Ethyl acetate extract of Smilax glabra Roxb roots and its major active compound astilbin promote osteoblastogenesis in vitro by upregulating bone cell differentiation-associated genes

Objective:To investigate the osteoblastogenic activity of the ethyl acetate(EtOAc)extract of Smilax glabra Roxb roots and its major active compound astilbin.Methods:Astilbin was isolated from EtOAc extract using silica gel chromatography combined with fraction crystallization.Chemical structure of astilbin was determined by analysis of the spectroscopic data in comparison with the literature.MTT method was used to detect the toxicity.Alkaline phosphatase(ALP)activity was determined by the spectrophotometric method at 405 nm using p-nitrophenyl phosphate as a substrate.Calcium deposition was stained with alizarin red-S,distained with cetylpyridium chloride,and quantified at 562 nm.In silico model for astilbin-ALP interaction was analyzed using AutoDock 4.2.6.The changes in expression of osteoblast differentiation related genes were determined using quantitative real-time PCR.Results:Both the EtOAc extract and astilbin had no toxicity toward osteoblast MC3T3-E1 cells at 5.0,10,25,and 50μg/mL.At 25μg/mL,they enhanced ALP activity and mineralization of osteoblasts up to 30%and 55%for the EtOAc extract and 22%and 41%for astilbin,respectively.Molecular docking analysis of astilbin-ALP interaction revealed Arg167,Asp320,His324,and His437 were key residues participating in hydrophobic interaction;meanwhile,His434 and Thr436 residues were involved in hydrogen bond formation in the active site of human tissue-nonspecific ALP.Moreover,the expression level of genes opn,col1,osx,and runx2 were up-regulated in astilbin treated samples with the fold changes as 2.2;3.7;4.1;2.3,respectively at 10μg/mL(P<0.05).Conclusions:The EtOAc extract and its major compound astilbin exhibit osteoblastogenic activity by up-regulating important markers for bone cell differentiation.It could be a new and promising osteogenic agent with dual actions for therapeutic applications.

Embryonic, genetic and clinical outcomes of fresh versus vitrified oocyte: A retrospective cohort study

Objective:To compare embryonic development,ploidy status and clinical outcomes between fresh and frozen-thawed oocytes.Methods:This retrospective cohort study evaluated 83 fertilization cycles including both fresh and frozen oocytes from 79 patients at the HP Fertility Center of Hai Phong International Hospital of Obstetrics and Pediatrics in vietnam.The patient underwent several ovarian stimulation cycles to accumulate a certain number of oocytes that would be vitrified.In the last oocyte retrieval,all patient’s oocytes including both frozen and fresh would be fertilized.The outcomes included the rates of oocyte survival,cleavage embryo,blastocyst,ploidy status,pregnancy,biochemical pregnancy and clinical pregnancy.Results:The oocyte survival rate after thawing was 96.5%.No statistically significant difference was found when comparing fresh and frozen oocytes regarding fertilization rate(78.1%vs.75.5%,P=0.461),usable cleavage embryo rate(86.9%vs.87.2%,P=0.916)but usable blastocyst rate was found higher statistically in the frozen oocyte group(44.4%vs.54.0%,P=0.049).The percentages of euploid,aneuploid and mosaic embryos between the fresh group and the vitrified group had no significant differences(33.8%vs.31.6%,P=0.682;51.0%vs.54.2%,P=0.569;15.2%vs.12.4%,P=0.787;respectively).The rates of pregnancy,biochemical pregnancy and clinical pregnancy had no statistical difference(68.8%vs.64.8%,P=0.764;12.5%vs.3.6%,P=0.258;37.5%vs.46.4%,P=0.565).17 Mature oocytes are the minimum to have at least one euploid embryo.Conclusions:Oocyte vitrification does not affect embryonic,genetic and clinical results.The number of mature oocytes should be considered for fertilization in some cases.

Ardisia gigantifolia ethanolic extract inhibits cell proliferation and targets cancer stem cells in gastric cancer

Objective:To evaluate the effects of ethanol extract from Ardisia gigantifolia leaves on cell proliferation and cancer stem cell(CSC)number in gastric cancer.Methods:The inhibitory effect of Ardisia gigantifolia extract on the proliferation of MKN45 and MKN74 gastric cancer cells was assessed using 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide assay.Non-adherent culture(3D)model was used to evaluate the effect of the extract on tumorsphere size and number.Moreover,the expression of CD44,ALDH,and p21 was determined by immunofluorescence analysis.Flow cytometric analysis was performed to evaluate cell cycle arrest and the expression of gastric CSC markers CD44 and ALDH.Real-time PCR analysis was also carried out to assess the effect of the extract on the expression of cell cycle-regulated genes.Results:Ardisia gigantifolia extract effectively inhibited cell proliferation with an IC_(50)of 55.7μg/m L in MKN45 cells and 123.6μg/m L in MKN74 cells.The extract also arrested cell cycle in the G_(0)/G_(1)phase as well as significantly reduced the size and number of tumorspheres.The markedly increased expression of p21 was observed at both m RNA and protein levels in the extract-treated adherent cells and tumorspheres.In addition,Ardisia gigantifolia extract significantly reduced the number of CD44-and/or ALDH-expressing gastric CSC.Conclusions:The development of gastric CSC can be inhibited by the ethanol extract of Ardisia gigantifolia.

Determination of groundwater solute transport parameters in finite element modelling using tracer injection and withdrawal testing data

The groundwater tracer injection and withdrawal tests are often carried out for the determination of aquifer solute transport parameters.However,the parameter analyses encounter a great difficulty due to the radial flow nature and the variability of the temporal boundary conditions.An adaptive methodology for the determination of groundwater solute transport parameters using tracer injection and withdrawal test data had been developed and illustrated through an actual case.The methodology includes the treatment of the tracer boundary condition at the tracer injection well,the normalization of tracer concentration,the groundwater solute transport finite element modelling and the method of least squares to optimize the parameters.An application of this methodology was carried out in a field test in the South of Hanoi city.The tested aquifer is Pleistocene aquifer,which is a main aquifer and has been providing domestic water supply to the city since the French time.Effective porosity of 0.31,longitudinal dispersivity of 2.2 m,and hydrodynamic dispersion coefficients from D=220 m^(2)/d right outside the pumping well screen to D=15.8 m^(2)/d right outside the tracer injection well screen have been obtained for the aquifer at the test site.The minimal sum of squares of the differences between the observed and model normalized tracer concentration is 0.00119,which is corresponding to the average absolute difference between observed and model normalized concentrations of 0.0355(while 1 is the worst and 0 is the best fit).

Attribute reduction,also known as feature selection,for decision information systems is one of the most pivotal issues in machine learning and data mining.Approaches based on the rough set theory and some extensions were proved to be efficient for dealing with the problemof attribute reduction.Unfortunately,the intuitionistic fuzzy sets based methods have not received much interest,while these methods are well-known as a very powerful approach to noisy decision tables,i.e.,data tables with the low initial classification accuracy.Therefore,this paper provides a novel incremental attribute reductionmethod to dealmore effectivelywith noisy decision tables,especially for highdimensional ones.In particular,we define a new reduct and then design an original attribute reduction method based on the distance measure between two intuitionistic fuzzy partitions.It should be noted that the intuitionistic fuzzypartitiondistance iswell-knownas aneffectivemeasure todetermine important attributes.More interestingly,an incremental formula is also developed to quickly compute the intuitionistic fuzzy partition distance in case when the decision table increases in the number of objects.This formula is then applied to construct an incremental attribute reduction algorithm for handling such dynamic tables.Besides,some experiments are conducted on real datasets to show that our method is far superior to the fuzzy rough set based methods in terms of the size of reduct and the classification accuracy.

A Simple Approach to the Fabrication of Graphene-Carbon Nanotube Hybrid Films on Copper Substrate by Chemical Vapor Deposition

In this study,graphene-carbon nanotube（CNT） hybrid films were directly synthesized on polycrystalline copper（Cu） substrates by thermal chemical vapor deposition（CVD） method.Graphene films were synthesized on Cu substrate at 1000 ℃ in mixture of gases:argon（Ar）,hydrogen（H2）,and methane（CH4）.Then,carbon nanotubes（CNTs） were grown uniformly on the surface of graphene/Cu films at 750 ℃ in mixture of Ar,H2.and acetylene（C2H2） gases.Ferric salt FeCl3solution deposited onto the surface of graphene/Cu substrate by spin coating method was used as precursor for the growth of the CNTs.The density and quality of the CNTs on the surface of graphene/Cu films can be controlled by varying the concentration of FeCl3salt catalyst.

We describe a new species of megophryid frog from Phu Yen Province in southern vietnam. Leptolalax macrops sp. nov. is distinguished from its congeners by a combination of the following morphological attributes：（1） body size medium （SVL 28.0-29.3 mm in three adult males, 30.3 mm in single adult female）; （2） supra-axillary glands present, creamy white; ventrolateral glands indistinct; （3） tympanum externally distinct; （4） dorsal skin roughly granular with larger tubercles, dermal ridges on dorsum absent; （5） rudimentary webbing present between fingers I-II and I1-111; rudimentary webbing between all toes; fingers and toes without dermal fringes; （6） in life ventral surface greyish-violet with white speckling; （7） supratympanic fold distinct, dark brown in life; （8） iris bicolored, typically golden in upper half, fading to golden green in lower half; （9） tibia short （TbL/SVL 0.44-0.45 in males）; and （10） eyes large and protuberant （ED/SVL 0.15-0.16 in males） From all congeners for which comparable sequences are available, the new species differs markedly in the 16S rRNA mitochondrial gene sequence （P-distance〉5.7%）. The new species is currently known only from montane evergreen tropical forests of Song Hinh District, Phu Yen Province, and M＇Drak District of Dak Lak Province at elevations of 470-630 m a.s.l.. We suggest the new species should be considered as Data Deficient following the IUCN＇s Red List categories. We also report a previously unknownLeptolalax mtDNA lineage from an evergreen tropica forest in the Hoa Thinh District of Phu Yen Province which may also represent an undescribed species.

An isogeometric approach to free vibration analysis of bi-directional functionally graded porous doubly-curved shallow microshells with variable length-scale parameters

This study uses iso-geometric investigation,which is based on the non-uniform rational B-splines(NURBS)basis function,to investigate natural oscillation of bi-directional functionally graded porous(BFGP)doublycurved shallow microshells placed on Pasternak foundations with any boundary conditions.The characteristics of the present material vary in both thickness and axial directions along the x-axis.To be more specific,a material length-scale coefficient of the microshell varies in both thickness and length directions as the material's mechanical properties.One is able to develop a differential equation system with varying coefficients that regulate the motion of BFGP double-curved shallow microshells by using Hamilton principle,Kirchhoff-Love hypothesis,and modified couple stress theory.The numerical findings are reported for thin microshells that are spherical,cylindrical,and hyperbolic paraboloidal,with a variety of planforms,including rectangles and circles.The validity and effectiveness of the established model are shown by comparing the numerical results given by the proposed formulations with previously published findings in many specific circumstances.In addition,influences of length scale parameters,power-law indexes,thickness-to-side ratio,and radius ratio on natural oscillation responses of BFGP microshells are investigated in detail.

This study measured the yields of the radionuclides^(82)Sr,^(83(m+g))Sr,^(85m)Sr,^(85g)Sr,^(87m)Sr,^(81(g+0.976m))Rb,^(82m)Rb,^(83g)Rb,^(84(m+g))Rb,and^(86(m+g))Rb produced in^(nat)Sr(γ,xnyp)multiparticle reactions with bremsstrahlung end-point energies of 55,60,and 65 MeV.The bremsstrahlung radiation was generated using the 100-MeV electron linear accelerator at the Pohang Accelerator Laboratory,Korea,and the reaction yields were derived from the induced activitiesmeasured using off-lineγ-ray spectrometry.To obtain accurate experimental results,we performed the necessaryγ-ray interference corrections.The experimental results were compared with the theoretical predictions obtained using the TALYS-1.95 statistical nuclear model code.The calculations were performed using six different level-density models to ascertain which model best fitted the experimental results.The dependence of the reaction yield on the incident bremsstrahlung energy and on the number of nucleons emitted by the photonuclear reactions was also investigated.

20-Hydroxyecdysone from Dacrycarpus imbricatus bark inhibits the proliferation of acute myeloid leukemia cells

Objective:To investigate the anti-proliferative effects of 20-hydroxyecdysone isolated from the bark of Dacrycarpus imbricatus(Blume) de Laub.Methods:Column chromatography was used for isolation of compounds from plant material.The structure of the isolated compound was identified by mass spectrometry and nuclear magnetic resonance techniques,including HSQC,HMBC,NOE-difference experiments.The isolated compound was tested for its anti-proliferative activity in acute myeloid leukemia(AML) and OCI-AML cells.Results:Compound 1 was isolated from the ethyl acetate fraction of Dacrycarpus imbricatus barks by column chromatography.Its chemical structure was identified as 20-hydroxyecdysone(20HE),a cholestane-type ecdysteroid,by a combination of mass spectrometry and nuclear magnetic resonance spectrometric analyses.Our goal was to test the anti-proliferative activity of 20 HE using the OCI-AML cell line.20 HE significantly decreased OCI cell number at a concentration of 1 mg/mL,whereas lower concentrations were ineffective.Moreover,this decrease was due to partial blockage of the G_1/S phase of the cell cycle,with a reduction of cells in the G_2M phase,not due to increased apoptosis.Conclusions:This indicates that 20 HE significantly decreases the number of cells in the G_1/S phase of the cell cycle in human AML cells.This is the first time that the anti-proliferative activity of 20 HE against a human tumor cell line has been reported.
