A reproducible and sensitive method for generating high-quality transcriptomes from single whitefly salivary glands and other low-input tissues

Transcriptomic studies are an important tool for understanding the molecular pathways underlying host plant use by agricultural pests,including vectors of damaging plant *** far,bulk RNA-Seq has been the main approach for non-model *** method relies on pooling large numbers of whole organisms or hundreds of individually dissected *** latter approach is logistically challenging,may introduce artifacts of handling and storage,and is not compatible with biological ***,we tested an approach to generate transcriptomes of individual salivary glands and other low-input body tissues from whiteflies(Bemisia tabaci MEAM1),which are major vectors of plant *** comparing our outputs to published bulk RNA-Seq datasets for whole whitefly bodies and pools of salivary glands,we demonstrate that this approach recovers similar numbers of transcripts relative to bulk RNA-Seq in a tissue-specific manner,and for some metrics,exceeds performance of bulk tissue *** generated from individual salivary glands also yielded additional novel transcripts not identified in pooled salivary gland datasets,and had hundreds of enriched transcripts when compared with whole head ***,our study demonstrates that it is feasible to produce high quality,replicated transcriptomes of whitefly salivary glands and other low-input *** anticipate that our approach will expand hypothesis-driven research on salivary glands of whiteflies and other Hemiptera,thus enabling novel control strategies to disrupt feeding and virus transmission.

Tip-enhanced Raman spectroscopy(TERS)imaging is a super-resolution imaging technique that features the merits of both surface-enhanced Raman spectroscopy(SERS)and scanning probe microscopy(SPM),such as the high chemical sensitivity from the former and the nanoscale spatial resolution from the *** advantages make TERS an essential nanospectroscopic characterization technique for chemical analysis,materials science,bio-sensing,*** probes,the most critical factor determining the TERS imaging quality,are expected to provide a highly confined electromagnetic hotspot with a minimized scattering background for the generation of Raman signals with high spatial *** two decades of development,numerous probe design concepts have been proposed and *** review provides a comprehensive overview of the state-of-the-art TERS probe designs,from the working mechanism to the practical *** start with reviewing the recent development of TERS configurations and the corresponding working mechanisms,including the SPM platforms,optical excitation/collection techniques,and probe preparation *** then review the emerging novel TERS probe designs,including the remote-excitation probes,the waveguide-based nanofocusing probes,the metal-coated nanofocusing probes,the nanowire-assisted selective-coupling probes,and the tapered metal-insulator-metal *** discussion focuses on a few critical aspects,including the surface-plasmon-polariton(SPP)hotspot excitation technique,conversion efficiency,working frequency,and *** the end,we review the latest TERS applications and give a perspective on the future of TERS.

This study assessed the effectiveness of three novel control technologies for particulate matter（PM） and volatile organic compound（VOC） removal from commercial meat cooking operations. All experiments were conducted using standardized procedures at university of california, riverside＇s commercial test cooking facility. PM mass emissions collected using South Coast Air Quality Management District（SCAQMD） Method 5.1, as well as a dilution tunnel-based PM method showed statistically significantly reductions for each control technology when compared to baseline testing（i.e., without a catalyst）. Overall, particle number emissions decreased with the use of control technologies, with the exception of control technology 2（CT2）, which is a grease removal technology based on boundary layer momentum transfer（BLMT） theory. Particle size distributions were unimodal with CT2 resulting in higher particle number populations at lower particle diameters. Organic carbon was the dominant PM component（ 99%） for all experiments. Formaldehyde and acetaldehyde were the most abundant carbonyl compounds and showed reductions with the application of the control technologies. Some reductions in mono-aromatic VOCs were also observed with CT2 and the electrostatic precipitator（ESP） CT3 compared to the baseline testing.
