The VP1 protein of foot-and-mouth disease virus serotype A was prokaryotically expressed and purified to replace the traditional virus antigen for estab- lishing a fast, safe, effective indirect ELISA method, so as to detecting antibody of foot-and-mouth disease virus serotype A. Western-Blot test showed that the VP1 recombinant protein could be used as detective antigen as it can be specifically recognized by bovine positive serum of FMDV serotype A. By employing matrix titra- tion method, the optimal parameters were obtained as follows： 1 mg/L VP1 protein as coating antigen, Vserum：Vblocking solution = 1：50 dilution for serum and Vsecondary enzyme-linked antibedies：Vblocking solution ---1：2 000 for enzyme combined antibodies. The results showod that the sensitivity and specificity of this method were 94.32% and 99.09% respectively, the coefficients of variations in intra-assay and inter-assay reproducibility tests was lower than 8%. Compared with liquid phase blocking ELISA kits, the agreement of 201 serum samples reached 92.54%. The VP1-ELISA method established here is specific, sensitive, stable and simple, which can be used to monitor the antibody level of FMD serotype A.

Cryopreservation techniques for mammalian oocytes and embryos have rapidly progressed during the past two decades,emphasizing their importance in various assisted reproductive *** and live births resulting from cryopreserved oocytes and embryos of several species including humans have provided proof of principle and led to the adoption of cryopreservation as an integral part of clinical in vitro *** progress has been achieved in the development and application of the cryopreservation of mammalian oocytes and embryos,including preservation of the reproductive potential of patients who may become infertile,establishment of cryopreserved oocyte banks,and transport of oocytes and embryos ***,the success rates are still far lower than those obtained with fresh oocytes and embryos,and there are still obstacles that need to be *** this review,we address the major obstacles in the development of effective cryopreservation *** knowledge may help to eliminate these hurdles by revealing which aspects need ***,this information may encourage further research by cryobiologists and increase the practical use of cryopreservation as a major part of assisted reproductive technologies for both humans and animal species.

The aim of the present study was to evaluate the interaction of different concentrations of butylated hydroxytoluene (BHT) in tris-based extender on semen quality parameters in post-thawed dog ***-four ejaculates were collected by artificial vagina from eight male Beagle *** semen were diluted with tris-based extender supplemented with different concentrations (0.0 mM, 0.5 mM,1.0 mM,1.5 mM and 2.0 mM) of BHT (BHT0.5,BHT1.0,BHT1.5,BHT2.0 respectively),at a final concentration of 200×10～6 spermatozoa/mL.

AIM:To isolate biliary lipid-carrying vesicles from isolated perfused rat livers after taurohyodeoxycholic acid(THDC)*** lipid vesicles have been implicated in hepatic disease and THDC was used since it increases biliary phospholipid ***:Rat livers were isolated and perfused via the hepatic portal vein with THDC dissolved in Krebs Ringer Bicarbonate solution,pH 7.4,containing 1 mmol/L CaCl2,5 mmol/L glucose,a physiological amino acid mixture,1%bovine serum albumin and 20%(v/v)washed human erythrocytes at a rate of 2000 nmol/min for *** livers were then removed,homogenized and subjected to centrifugation,and the microsomal fraction was obtained and further centrifuged at 350000 g for 90 min to obtain subcellular *** were analyzed for total phospholipid,cholesterol,protein and alkaline phosphodiesterase I(PDE).RESULTS:No significant changes were observed in the total phospholipid,cholesterol and protein contents of the gradient fractions obtained from the microsomal ***,the majority of the gradient fractions(ρ=1.05-1.07 g/mL andρ=1.95-1.23 g/mL)obtained from THDC-infused livers had significantly higher PDE activity compared to the control *** low density gradient fraction(ρ=1.05-1.07 g/mL)which was envisaged to contain the putative vesicle population isolated from THDC-perfused livers had relatively small amounts of phospholipids and protein when compared to the relevant control fractions;however,they displayed an increase in cholesterol and PDE *** phospholipids were also isolated by thin layer chromatography and subjected to fractionation by high performance liquid chromatography;however,no differences were observed in the pattern of the fatty acid composition of the phospholipids isolated from THDC and control perfused *** density gradient fractions(ρ=1.10-1.23 g/mL)displayed an increase in all the parameters measured from both control and THDCinfused ***:No significant c

A new method of experimental infection of ovine progressive pneumonia virus (OPPV), aerosol nebulization (Nb), was compared to intravenous (IV) and oral (PO) methods of experimental infection. Seven month old lambs were given 3.5 × 107 TCID50 of Dubois OPPV LMH19 isolate using IV, PO, or Nb methods and were monitored for infection using cELISA and OPPV quantitative (q) PCR for 35 weeks. Four out of four sheep in the IV group, six out of six sheep in the Nb group, but only two out of six sheep in the PO group became infected by OPPV;whereas the uninoculated controls (n = 2) and a sentinel control (n = 1) remained uninfected during the course of the study. The time to a cELISA or OPPV qPCR positive result in the Nb group was quicker and statistically different from the time to a cELISA or OPPV qPCR positive result in the PO group (cELISA P value = 0.0021 and OPPV qPCR P value = 0.0007). When the Nb and IV groups were compared, sheep became cELISA and OPPV qPCR positive at similar times (cELISA P value = 0.6 and OPPV qPCR P value = 0.1). In addition, sheep became OPPV qPCR positive prior to cELISA in both the IV and Nb groups (IV P value = 0.027 and Nb P value = 0.007). Aerosol nebulization is a more natural experimental method of transmitting OPPV and may be valuable for testing potential vaccines or specific host genetics.

Odontogenic tumours are a heterogeneous group of lesions that develop in the oral cavity region and are characterized by the formation of tumoural structures that differentiate as teeth. Due to the diversity of their histopathological characteristics and clinical behaviour, the classification of these tumours is still under debate. Alterations in morphogenesis pathways such as the Hedgehog,MAPK and WNT/β-catenin pathways are implicated in the formation of odontogenic lesions, but the molecular bases of many of these lesions are still unknown. In this study, we used genetically modified mice to study the role of IKKβ(a fundamental regulator of NF-κB activity and many other proteins) in oral epithelial cells and odontogenic tissues. Transgenic mice overexpressing IKKβ in oral epithelial cells show a significant increase in immune cells in both the oral epithelia and oral submucosa. They also show changes in the expression of several proteins and mi RNAs that are important for cancer development. Interestingly, we found that overactivity of IKKβ in oral epithelia and odontogenic tissues, in conjunction with the loss of tumour suppressor proteins(p53, or p16 and p19), leads to the appearance of odontogenic tumours that can be classified as ameloblastic odontomas, sometimes accompanied by foci of secondary ameloblastic carcinomas. These tumours show NF-κB activation and increased β-catenin *** findings may help to elucidate the molecular determinants of odontogenic tumourigenesis and the role of IKKβ in the homoeostasis and tumoural transformation of oral and odontogenic epithelia.

Acute Toxicity of Oxyclozanide Suspension in Mice by Oral Administration

The safety of oxyclozanidc suspension was preliminarily evaluated through acute toxicity test in mice. Administration dose, formal trial grouping and group interval were determined in pre-trial using incremental method. Formal test was performed using simplified karber＇s method. Changes in sign of mice after ad- ministration were observed; the mortality rate was statistically calculated, and the time of death was recorded; the median lethal dose （LD50） and 95% confidence limit of oxyclozanide suspension were calculated. The results showed the LD50 of oxyclozanide suspension in mice by oral administration was 1. 679 g/kg, and the 95% confidence interval was 1. 439 - 1. 947 g/kg. According to toxicity grading of chemicals, oxyclozanide suspension was low toxic substance.

The bacterium Helicobacter pylori(***)infects the stomachs of approximately 50%of all *** its universal occurrence,high infectivity and virulence properties it is considered as one of the most severe global burdens of modern *** has accompanied humans for many thousands of years,and due to its high genetic variability and vertical transmission,its population genetics reflects the history of human ***,especially complex demographic events such as the colonisation of Europe cannot be resolved with population genetic analysis of modern *** strains *** is best exemplified with the reconstruction of the 5300-year-old *** genome of the Iceman,a European Copper Age *** analysis provided precious insights into the ancestry and evolution of the pathogen and underlined the high complexity of ancient European population *** this review we will provide an overview on the molecular analysis of *** in mummified human remains that were done so far and we will outline methodological advancements in the field of ancient DNA research that support the reconstruction and authentication of ancient *** genome sequences.
