Detection of arboviruses in Culicoides(Diptera:Ceratopogonidae)collected from animal farms in the border areas of Yunnan Province,China

Biting midges of the genus Culicoides(order Diptera,family Ceratopogonidae)are potential biological vectors for the transmission of certain arboviruses among humans,livestock,and wild animals.This study collected a total of 405 Culicoides individuals from seven animal farms located in five counties in the border areas of Yunnan Province,China,and examined the Culicoides species composition and the major arboviruses carried by the Culicoides species.The collected Culicoides were classified into seven species with variable abundances:Culicoides arakawae(5.43%,22/405),Culicoides homotomus(1.23%,5/405),Culicoides obsoletus(19.75%,80/405),Culicoides orientalis(17.28%,70/405),Culicoides oxystoma(29.38%,119/405),Culicoides peregrinus(5.68%,23/405),and Culicoides nipponensis(21.23%,86/405).Among the seven species,C.oxystoma and C.nipponensis were distributed in all the five counties with abundances of 13.33–44.87%and 10.00–46.83%,respectively,suggesting that these were the dominant species of Culicoides widespread on animal farms in the border areas.PCR was used to detect major arboviruses in the collected Culicoides specimens,including bluetongue virus(BTV),Japanese encephalitis virus,Dengue virus,Zika virus,African swine fever virus,and African horse sickness virus.Among the tested viruses,only BTV serotype 1 was tested positive in C.oxystoma specimens collected from a buffalo farm.Culicoides oxystoma was the dominant species on animal farms in the sampled areas,but it has not previously been documented as positive for BTV in China.The current results thus suggest that C.oxystoma could be an important vector for BTV transmission in these border areas,which,however,needs to be confirmed by further comprehensive experiments.Overall,the present study provides the first profile of Culicoides species on animal farms in the China,Vietnam,and Myanmar border areas,establishes the prevalence of arboviruses carried by these Culicoides species,and suggests the vector potential of C.oxystoma species for the transmission of BTV.

Human bocavirus in children suffering from acute lower respiratory tract infection in Beijing Children＇s Hospital

Background Human bocavirus （HBoV） is a parvovirus recently found to possibly cause respiratory tract disease in children and adults. This study investigated HBoV infection and its clinical characteristics in children younger than five years of age suffering from acute lower respiratory tract infection in Beijing Children＇s Hospital. Methods Nasopharyngeal aspirates were collected from children suffering from acute lower respiratory tract infection during the winters of 2004 to 2006 （from November through the following February）. HBoV was detected by polymerase chain reaction amplification and virus isolation and the amplification products were sequenced for identification. Results HBoV infection was detected in 16 of 333 study subjects. Coinfections with respiratory syncytial virus were detected in 3 of 16 HBoV positive patients with acute lower respiratory tract infection. The median age for HBoV positive children was 8 months （mean age, 17 months; range, 3 to 57 months）. Among the HBoV positive children, 14 were younger than 3 years old, 9 were younger than 1 year old and 7 were younger than 6 months. These 16 positive HBoV children exhibited coughing and abnormal chest radiography findings and more than 60% of these children had wheezing and fever. Ten children were clinically diagnosed with pneumonia, 2 bronchiolitis, 2 acute bronchitis and 2 asthma. One child died. Conclusions HBoV was detected in about 5% of children with acute lower respiratory infection seen in Beijing Children＇s HosPital. Further investigations regarding clinical and epidemiologic characteristics of HBoV infection are needed.

Ebola virus disease reemerged in Western Africa in 2014.Chinese Center for Disease Control and Prevention dispatched the first Ebola virus（EBOV）detection team to run newly established Sierra Leone-China Friendship Biological Safety laboratory.The aims of study were to understand epidemiology,clinical manifestations and survival time of EBOV in patient＇s blood.A total of 913specimens were tested between March 11 and April20, 2015. EBOV positivity occurred in 7.37% of the blood and 0.53% in throat swabs.

Parvalbumin-positive retinal ganglion cells(PV+RGCs)are an essential subset of RGCs found in various species.However,their role in transmitting visual information remains unclear.Here,we characterized PV+RGCs in the retina and explored the functions of the PV+RGC-mediated visual pathway.By applying multiple viral tracing strategies,we investigated the downstream of PV+RGCs across the whole brain.Interestingly,we found that the PV+RGCs provided direct monosynaptic input to PV+excitatory neurons in the superficial layers of the superior colliculus(SC).Ablation or suppression of SC-projecting PV+RGCs abolished or severely impaired the flight response to looming visual stimuli in mice without affecting visual acuity.Furthermore,using transcriptome expression profiling of individual cells and immunofluorescence colocalization for RGCs,we found that PV+RGCs are predominant glutamatergic neurons.Thus,our findings indicate the critical role of PV+RGCs in an innate defensive response and suggest a non-canonical subcortical visual pathway from excitatory PV+RGCs to PV+SC neurons that regulates looming visual stimuli.These results provide a potential target for intervening and treating diseases related to this circuit,such as schizophrenia and autism.

The first imported Middle East respiratory syndrome （MERS） case in China was identified in May 2015. We determined the kinetics of antibody （IgG and IgM） and neutralizing antibodies against MERS-coronavirus （MERS-CoV） in this case before discharge. Moreover, no seroconversion was found among 53 close contacts by anti-MERS IgG antibody enzyme-linked immunosorbent assay （ELISA） of paired serum samples. These findings suggest that neither community nor nosocomial transmission of MERS-CoV occurred in China.

Interferon-inducible transmembrane protein 3(IFITM3)inhibits influenza virus infection by blocking viral membrane fusion,but the exact mechanism remains elusive.Here,we investigated the function and key region of IFITM3 in blocking influenza virus entry mediated by hemagglutinin(HA).The restriction of IFITM3 on HAmediated viral entry was confirmed by pseudovirus harboring HA protein from H5 and H7 influenza viruses.Subcellular co-localization and immunocoprecipitation analyses revealed that IFITM3 partially co-located with the full-length HA protein and could directly interact with HA_(2) subunit but not HA_(1) subunit of H5 and H7 virus.Truncated analyses showed that the transmembrane domain of the IFITM3 and HA_(2) subunit might play an important role in their interaction.Finally,this interaction of IFITM3 was also verified with HA_(2) subunits from other subtypes of influenza A virus and influenza B virus.Overall,our data demonstrate for the first time a direct interaction between IFITM3 and influenza HA protein via the transmembrane domain,providing a new perspective for further exploring the biological significance of IFITM3 restriction on influenza virus infection or HA-mediated antagonism or escape.

Nonpharmaceutical interventions(NPIs)have been commonly deployed to prevent and control the spread of the coronavirus disease 2019(COVID-19),resulting in a worldwide decline in influenza prevalence.However,the influenza risk in China warrants cautious assessment.We conducted a cross-sectional,seroepidemiological study in Shandong Province,Northern China in mid-2021.Hemagglutination inhibition was performed to test antibodies against four influenza vaccine strains.A combination of descriptive and meta-analyses was adopted to compare the seroprevalence of influenza antibodies before and during the COVID-19 pandemic.The overall seroprevalence values against A/H1N1pdm09,A/H3N2,B/Victoria,and B/Yamagata were 17.8%(95%CI 16.2%–19.5%),23.5%(95%CI 21.7%–25.4%),7.6%(95%CI 6.6%–8.7%),and 15.0(95%CI 13.5%–16.5%),respectively,in the study period.The overall vaccination rate was extremely low(2.6%).Our results revealed that antibody titers in vaccinated participants were significantly higher than those in unvaccinated individuals(P<0.001).Notably,the meta-analysis showed that antibodies against A/H1N1pdm09 and A/H3N2 were significantly low in adults after the COVID-19 pandemic(P<0.01).Increasing vaccination rates and maintaining NPIs are recommended to prevent an elevated influenza risk in China.

目的　应用RT -PCR方法扩增风疹病毒E1膜蛋白的核苷酸片段作为检测该病毒感染的靶目标。方法　对发疹后 4周 ,ELISA检测风疹病毒感染 (IgM阳性 )的病人血清样品提取RNA ,用RT -PCR方法扩增风疹病毒E1膜蛋白的 14 2bp核苷酸片段。测序并与英国代表株Thomas株的相应核苷酸序列进行比较。结果　 10份样品中 ,4份样品扩增到一条DNA片段。测序结果分析表明 3份样品扩增的风疹病毒E1膜蛋白的核苷酸片段序列与英国代表株Thomas株的相应核苷酸序列有 3个核苷酸的变异。一份样品扩增的风疹病毒E1膜蛋白的核苷酸片段序列与英国代表株Thomas株的相应核苷酸序列 4个核苷酸的差异。结论　该片段的核苷酸序列很稳定 ,适用于作为RT -PCR方法检测该病毒感染的靶目标。

Progress in the mechanism and targeted drug therapy for COPD

Chronic obstructive pulmonary disease(COPD)is emphysema and/or chronic bronchitis characterised by long-term breathing problems and poor airflow.The prevalence of COPD has increased over the last decade and the drugs most commonly used to treat it,such as glucocorticoids and bronchodilators,have significant therapeutic effects;however,they also cause side effects,including infection and immunosuppression.Here we reviewed the pathogenesis and progression of COPD and elaborated on the effects and mechanisms of newly developed molecular targeted COPD therapeutic drugs.Among these new drugs,we focussed on thioredoxin(Trx).Trx effectively prevents the progression of COPD by regulating redox status and protease/anti-protease balance,blocking the NF-κB and MAPK signalling pathways,suppressing the activation and migration of inflammatory cells and the production of cytokines,inhibiting the synthesis and the activation of adhesion factors and growth factors,and controlling the cAMP-PKA and PI3K/Akt signalling pathways.The mechanism by which Trx affects COPD is different from glucocorticoid-based mechanisms which regulate the inflammatory reaction in association with suppressing immune responses.In addition,Trx also improves the insensitivity of COPD to steroids by inhibiting the production and internalisation of macrophage migration inhibitory factor(MIF).Taken together,these findings suggest that Trx may be the ideal drug for treating COPD.

Pseudorabies(PR),also called Aujeszky's disease,is a highly infectious disease caused by pseudorabies virus(PRV).Without specific host tropism,PRV can infect a wide variety of mammals,including pig,sheep,cattle,etc.,thereby causing severe clinical symptoms and acute death.PRV was firstly reported in China in 1950s,while outbreaks of emerging PRV variants have been documented in partial regions since 2011,leading to significant economic losses in swine industry.Although scientists have been devoting to the design of diagnostic approaches and the development of vaccines during the past years,PR remains a vital infectious disease widely prevalent in Chinese pig industry.Especially,its potential threat to human health has also attracted the worldwide attention.In this review,we will provide a summary of current understanding of PRV in China,mainly focusing on PRV history,the existing diagnosis methods,PRV prevalence in pig population and other susceptible mammals,molecular characteristics,and the available vaccines against its infection.Additionally,promising agents including traditional Chinese herbal medicines and novel inhibitors that may be employed to treat this viral infection,are also discussed.
