G protein-coupled receptor kinase 2 （GRK2） is an important serine/threonine-kinase regulating different membrane receptors and intraceUular proteins. Attenuation of Drosophila Gprk2 in embryos or adult flies induced a defective differentiation of somatic muscles, loss of fibers, and a flightless phenotype. In vertebrates, GRK2 hemizygous mice contained less but more hypertrophied skeletal muscle fibers than wild-type littermates. In C2C12 myoblasts, overexpression of a GRK2 kinase-deficient mutant （K220R） caused precocious differentiation of ceUs into immature myotubes, which were wider in size and contained more fused nuclei, while GRK2 overexpression blunted differentiation. Moreover, p38MAPK and Akt pathways were activated at an earlier stage and to a greater extent in K220R-expressing cells or upon kinase downregulation, while the activation of both kinases was impaired in GRK2-overexpressing cells. The impaired differentiation and fewer fusion events promoted by enhanced GRK2 levels were recapitulated by a p38MAPK mutant, which was able to mimic the inhibitory phosphorylation of p38MAPK by GRK2, whereas the blunted differentiation observed in GRK2-expressing clones was rescued in the presence of a constitutively active upstream stimulator of the p38MAPK pathway. These results suggest that balanced GRK2 function is necessary for a timely and complete myogenic process.

目的甲状腺髓样癌(MTC)占甲状腺癌的5%～8%。血清降钙素和癌胚抗原(CEA)的测定已被广泛应用于MTC进展的监测。但临床仍希望有能够预测预后的相关检查。该研究的目的是评价F-多巴(DOPA)PET/CT显像在MTC复发患者中的预后预测价值。方法 60例MTC复发患者[年龄44～82岁,平均(64±13)岁]行F-DOPAPET/CT显像用于再分期,显像时间在手术后至少半年。记录患者的CEA和降钙素水平,PET/CT显像所示局部复发、淋巴结受累及远处转移情况。以峰值活度的40%作为阈值对每个病灶自动设置感兴趣容积(VOI),计算代谢性肿瘤体积(MTV)、最大标准摄取值(SUV)、平均标准摄取值(SUV);同时以纵隔放射性摄取计算标化SUV和SUV,即nSUV和nSUV。所有患者行临床和放射影像学随访(21±11)个月。使用Kaplan-Meier生存率曲线和时序检验评价无进展生存(PFS)率、疾病特异性生存(DSS)率,分析FDOPA PET/CT显像较常规影像学检查的预后预测增益价值。对预后预测的相关因素采用单变量和多变量Cox回归分析法进行评价。结果F-DOPA PET/CT显像发现27例患者有异常病灶(45%),其余33例未见明显异常(55%)。PET/CT显像未见异常的患者PFS、DSS均高于有异常病灶的患者(P=0.018、0.04)。与常规影像学检查比较,F-DOPA PET/CT显像对PFS和DSS的预测有增益价值(P<0.001、P=0.012)。半定量的PET参数和临床资料或实验室检查结果均无法预测MTC复发患者PFS和DSS的降低。结论F-DOPA PET/CT显像在MTC复发患者疾病进展和死亡率上有预后预测价值。

Apple(Malus domestica Borkh)is an important fruit crop cultivated in a broad range of environmental *** fruit ripening is a physiological process,whose molecular regulatory network response to different environments is still not sufficiently investigated and this is particularly true of the peel *** this study,the influence of environmental conditions associated with low(20 m)and high(750 m)altitude on peel tissue ripening was assessed by physiological measurements combined with metabolomic and proteomic analyses during apple fruit development and *** apple fruit ripening was itself not affected by the different environmental conditions,several key color parameters,such as redness and color index,were notably induced by high *** with this observation,increased levels of anthocyanin and other phenolic compounds,including cyanidin-3-O-galactoside,quercetin-3-O-rhamnoside,quercetin-3-O-rutinoside,and chlorogenic acid were identified in the peel of apple grown at high ***,the high-altitude environment was characterized by elevated abundance of various carbohydrates(e.g.,arabinose,xylose,and sucrose)but decreased levels of glutamic acid and several related proteins,such as glycine hydroxymethyltransferase and glutamate–glyoxylate *** processes affected by high altitude were the TCA cycle,the synthesis of oxidative/defense enzymes,and the accumulation of photosynthetic *** the obtained data we were able to construct a metabolite-protein network depicting the impact of altitude on peel *** combined analyses presented here provide new insights into physiological processes linking apple peel ripening with the prevailing environmental conditions.

Vitamin D deficiency has been indicated as a pandemicemerging public health problem. In addition to the well-known role on calcium-phosphorus homeostasis in thebone, vitamin D-mediated processes have been recentlyinvestigated on other diseases, such as infections, can-cer and cardiovascular diseases. Recently, both the dis-covery of paracrine actions of vitamin D(recognized as"local vitamin D system") and the link of vitamin D with renin-angiotensin-aldosterone system and the fibroblast growth factor 23/klotho pathways highlighted its ac-tive cardiovascular activity. Focusing on hypertension, this review summarizes the more recent experimental evidence involving the vitamin D system and deficiency in the cardiovascular pathophysiology. In particular, we updated the vascular synthesis/catabolism of vitamin D and its complex interactions between the various endocrine networks involved in the regulation of blood pressure in humans. On the other hand, the conflicting results emerged from the comparison between obser-vational and interventional studies emphasize the frag-mentary nature of our knowledge in the field of vitamin D and hypertension, strongly suggesting the need of further researches in this field.

This paper is a pilot study that investigates the attitudes towards the official recognition of Hong Kong Sign Language(HKSL)by Hong Kong *** used video-chat software(mainly WhatsApp,and Facebook Messenger,but also FaceTime)to conduct long-distance semi-structured interviews with 30 participants grouped as deaf,hearing-related(hearing people that are closely involved in the Deaf community),and hearing-unrelated(hearing people that have little contact with deaf people and the Deaf community).Results show that the majority of participants(N=22)holds a supportive attitude towards the recognition of HKSL;Five participants hold a neutral position,and three participants hold a negative attitude towards *** discussed each type of attitude in *** show that participants’attitudes are positively related to their awareness of deaf people’s need,the understanding of‘language recognition’,and personal world *** other words,the more participants are aware,the more they foster official recognition,at least as a general *** also indicate that hearing people who are not involved in the Deaf community know very little about deaf people and the Deaf community,in *** the end of the paper,we also reflect on two issues:we argue that the standardization of HKSL plays an important role in deaf education and empowering citizenship awareness and participation.
