Background: Extensive degradation of amino acids in the rumen via microbial deamination decreases the postruminal availability of dietary indispensable amino acids. Together with the normal decrease in voluntary dry matter intake(DMI) around parturition in dairy cows, microbial metabolism contributes to a markedly negative balance of indispensable amino acids, including methionine which may be the first-limiting for milk production. The main objective of the current study was to profile changes in major bacterial species with key functions in cel ulose and hemicel ulose digestion, xylan breakdown, proteolytic action, propionic acid production, lactate utilization and ruminal biohydrogenation in cows supplemented with rumen-protected Methionine(SM; Smartamine M, Adisseo NA, Alpharetta,GA, USA) from-23 through 30 d relative to parturition. Because ~90% of the methionine in SM bypasses the rumen,~10% of the methionine is released into the rumen and can be utilized by ***: As expected, there was an increase in overall DMI after parturition(Day, P treatment × Day, P < 0.05) abundance prepartum of Fibrobacter succinogenes, Succinimonas amylolytica, and Succinivibrio dextrinosolvens. However, the greater DMI in cows fed SM after parturition(19.6 kg/d versus 13.9 kg/d) was associated with lower abundance of Fibrobacter succinogenes(2.13 × 10-3 versus 2.25 × 10-4) and Selenomonas ruminantium(2.98 × 10-1 versus 4.10 × 10-1). As one of the largest human activities, World Expo is an important source of anthropogenic Greenhouse Gas emission（GHG）, and the GHG emission and other environmental impacts of the Expo Shanghai 2010, where around 59,397 tons of waste was generated during 184 Expo running days, were assessed by life cycle assessment（LCA）. Two scenarios, i.e., the actual and expected figures of the waste sector, were assessed and compared, and 124.01 kg CO2-equivalent（CO2-eq.）, 4.43 kg SO2-eq., 4.88 kg NO-3-eq., and 3509 m3 water per ton tourist waste were found to be released in terms of global warming（GW）, acidification（AC）,nutrient enrichment（NE） and spoiled groundwater resources（SGWR）, respectively. The total GHG emission was around 3499 ton CO2-eq. from the waste sector in Expo Park, among which 86.47% was generated during the waste landfilling at the rate of 107.24 kg CO2-eq.,and CH4, CO and other hydrocarbons（HC） were the main contributors. If the waste sorting process had been implemented according to the plan scenario, around 497 ton *** could have been attained. Unlike municipal solid waste, with more organic matter content, an incineration plant is more suitable for tourist waste disposal due to its high heating value, from the GHG reduction perspective.

Stem cells are pluripotent cells, having a property of differentiating into various types of cells of human body. Several studies have developed mesenchymal stem cells(MSCs) from various human tissues,peripheral blood and body fluids. These cells are then characterized by cellular and molecular markers to understand their specific phenotypes. Dental pulp stem cells(DPSCs) are having a MSCs phenotype and they are differentiated into neuron, cardiomyocytes, chondrocytes, osteoblasts, liver cells and β cells of islet of pancreas. Thus, DPSCs have shown great potentiality to use in regenerative medicine for treatment of various human diseases including dental related problems. These cells can also be developed into induced pluripotent stem cells by incorporation of pluripotency markers and use for regenerative therapies of various diseases. The DPSCs are derived from various dental tissues such as human exfoliated deciduous teeth, apical papilla, periodontal ligament and dental follicle tissue. This review will overview the information about isolation, cellular and molecular characterization and differentiation of DPSCs into various types of human cells and thus these cells have important applications in regenerative therapies for various diseases. This review will be most useful for postgraduate dental students as well as scientists working in the field of oral pathology and oral medicine.

Objective:To examine the role of heat shock protein 90(Hsp90) in the maintenance of actin cytoskeleton in human neuroblastoma tumor ***:Co-precipitation experiments were performed to examine Hsp90 interaction with ***90 and actin interactions were evaluated by protein refolding and acto-myosin motility assays.17-(AUylamino)-17- demethoxygeldanamycin(17AAG) induced actin-cytoskeleton re-organization was examined by laser scanning confocal ***:It was shown that inhibition of Hsp90 by 17AAC accelerates detergent induced cell lysis of neuroblastoma tumor cells through destabilization of actin *** in vitro co-precipitation experiments showed that functional but not mutant Hsp90 binds with *** biochemical modifications,phopshorylation and oligomerization enhanced Hsp90 binding with F-actin.F-actin binding to Hsp90 interfered with Hsp90 chaperone activity in protein refolding assays,and Hsp90 binding to F-actin interfered with actin motility on myosin coated flow *** the combination treatment,17AAG irreversibly augmented the effect of cytochalasin D,an inhibitor of actin ***:It can be concluded that Hsp90 binds to F-actin in tumor cells and maintains the cellular integrity. The results display a novel element of Hsp90 inhibition in destabilizing the actin cytoskeleton of tumor cells,therefore suggest that 17AAG combination with cytoskeletal disruptor may be effective in combating cancer.

BACKGROUND There is scant literature on hepatocellular carcinoma(HCC)in patients with Budd-Chiari syndrome(BCS).AIM To assess the magnitude,clinical characteristics,feasibility,and outcomes of treatment in *** A total of 904 BCS patients from New Delhi,India and 1140 from Mumbai,India were *** prevalence and incidence of HCC were determined,and among patients with BCS-HCC,the viability and outcomes of interventional therapy were *** In the New Delhi cohort of 35 BCS-HCC patients,18 had HCC at index presentation(prevalence 1.99%),and 17 developed HCC over a follow-up of 4601 person-years,[incidence 0.36(0.22-0.57)per 100 person-years].BCS-HCC patients were older when compared to patients with BCS alone(P=0.001)and had a higher proportion of inferior vena cava block,cirrhosis,and long-segment vascular *** median alpha-fetoprotein level was higher in patients with BCS-HCC at first presentation than those who developed HCC at follow-up(13029 ng/mL vs 500 ng/mL,P=0.01).Of the 35 BCS-HCC,26(74.3%)underwent radiological interventions for BCS,and 22(62.8%)patients underwent treatment for HCC[transarterial chemoembolization in 18(81.8%),oral tyrosine kinase inhibitor in 3(13.6%),and transarterial radioembolization in 1(4.5%)].The median survival among patients who underwent interventions for HCC compared with those who did not was 3.5 years vs 3.1 mo(P=0.0001).In contrast to the New Delhi cohort,the Mumbai cohort of BCS-HCC patients were predominantly males,presented with a more advanced HCC[Barcelona Clinic Liver Cancer C and D],and 2 patients underwent liver *** HCC is not uncommon in patients with *** interventions and liver transplantation are feasible in select primary BCS-HCC patients and may improve outcomes.

In eukaryotic cells, chromatin transformation from euchromatin into heterochromatin as a means of controlling gene expression and replication has been known as the ‘‘accessibility hypothesis' '. The interplay of epigenetic changes including histone modifications, DNA methylation, RNA interference(RNAi) and other func- tional epigenetic components are intricate. It is believed that these changes are well-programmed, inherited and can be modified by environmental contaminant stressors. Environmentally-driven epigenetic alterations during development, e.g. embryonic, foetal or neonatal stage, may influence disease susceptibility in adulthood. Therefore, understanding how epigenome modifications develop in response to environmental chemicals and, how epigenetic-xenobiotic interactions influence human health will shed new insights into gene-environment interactions in the epidemiology of several diseases including cancer. In this review, we consider studies of chemical modifiers including nutritional and xenobiotic effects on epigenetic components in vitro or in vivo. By examining the most-studied epigenome modifications and how their respective roles are interlinked, we highlight the central role of xenbiotic- modified epigenetic mechanisms. A major requirement will be to study and understand effects following environmentally-relevant exposures. We suggest that the study of epigenetic toxicology will open up new opportunities to devise strategies for the prevention or treatment of at-risk populations.
