectrum from a combined normalized response spectrum （NRSc） which is obtained from pseudo-velocity spectrum with the ordinate scaled by different peak ground amplitudes （PGA, PGV, PGD） in different period regions. And a consecutive function./（/） used to normalize the ordinates is defined. Based on a comprehensive study of 220 strong ground motions recorded during recent eleven large worldwide earthquakes, the features of the NRSc are discussed and compared with the traditional normalized acceleration, velocity and displacement response spectra （NRSA, NRSv, NRSD）. And the relationships between ground amplitudes are evaluated by using a weighted mean method instead of the arithmetic mean. Then the NRSc is used to define the design spectrum with given peak ground amplitudes. At last, the smooth spectrum is compared with those derived by the former approaches, and the accuracy of the proposed spectrum is tested through an analysis of the dispersion of ground motion response spectra.

Remote sensing products are significant in the data assimilation of an ocean model. Considering the resolution and space coverage of different remote sensing data, two types of sea surface height（SSH） product are employed in the assimilation, including the gridded products from AVISO and the original along-track observations used in the generation. To explore their impact on the assimilation results, an experiment focus on the South China Sea（SCS） is conducted based on the Regional Ocean Modeling System（ROMS） and the four-dimensional variational data assimilation（4 DVAR） technology. The comparison with EN4 data set and Argo profile indicates that, the along-track SSH assimilation result presents to be more accurate than the gridded SSH assimilation, because some noises may have been introduced in the merging process. Moreover, the mesoscale eddy detection capability of the assimilation results is analyzed by a vector geometry–based algorithm. It is verified that, the assimilation of the gridded SSH shows superiority in describing the eddy＇s characteristics, since the complete structure of the ocean surface has been reconstructed by the original data merging.

Antarctic ice microalga Chlamydomonas sp. can thrive undisturbed under high UV radiation in the Antarctic ice layer. However, it is unknown that the initial adaptation mechanisms in protein level occurring in response to high UV radiation. Global-expression profiling of proteins in response to stress was analyzed by two-dimensional electrophoresis （2-DE） and image analysis. In the 2-DE analysis, protein preparation is the key step. Three different protein extract methods were compared, and the results showed that the trichloroacetic acid （TCA）-acetone fractional precipitation method was the fittest one. At the same time, the proteins in Chlamydomonas sp. were compared in 2-DE way, and the synthesis of seven protein spots was found disappeared and 18 decreased after exposed to UV-B radiation. In addition, 14 protein spots were enhanced or induced, among which two new peptides （20 and 21 kDa） appeared whose isoelectric point （pI） was 7.05 and 4.60 respectively. These changed proteins might act as key role in the acclimation of Antarctic ice microalga to UV-B radiation

Antarctic ice microalga can survive and thrive in cold channels or pores in the Antarctic ice layer. In order to understand the adaptive mechanisms to low temperature, in the present study we compared two-dimensional polyacrylamide gel electrophoresis （2-DE） profiles of normal and low temperature-stressed Antarctic ice microalga Chlamydomonas sp. cells. In addition, new protein spots induced by low temperature were identified with peptide mass fingerprinting based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry （MALDI-TOF-MS） and database searching. Well-resolved and reproducible 2-DE patterns of both normal and low temperature-stressed cells were acquired. A total of 626 spots was detected in control cells and 652 spots were detected in the corresponding low temperature-stressed cells. A total of 598 spots was matched between normal and stressed cells. Two newly synthesized proteins （a and b） in low temperature-stressed cells were characterized. Protein spot A （53 kDa, pl 6.0） was similar to isopropylmalate/homocitrate/citramalate synthases, which act in the transport and metabolism of amino acids. Protein spot b （25 kDa, pl 8.0） was related to glutathione S-transferase, which functions as a scavenger of active oxygen, free radicals, and noxious metabolites. The present study is valuable for the application of ice microalgae, establishing an ice microalga Chlamydomonas sp. proteome database, and screening molecular biomarkers for further studies.

Acclimatory responses to high-salt stress in Chlamydomonas (Chlorophyta, Chlorophyceae) from Antarctica

Antarctic ice microalga can survive and thrive in channels or pores containing high salinity in Antarctic ice layer. In this study, it was found that cell membrane permeability of green microalga Chlaraydomonas sp. L4 from Antarctic sea ice was high in cells treated with hypersalinity due to the induction of active oxygen and radicals. However, increased super oxide dismutase （SOD） scavenged harmful free radicals effectively to keep cell membrane integrity. Also, the analysis of membrane fatty acids demonstrated the content of saturated fatty acids and monounsaturated fatty acids in- creased and polyunsaturated fatty acids decreased under the high-salt treatment for 14 d, which effectively reduced the membrane fluidity and minimized the injury to cell membrane. The morpho- logical changes showed that hypersalinity induced the increase of cell volume and the consumption of starch granules. However, because of the increase in detoxification of vacuoles, electron-dense deposits and SOD activity under high-salt stress, the complete noninterference thylakoids, mito- chondria and cell nucleus maintained cellular fundamental metabolism. Global-expression profiling of proteins showed eight protein spots disappeared, 18 protein spots decreased and 18 protein spots were enhanced after the high-salt shock obviously （P 〈0.05）. One new peptide （pI 6.90; MW 51 kDa） was primarily confirmed as the processor of light reaction center protein CP43 in photosystem II, which increased photosynthesis ability of Chlamydomonas sp. L4 treated with high salinity.

In order to provide pertinent evidence of ion channel with immune response in the fish, whole cell patch-clamp technique was employed for potassium ion channel study in turbot （Scophthalmus maximus）. Lymphocytes were isolated by Percoll density gradient centrifugation from peripheral blood samples, and electrophysiological characters of the channel were analyzed. In the recorded cells, activated voltage of the channels was -42.5±3.7 mV and the average peak current was 313.12±28.2 pA. The channel was identified as voltage dependent, the current was outward and it could be inhibited by 10 mmol/dma TEA or 5 mmol/dm^3 4-AP, a specific potassium channel inhibitor, identifying the existence of potassium channel in peripheral lymphocytes of the turbot.
