Translating bacteriophage-derived depolymerases into antibacterial therapeutics:Challenges and prospects

Predatory bacteriophages have evolved a vast array of depolymerases for bacteria capture and *** depolymerases are enzymes responsible for degrading diverse bacterial surface *** are exploited as antibiofilm agents and antimicrobial adjuvants while rarely inducing bacterial resistance,making them an invaluable asset in the era of antibiotic *** depolymerases have been investigated preclinically,with evidence indicating that depolymerases with appropriate dose regimens can safely and effectively combat different multidrug-resistant pathogens in animal infection ***,some formulation approaches have been developed for improved stability and activity of ***,depolymerase formulation is limited to liquid dosage form and remains in its infancy,posing a significant hurdle to their clinical translation,compounded by challenges in their applicability and *** development must address these obstacles for clinical ***,after unravelling the history,diversity,and therapeutic use of depolymerases,we summarized the preclinical efficacy and existing formulation findings of recombinant ***,the challenges and perspectives of depolymerases as therapeutics for humans were assessed to provide insights for their further development.

Resource recycling from waste-water and sludge is an important part of the 14th Five-Year Plan in *** emerging titanium-based coagulants have drawn growing attentions due to their strong coagulation capability in water purification and value-added Ti-loaded sludge *** and recovery of the high value-added sludge into functional nanomaterials is highly significant for both sludge reduction and environmental *** present study was carried out to investigate the recycle of the coagulated Ti-loaded sludge to produce functional C/TiO_(2)composites as the anode materials for lithium-ion batteries(LIBs).It is the first time that the application of the Ti-loaded wastewater sludge derived C/TiO_(2)was evaluated for *** experimental results showed that the carbon coating through in-situ carbonization of the sludge produced the C/TiO_(2)composites with a high specific surface area,stable structural integrity,and excellent electrochemical properties that would facilitate Li+diffusion in long-term LIBs *** C/TiO_(2)composites calcinated from the polytitanium sulfate-coagulated sludge at 800℃(N_(2))exhibited the best electrochemical performance during the cycling tests(601 m Ah/g at 100 m A/g after 200 cycles).The research work demonstrates the promising prospect of the recycle and value-added utilization of the Ti-loaded sludge in the production of high-performance C/TiO_(2)composites for energy storage *** study provides a new way for the management and reuse of Ti-loaded waste-sludge.

Background Soil erosion affects the stability of terrestrial ecosystems and ecosystem services by directly or indirectly impacting the cycling of soil materials and energy and reducing the fertility of grassland soils. However, research on microbial adaptation to grazing and soil erosion is limited, particularly in relation to grassland ecosystem restora-tion. Here, we assess microbial communities subjected to simulated soil erosion and grazing in a semi-arid grassland of Inner Mongolia, *** No significant change was observed in soil variables. However, the structure of the soil microbial commu-nity underwent significant changes as a result of soil erosion and soil erosion plus grazing, leading to a significant increase in the relative abundance of Cyanobacteria(116.80% vs 116.38%). Wind erosion and deposition contributed to an increase in the network complexity of soil bacterial and fungal communities. However, much of this effect was alleviated by grazing. Simultaneously, aeolian processes and grazing regulate soil microbial community assembly, leading to inconsistent patterns of change in bacterial and fungal communities. Under wind erosion and deposi-tion, the relative contribution of deterministic processes(4.44% vs 31.11%) in bacterial communities increased, while the relative contribution of stochastic processes(2.23% vs 20%) in fungal communities reduced. Grazing resulted in a decrease in the relative contribution of deterministic processes(8.89%) in the bacterial community and an increase in the relative contribution of stochastic processes(8.89%) in the fungal *** This study presents a comprehensive investigation of the response of soil microbial communities to aeolian erosion–deposition and grazing in a semi-arid grassland. Aqueous zinc(Zn)batteries with Zn metal anodes are promising clean energy storage devices with intrinsic safety and low ***,Zn dendrite growth severely restricts the use of Zn *** effectively suppress Zn dendrite growth,we propose a bilayer separator consisting of commercial butter paper and glassfiber *** dense cellulose-based butter paper(BP)with low zincophilicity and high mechanical properties prevents the pore-filling behavior of deposited Zn and related separator piercing,effectively suppressing the Zn dendrite *** a result,the bilayer separators endow the ZnjjZn symmetrical batteries with a superlong cycling life of Zn anodes(over 5000 h)at 0.5 mA cm^(-2) and the full batteries enhanced capacity retention,demonstrating the advancement of the bilayer separator to afford excellent cyclability of aqueous metal batteries.

Mutualistic interactions between marine phototrophs and associated bacteria are an important strategy for their success-ful survival in the ocean,but little is known about their metabolic ***,bacterial communities in the algal sphere(AS)and bulk solution(BS)of nine marine red algal cultures were analyzed,and Roseibium and Phycisphaera were identified significantly more abundantly in AS than in *** metabolic features of Roseibium RMAR6-6(isolated and genome-sequenced),Phycisphaera MAG 12(obtained by metagenomic sequencing),and a marine red alga,Porphyridium purpureum CCMP1328(from GenBank),were analyzed ***6-6 has the genetic capability to fix nitrogen and produce B vitamins(B1,B2,B5,B6,B9,and B12),bacterioferritin,dimethylsulfoniopropionate(DMSP),and phenylacetate that may enhance algal growth,whereas MAG 12 may have a limited metabolic capability,not producing vitamins B9 and B12,DMSP,phenylacetate,and siderophores,but with the ability to produce bacitracin,possibly modulating algal *** CCMP1328 lacks the genetic capability to fix nitrogen and produce vitamin B12,DMSP,phenylacetate,and *** was shown that the nitrogen-fixing ability of RMAR6-6 promoted the growth of ***-pureum,and DMSP reduced the oxidative stress of *** metabolic interactions between strain RMAR6-6 and *** CCMP1328 were also investigated by the transcriptomic analyses of their monoculture and *** together,potential metabolic relationships between Roseibium and *** were *** study provides a bet-ter understanding of the metabolic relationships between marine algae and algae-associated bacteria for successful growth.

Mosquito-borne viruses(MBVs)are a large class of viruses transmitted mainly through mosquito bites,including dengue virus,Zika virus,Japanese encephalitis virus,West Nile virus,and chikungunya virus,which pose a major threat to the health of people around the *** global warming and extended human activities,the incidence of many MBVs has increased *** saliva contains a variety of bioactive protein *** not only enable blood feeding but also play a crucial role in regulating local infection at the bite site and the remote dissemination of MBVs as well as in remodeling the innate and adaptive immune responses of host ***,we review the physiological functions of mosquito salivary proteins(MSPs)in detail,the influence and the underlying mechanism of MSPs on the transmission of MBVs,and the current progress and issues that urgently need to be addressed in the research and development of MSP-based MBV transmission blocking vaccines.

The Five-hundred-meter Aperture Spherical radio Telescope(FAST)has the largest aperture and a 19-beam L-band receiver,making it powerful for investigating the neutral hydrogen atomic gas(HI)in the *** present HiFAST(https://***),a dedicated,modular,and self-contained calibration and imaging pipeline for processing the H?data of *** pipeline consists of frequency-dependent noise diode calibration,baseline fitting,standing wave removal using an FFT-based method,flux density calibration,stray radiation correction,and gridding to produce data *** modules can be combined as needed to process the data from most FAST observation modes:tracking,drift scanning,On-The-Fly mapping,and most of their *** HiFAST,the root-mean-square(RMS)noises of the calibrated spectra from all 19 beams were only slightly(~5%)higher than the theoretical *** results for the extended source M33 and the point sources are consistent with the results from *** moment maps(0,1 and 2)of M33 agree well with the results from the Arecibo Galaxy Environment Survey(AGES)with a fractional difference of less than 10%.For a common sample of 221 sources with signal-to-noise ratio S/N>10 from the Arecibo Legacy Fast ALFA(ALFALFA)survey,the mean value of fractional difference in the integrated flux density,Sint,between the two datasets is approximately 0.005%,with a dispersion of 15.4%.Further checks on the integrated flux density of 23 sources with seven observations indicate that the variance in the flux density of the source with luminous objects(Sint>2.5 Jy km s-1)is less than 5%.Our tests suggest that the FAST telescope,with the efficient,precise,and user-friendly pipeline HiFAST,will yield numerous significant scientific findings in the investigation of the H?in the universe.
