Spatiotemporal regulation of gene expression by polycomb repressive complex 2(PRc2)is critical for animal and plant *** Arabidopsis fertilization independent seed(FIS)-PRC2 complex functions specifically during plant reproduction from gametogenesis to seed *** a double fertilization event,triploid endosperm proliferates early,followed by the growth of a diploid embryo,which replaces the endosperm in Arabidopsis and many *** genes critical for endosperm proliferation such as IKkU2 and MINI3 are activated after *** we report that two MADS-box AGAMOUS-LIKE(AGL)proteins associate with the key endosperm proliferation loci and recruit the FIS-PRC2 repressive complex at 4-5 days after pollination(DAP).Interestingly,AGL9 and AGL15 only accumulate toward the end of endosperm proliferation at 4-5 DAP and promote the deposition of H3K27me3 marks at key endosperm proliferation *** of AGL9 and AGL15 or overexpression of AGL9 or AGL15 significantly influence endosperm proliferation and ***-wide analysis with cleavage Under Targets and tagmentation(CUT&Tag)sequencing and RNA sequencing revealed the landscape of endosperm H3K27me3 marks and gene expression profiles in Col-0 and agl9 ***&Tag qPCR also demonstrated the occupancy of the two MADS-box proteins and FIS-PRC2 on a few representative target *** studies suggest that MADS-box proteins could potentially recruit PRc2 to regulate many other developmental processes in plants or even in fungi and animals.

The progressive degradation in the trabecular meshwork(TM)is related to age-related ocular diseases like primary open-angle ***,the molecular basis and biological significance of the aging process in TM have not been fully ***,we established a dynamic single-cell transcriptomic landscape of aged macaque TM,wherein we classified the outflow tissue into 12 cell subtypes and identified mitochondrial dysfunction as a prominent feature of TM ***,we divided TM cells into 13 clusters and performed an in-depth analysis on cluster O,which had the highest aging score and the most significant changes in cell proportions between the two ***,we found that the APOE gene was an important differentially expressed gene in cluster O during the aging process,highlighting the close relationship between cell migration and extracellular matrix regulation,and TM *** work further demonstrated that silencing the APOE gene could increase migration and reduce apoptosis by releasing the inhibition on the PI3K-AKT pathway and downregulating the expression of extracellular matrix components,thereby increasing the aqueous outflow rate and maintaining intraocular pressure within the normal *** work provides valuable insights for future clinical diagnosis and treatment of glaucoma.

The present study aimed to dynamically observe the segmental and global myocardial movements of the left ventricle during coronary artery bypass grafting by transesophageal speckle-tracking echocardiography,and to assess the effect of sevoflurane on cardiac ***-four patients scheduled for the off-pump coronary artery bypass grafting were randomly divided into a sevoflurane-based anesthesia(AS)group and a propofolbased total intravenous anesthesia(AA)*** AS group demonstrated a higher absolute value of left ventricular global longitudinal strain than that of the AA group at both T1(after harvesting all grafts and before coronary anastomosis)and T_(2)(30 min after completing all coronary anastomoses)(P<0.05).Moreover,strain improvement in the segment with the highest preoperative strain was significantly reduced in the AS group,compared with the AA group at both T1 and T_(2)(P<0.01).The flow of the left internal mammary artery-left anterior descending artery graft was superior,and the postoperative concentration of troponin T decreased rapidly in the AS group,compared with the AA group(P<0.05).Compared with total intravenous anesthesia,sevoflurane resulted in a significantly higher global longitudinal strain,stroke volume,and cardiac *** also led to an amelioration in the condition of the arterial ***,sevoflurane significantly reduced strain improvement in the segmental myocardium with a high preoperative strain *** findings need to be replicated in larger studies.

We present an azimuthal-rotation-controlled dynamic nanoinscribing(ARC-DNI)process for continuous and scalable fabrication of asymmetric nanograting structures with tunable periods and shape profiles.A sliced edge of a nanograting mold,which typically has a rectangular grating profile,slides over a polymeric substrate to induce its burrfree plastic deformation into a linear *** this continuous nanoinscribing process,the“azimuthal angle,”that is,the angle between the moving direction of the polymeric substrate and the mold’s grating line orientation,can be controlled to tailor the period,geometrical shape,and profile of the inscribed *** modulating the azimuthal angle,along with other important ARC-DNI parameters such as temperature,force,and inscribing speed,we demonstrate that the mold-opening profile and temperature-and time-dependent viscoelastic polymer reflow can be controlled to fabricate asymmetric,blazed,and slanted nanogratings that have diverse geometrical profiles such as trapezoidal,triangular,and ***,period-and profile-tunable ARC-DNI can be utilized for the practical fabrication of diverse optical devices,as is exemplified by asymmetric diffractive optical elements in this study.

Background Ictal examination based on video-based electroencephalography(EEG)is crucial for locating and lat-eralizing *** this study,we aimed to evaluate the quality of ictal examination in the Comprehensive Epilepsy Center of West China Hospital,Sichuan university,in order to provide information for quality improvement in daily clinical *** Video recordings of 100 patients with epilepsy were retrospectively *** performance of the ictal examination was independently reviewed by two epileptologists using an ictal examination *** In this retrospective analysis,589 seizure episodes from 100 patients with epilepsy were *** ages of the patients ranged from 3 to 77 years,with a mean age of 25.8±12.8 *** the 589 seizure episodes,a majority(93.7%)were focal *** 226(38.4%)seizures,the medical staff arrived at the *** them,153(153/226,64.7%)seizure episodes,the medical staff arrival at the bedside within 30 s of onset,and 120(120/226,53.1%)seizures were tested by the medical *** compliance rates for"safety"and"visibility"reached 80%or higher while"naming","retelling",and“memory testing”only reach less than 3%.Conclusions Our survey identified the main problems in ictal *** is challenging to complete a standard-ized examination for new trainees at Epilepsy Monitoring *** strengthening training in ictal examination and understanding of semiology may improve patients’examination ***,further study of the implemen-tation of training is necessary.
