Hepatocellular carcinoma(HCC),a prevalent solid carcinoma of significant concern,is an aggressive and often fatal disease with increasing global incidence rates and poor therapeutic *** etiology and pathological progression of non-alcoholic steatohepatitis(NASH)-related HCC is multifactorial and ***,no single animal model can accurately mimic the full NASH-related HCC pathological progression,posing considerable challenges to transition and mechanistic ***,a novel conditional inducible wild-type human HRAS overexpressed mouse model(HRAS-HCC)was established,demonstrating 100%morbidity and mortality within approximately one month under normal dietary and lifestyle *** symptoms of HCC such as ascites,thrombus,internal hemorrhage,jaundice,and lung metastasis were successfully replicated in ***-depth pathological features of NASH-related HCC were demonstrated by pathological staining,biochemical analyses,and typical marker gene *** murine anti-PD-1 and sorafenib treatment effectively prolonged mouse survival,further confirming the accuracy and reliability of the *** on protein-protein interaction(PPI)network and RNA sequencing analyses,we speculated that overexpression of HRAS may initiate the THBS1-COL4A3 axis to induce NASH with severe fibrosis,with subsequent progression to ***,our study successfully duplicated natural sequential progression in a single murine model over a very short period,providing an accurate and reliable preclinical tool for therapeutic evaluations targeting the NASH to HCC continuum.

Since 2015,stroke has become the leading cause of death and disability in China,posing a significant threat to the health of its citizens as a major chronic non-communicable *** to the China Stroke High-risk Population Screening and Intervention Program,an estimated 17.8 million[95%confidence interval(CI)17.6–18.0million]adults in China had experienced a stroke in 2020,with 3.4 million(95%CI 3.3–3.5 million)experiencing their first-ever stroke and another 2.3 million(95%CI 2.2–2.4 million)dying as a ***,approximately 12.5%(95%CI 12.4%–12.5%)of stroke survivors were left disabled,as defined by a modified Rankin Scale score greater than 1,equating to 2.2 million(95%CI 2.1–2.2 million)stroke-related disabilities in *** the population ages and the prevalence of risk factors like diabetes,hypertension,and hyperlipidemia continues to rise and remains poorly controlled,the burden of stroke in China is also increasing.A large national epidemiological survey initiated by the China Hypertension League in 2017 showed that the prevalence of hypertension was 24.7%;the awareness,treatment,and control rates in hypertensive patients were:60.1%,42.5%,and 25.4%,respectively.A nationally representative sample of the Chinese mainland population showed that the weighted prevalence of total diabetes diagnosed by the American Diabetes Association criteria was 12.8%,suggesting there are 120 million adults with diabetes in China,and the awareness,treatment,and control rates in diabetic patients were:43.3%,49.0%,and 49.4%,***“Sixth National Health Service Statistical Survey Report in 2018”showed that the proportion of the obese population in China was 37.4%,an increase of 7.2 points from *** from 1599 hospitals in the Hospital quality monitoring System and Bigdata Observatory Platform for Stroke of China(BOSC)showed that a total of 3,418,432 stroke cases[mean age±standard error(SE)was(65.700±0.006)years,and 59.1%were male]were admitted dur

Avian pathogenic Escherichia coli(APEC)belonging to extraintestinal pathogenic ***(ExPEC)can cause severe infections in extraintestinal tissues in birds and humans,such as the lungs and ***(microcin production regulation,locus A,herein renamed AbsR,a blood survival regulator),a member of the MarR(multiple antibiotic resistance regulator)transcriptional regulator family,governs the expression of capsule biosynthetic genes in human ExPEC and represents a promising druggable target for ***,a deep understanding of the AbsR regulatory mechanism as well as its regulon is *** this study,we present a systems-level analysis of the APEC AbsR regulon using ChIP-Seq(chromatin immunoprecipitation sequencing)and RNA-Seq(RNA sequencing)*** found that AbsR directly regulates 99 genes and indirectly regulates 667 ***,we showed that:1)AbsR contributes to antiphagocytotic effects by macrophages and virulence in a mouse model for systemic infection by directly activating the capsular gene cluster;2)AbsR positively impacts biofilm formation via direct regulation of the T2SS(type II secretion system)but plays a marginal role in virulence;and 3)AbsR directly upregulates the acid tolerance signaling system EvgAS to withstand acid stress but is dispensable in ExPEC ***,our data indicate that the role of AbsR in virulence gene regulation is relatively conserved in ExPEC ***,this study provides a comprehensive analysis of the AbsR regulon and regulatory mechanism,and our data suggest that AbsR likely influences virulence primarily through the control of capsule ***,we found that AbsR severely represses the expression of the type I-F CRISPR(clustered regularly interspaced short palindromic repeats)-Cas(CRISPR associated)systems,which could have implications in CRISPR biology and application.

Globally,groundwater has globally emerged as a crucial freshwater source for domestic,irrigation,and industrial *** evaluation of groundwater quality in the Toshka region is imperative to ensure its suitability for the extensive agricultural and industrial activities underway in this promising,groundwater-dependent development *** is particularly significant as Egypt increasingly relies on groundwater reserves to address freshwater deficits and to implement mega-development projects in barren *** this study,fifty-two samples were collected from the recently drilled wells tapping into the Nubian Sandstone Aquifer(NSA)in the Toshka *** quality was assessed through hydrochemical analysis,Piper diagram,and various indicators such as Na%,SAR,RSC,KR,MH and *** hydrochemical analysis revealed improved groundwater quality characteristics,attributed to continuous recharge from Lake *** Piper diagram categorised most of the water samples as"secondary salinity"water *** all wells proved suitable for irrigation with only two wells unsuitable based on MH values and six wells based on KR *** Total Hardness(TH)values,all samples were classified as"Soft",indicating their suitability for domestic and industrial *** quality Index(WQI)results concluded that all samples met WHO and FAO guidelines for drinking and irrigation,*** distribution maps,constructed using GIS,facilitate the interpretation of the *** monitoring of quality parameters is essential to detect any deviation from permissible limits.

Participatory Forest Management and Gender Inclusiveness within the Community Forest Management Groups of Bhutan

Community forest management groups (CFMGs) in Bhutan exhibit participatory forest management practices that recognize the importance of community’s collective participation in the management of natural forest resources. This approach involves the community in the stewardship of designated forest areas and resources to ensure sustainable livelihoods and realization of forest conservation objectives. The increase of CFMGs in the country has been successful. However, research on the extent of gender-inclusive participation in CFMGs is either insufficient or missing vis-à-vis the allocation of decision-making power. Therefore, this study analyzes the factors influencing gender participation in CFMGs and their integration into decision-making processes. Primary data were collected from 12 study sites spanning 4 regions, complemented by secondary data from the Forest Department. Regression models were used to identify factors significantly influencing CFMG member participation in decision-making. The empirical results of this study reveal that gender is a significant factor influencing participation in CFMG decision-making. The study concludes that there is insufficient participation of women members in decision-making processes. Therefore, consideration of gender should be included in the development phase of the CFMG policy in addition to promoting awareness of inequity between gender and the promotion of leadership roles for women in CFMGs.

Normothermic machine perfusion(NMP)is an innovative technique used in solid organ transplantation that involves perfusing the organ with specialized solution or leukocyte depleted red blood cells at near-normal body temperature aiming at mimicking physiological conditions,and providing optimal conditions for organ preservation which leads to reduced risk of ischemia reperfusion injury(IRI)when compared to the standard static cold storage(SCS)(1).One of the key advantages of NMP is its ability to assess the quality of the organ in real-time and assessment of the organ performance prior to *** continuously monitoring parameters such as blood flow,oxygen consumption,and lactate production,clinicians can evaluate the viability of the organ and make more informed decisions about its suitability for *** real-time assessment can help reduce the risk of transplanting organs that may not function optimally or have a higher likelihood of complications ***,NMP may expand the pool of donor organs by allowing for the use of organs that may have been deemed marginal or unsuitable for transplantation using traditional methods(1,2).

Excavation of underground caverns,such as mountain tunnels and energy-storage caverns,may cause the damages to the surrounding rock as a result of the stress *** this influenced zone,new cracks and discontinuities are created or propagate in the rock ***,it is effective to measure and evaluate the acoustic emission(AE)events generated by the rocks,which is a small elastic vibration,and permeability *** authors have developed a long-term measurement device that incorporates an optical AE(O-AE)sensor,an optical pore pressure sensor,and an optical temperature sensor in a single multi-optical measurement probe(MOP).Japan Atomic Energy Agency has been conducting R&D activities to enhance the reliability of high-level radioactive waste(HLW)deep geological disposal *** a high-level radioactive disposal project,one of the challenges is the development of methods for long-term monitoring of rock mass ***,in January 2014,the long-term measurements of the hydro-mechanical behavior of the rock mass were launched using the developed MOP in the vicinity of 350 m below the surface at the Horonobe Underground Research *** measurement results show that AEs occur frequently up to 1.5 m from the wall during *** addition,hydraulic conductivity increased by 2e4 orders of *** analysis revealed that the hydraulic behavior of the rock mass affected the pore pressure fluctuations and caused *** on this,a conceptual model is developed to represent the excavation damaged zone(EDZ),which contributes to the safe geological disposal of radioactive waste.

Tracking PCB Contamination in Ontario Great Lakes Tributaries: Development of Methodologies and Lessons Learned for watershed Based Investigations

Project Trackdown is an investigative environmental program aimed at tracking sources of polychlorinated biphenyl (PCB) contamination in Great Lakes tributaries. The program uses a multimedia weight of evidence approach for identifying sources of PCBs to the environment. PCB concentrations in environmental media (sediment, water, suspended sediment and soil), passive samplers and/or exposed biota (mussels, young-of-the-year fish and benthic invertebrates) are used in combination to evaluate bioavailability and identify local anomalies within a tributary. These lines of evidence can be assessed with simple chemometric techniques and fingerprinting of PCB congener profiles, and, combined with anecdotal information such as land use history and tributary alterations, may be used to identify ongoing and locally controllable sources of PCBs to the Great Lakes. The program was successful at developing environmental triggers to differentiate potential source areas from background PCB conditions in urban areas, allowing efforts to focus on identifying active ongoing sources of PCB contamination. Project Trackdown has been carried out in three tributaries to Lake Ontario (Cataraqui River, Etobicoke Creek and Twelve Mile Creek) and two tributaries that flow into the Detroit River (Turkey Creek and Little River). Local ongoing PCB sources have been identified in four projects, leading to abatement or remediation measures. As a collaborative initiative between the Ontario Ministry of the Environment and Environment Canada, Project Trackdown has successfully identified several PCB sources leading to substantial cleanup efforts aimed ultimately at reducing PCB contamination to the Great Lakes.
