Urban air pollution,climate and its impact on asthma morbidity

Objective:To study the mechanism of formation of air quality and to determine the impact of the studied factors on asthma morbidity in ***:The evaluation of air pollution in vladivostok was done using long-term(2008-2012) monitoring data(temperature,humidity,atmospheric pressure,wind speed,etc.).The levels of suspended particulate matter,nitrogen and sulfur dioxide,carbon monoxide,ammonia,formaldehyde(mg/m^3) in six stationary observation posts were *** studied the aerosol suspensions of solid particles,which were collected during snowfall from precipitation(snow) and air in 14 districts with different levels of anthropogenic *** snow was analyzed on laser *** impact of air pollution on the distribution of asthma morbidity was evaluated in various age groups by data of federal statistical observation obtained from 8 adults and 7 children municipal clinics in vladivostok(2008-2012).Results:The content of suspended particulate components of pollution remained more stable,due to the features of atmospheric circulation,rugged terrain and residential *** nano- and micro-sized particles(0-50 μm),which can absorb highly toxic metals,prevail in dust *** respirable fractions of particles,even in small doses,can contribute to the increase in asthma morbidity in the ***:We determined that asthma morbidity depends from general air pollution(in the range of 18.3%).It was detected that the highest age-specific dependence is associated with the content of particulate matter,carbon monoxide and nitrogen dioxide in air.

Fatty acids composition of erythrocytes in arterial hypertension associated with dyslipidemia

Studied the fatty acid (FA) composition of erythrocyte membranes in patients with arterial hypertension associated with dyslipidemia. The fatty acid composition of erythrocyte membranes was studied by gaz chromatography. In all hypertensive patients are disturbed lipid FA composition of erythrocytes: increase in saturated fatty acids (15:0, 16:0), monoenic fatty acids (18:1n9, 18:1n7), polyunsaturated fatty acids (PUFA) family n6 (20:4n6), decrease in polyunsaturated fatty acids family n3:22:5n3, 22:6n3, the total value of n3 PUFA. Cells endogenous deficiency of n3 PUFA leads to the changes of the plasmatic membranes physicochemical properties, activation of eicosanoids synthesis with proinflammatory and vasoconstriction activity, the formation of the systemic inflammatory syndrome and conditions for the atherogenesis. Thus, structural and functional changes in erythrocyte membranes in hypertension developed as a part of the common systemic disorder of lipid metabolism.

Bronchial inflammatory profile in interferon-gamma-mediated immune response in asthma patients during airway response to cold stimulus

Objective:To evaluate the inflammatory pattern and the interferon(IFN)-γin the bronchial secretion of asthma patients in response to acute cold *** and methods:We enrolled 42 patients with *** assessed asthma by Asthma Control Test,the lung function by spirometry before and after the bronchodilator test,followed by collecting induced *** next day,we collected exhaled breath condensate(EBC)and conducted a 3-minute isocapnic hyperventilation with cold air(IHCA),followed by collecting spontaneously produced ***:Group 1 included 20 patients with cold airway hyperresponsiveness(CAHR),and group 2 included 22 patients without *** both groups,a high level of neutrophils in bronchial secretion was observed before and after *** response to IHCA,the number of epitheliocytes in the sputum decreased to a greater extent in patients of group *** baseline epitheliocytes and the concentration of IFN-γafter IHCA had an inverse relationship(r=-0.60;P=0.017).The baseline IFN-γin EBC before and after IHCA was lower in group *** response to cold exposure directly correlated with IFN-γlevels after IHCA(Rs=0.42;P=0.014).Conclusion:In asthma patients with CAHR,there is a relationship between the persistence of mixed inflammation and the level of IFN-γin the ***-γin response to IHCA is decreased with increased cytokine utilization during cold bronchospasm,which is accompanied by the mobilization of neutrophils and the shift in the cytokine spectrum of the respiratory tract towards the T helper cells(Th)1 immune response.

目的探讨温度感受器瞬时受体电位蛋白-8(transient receptor potential melastatin 8,TRPM8)在人气道上皮细胞感受冷刺激过程中发挥的作用及其机制。方法用薄荷醇及冷空气(18℃)刺激人气道上皮16HBE细胞:1以TRPM8通道特异性拮抗剂BCTC、TRPM8 shRNA为干预手段,动态观察在薄荷醇、冷刺激作用下细胞内Ca2+荧光强度变化来监测气道上皮细胞上TRPM8的功能;2以磷脂酰肌醇4,5-二磷酸(phosphatidylinositol 4,5-bisphosphate,PIP2)荧光分子探针PLCδ1-PH-GFP转染细胞,通过动态观察各组细胞内PLCδ1-PH-GFP从细胞膜至细胞质的荧光转位变化来检测PIP2的动态变化来研究冷刺激作用下TRPM8的信号通路。结果 1细胞内相对钙离子浓度变化:薄荷醇组处理4 min后、冷刺激组处理6 min后分别为(5.80±0.34)、(5.02±0.36),明显高于对照组(1.04±0.12)、(1.03±0.08)(P<0.05),1 mmol/L薄荷醇组+BCTC组、1 mmol/L薄荷醇组+转染TRPM8 shRNA组最高值分别为(2.68±0.19)、(1.08±0.16),明显低于1 mmol/L薄荷醇组,18℃+BCTC组、18℃+转染TRPM8 shRNA组最高值分别为(1.77±0.22)、(1.02±0.18),明显低于18℃组(均P<0.05);2PLCδ1-PH-GFP从细胞膜至细胞质的转位变化(Fm/Fc值):1 mmol/L薄荷醇组、冷刺激组最高值分别为(0.90±0.01)、(0.90±0.01),明显高于对照组最高值(1.01±0.02)(P<0.05),细胞处理200 s时1 mmol/L薄荷醇+BCTC15μmol/L组、1 mmol/L薄荷醇组+转染TRPM8 shRNA组分别为(0.86±0.03)、(0.89±0.02),明显低于1 mmol/L薄荷醇组(0.36±0.06)(P<0.05),细胞处理220 s时18℃+BCTC15μmol/L组、18℃+转染TRPM8 shRNA组分别为(0.82±0.02)、(0.93±0.01),明显低于18℃组(0.40±0.06)(P<0.05)。结论TRPM8受体是人气道上皮细胞感受冷刺激的主要受体,并通过TRPM8→Ca2+→PLC→PIP2信号通路引起气道黏液高分泌等反应。
