Genetic composition,genetic diversity and small-scale environmental variation matter for the experimental reintroduction of a rare plant

Aims reintroduction has become an important tool for the management of endangered plant *** tested the little-explored effects of small-scale environmental variation,genotypic composition(*** of genotypes)and genotypic diversity on the population survival of the regionally rare clonal plant ranunculus *** this species of periodically inundated lakeshores,genetic differentiation had been reported between populations and between short-flooded and long-flooded microsites within *** We established 306 experimental test populations at a previously unoccupied lakeshore,comprising either monocultures of 32 genotypes,mixtures of genotypes within populations or mixtures of genotypes between *** 2000,three years after planting out at the experimental site,a long-lasting flood caused the death of half of the experimental *** 2003,an extreme drought resulted in the lowest summer water levels ever *** findings Despite these climatic extremes,27%of the established populations survived in December *** success of experimental populations largely differed between ***,the success of genotype monocultures depended on genotype and source *** differentiation between microsites played a minor role for the success of *** the flood,populations planted with genotypes from different source populations increased in abundance,whereas populations with genotypes from single source populations and genotype monocultures *** 2014,several small patches of ranunculus clones were still present,but plants were strongly intermingled,which precluded their assignment to the original *** conclude that sources for reintroductions need to be selected ***,mixtures of plants from different populations appear to be the best choice for successful reintroduction,at least in unpredictably varying environments.

Electrochemical Immunosensors for the Diagnosis of Celiac Disease

Celiac disease is a permanent intolerance to gluten proteins of wheat, rye, barley, and oats in genetically susceptible individuals. The clinical picture is characterized by inflammation and damage of the small intestinal mucosa and malabsorption of essential nutrients. Therapeutically, a lifelong strict gluten-free diet is necessary. The diagnosis of celiac disease is complex and includes symptomatology, serology, small intestinal histology, and genetic status. Serological testing plays a central role within the diagnostic procedure and is based on the measurement of disease-specific antibodies against gluten proteins (antigen) and tissue transglutaminase (autoantigen). Immunofluorescence detection and enzyme-linked immunosorbent assays are currently most often applied for antibody testing. However, these tests are expensive and time-consuming. Therefore, simple and rapid alternative methods have been developed during the last years, and electro-chemical immunosensors seem to be the most promising analytical tools. The architecture of these sensors may comprise the following elements: working and reference electrodes, covalent or noncovalent binding of the antigen to the surface of the working electrode by means of a functional monolayer, and blocking of unreacted binding sites. The analytical procedure is initiated by adding the analyte (serum antibodies) and an analyte-specific second antibody, which is usually labeled with an enzyme. The special reaction of the enzyme with an appropriate substrate results in a product that initiates a current that can be measured by different electrical methods. A number of different electrochemical immunosensors variable in different electrodes, binding systems, secondary antibodies, and current measurements have been developed. Most of them have been tested with real human serum samples of celiac patients and healthy individuals, and some of them reached disease sensitivity and specificity comparable with traditional analytica

Background： During processing in a desolventizer/toaster（DT）, rapeseed meal（rSM） is heated to evaporate the hexane and to reduce the level of heat-labile anti-nutritional factors such as glucosinolates（GSL）. However, excessive heat treatment may reduce amino acid（AA） content in addition to lower AA digestibility and availability in rSM. The objective of the present study was to produce from one batch of a 00-rapeseed variety（17 μmol GSL/g dry matter（DM）, seed grade quality） five differently processed rSM under standardized and defined conditions in a pilot plant,and to determine the impact of these different treatments on protein solubility and chemical composition, in particular with regard to contents of AA including reactive Lys（rL ys） and levels of total and individual ***： four rSM were exposed to wet toasting conditions（WetT C） with increasing residence time in the DT of 48,64, 76, and 93 min. A blend of these four rSM was further processed, starting with saturated steam processing（〈 100 °C）and followed by exposure to dry toasting conditions（DryT C） to further reduce the GSL content in this ***： The contents of neutral detergent fiber and neutral detergent fiber bound crude protein（CP） increased linearly（P 〈 0.05）, as residence time of rSM in the DT increased from 48 to 93 min, whereas contents of total and most individual GSL and those of Lys, rL ys, Cys, and the calculated ratio of Lys：CP and r Lys：CP decreased linearly（P ≤ 0.05）.The combination of wet heating and DryT C resulted in the lowest GSL content compared to rSM produced under WetT C, but was associated with lowest protein ***： It can be concluded that by increasing residence time in the DT or using alternative processing conditions such as wet heating combined with DryT C, contents of total and individual GSL in rSM can be substantially *** in vivo studies are warranted to elucidate if and to which extent the observed differences in protein quality and

Aims We investigated the regulation of the water status in three predominant perennial C3 phreatophytes(Alhagi sparsifolia,Populus euphratica,Tamarix ramosissima)at typical sites of their occurrence at the southern fringe of the hyperarid Taklamakan Desert(north-west China).Methods In the foreland of the river oasis of Qira(Cele),we determined meteorological variables,plant biomass production,plant water potentials(WL)and the water flux through the *** calculated the hydraulic conductance on the flow path from the soil to the leaves(kSL)and tested the effects of kSL,WL and the leaf-to-air difference in the partial pressure of water vapour(Dw)on stomatal regulation using regression *** findings Despite high values of plant water potential at the point of turgor loss,all plants sustained WL at levels that were high enough to maintain transpiration throughout the growing *** ***,stomatal resistance(rs;related to leaf area or leaf mass)was most closely correlated with kSL;whereas in ***,~70%of the variation in rs was explained by *** ***,leaf area-related rs was significantly correlated with WL and *** regulation mechanisms are in accordance with the growth patterns and the occurrence of the species in relation to their distance to the ground water.
