One ring is sufficient to inhibit α-synuclein aggregation

Parkinson's disease,the second most prevalent neurodegenerative disorder worldwide,is characterized by a progressive loss of dopaminergic neurons in substantia nigra pars compacta,causing motor symptoms.This disorder's main hallmark is the formation of intraneuronal protein inclusions,named Lewy bodies and neurites.The major component of these arrangements is α-synuclein,an intrinsically disordered and soluble protein that,in pathological conditions,can form toxic and cell-to-cell transmissible amyloid structures.Preventing α-synuclein aggregation has attracted significant effort in the search for a disease-modifying therapy for Parkinson's disease.Small molecules like Synu Clean-D,epigallocatechin gallate,trodusquemine,or anle138 b exemplify this therapeutic potential.Here,we describe a subset of compounds containing a single aromatic ring,like dopamine,ZPDm,gallic acid,or entacapone,which act as molecular chaperones against α-synuclein aggregation.The simplicity of their structures contrasts with the complexity of the aggregation process,yet the block efficiently α-synuclein assembly into amyloid fibrils,in many cases,redirecting the reaction towards the formation of non-toxic off-pathway oligomers.Moreover,some of these compounds can disentangle mature α-synuclein amyloid fibrils.Their simple structures allow structure-activity relationship analysis to elucidate the role of different functional groups in the inhibition of α-synuclein aggregation and fibril dismantling,making them informative lead scaffolds for the rational development of efficient drugs.

Protein misfolding and aggregation into amyloid fibrils is the main pathological hallmark of neurodegenerative diseases,including Alzheimer’s,Parkinson’s,Huntington’s,and prion diseases(Chiti and Dobson,2017).These insoluble fibrillar deposits possess a common structure characterized by a cross-β-sheet conformation in which β-strands run transversely to the fiber axis and form an intermolecular network of hydrogen bonds.

The scientific community has focused the agenda of studies of climate change on lower-end warming and simple risk analyses,because more realistic complex assessments of risk are more difficult,the benchmark of the international targets is the Paris Agreement goal of limiting warming to dering,at least,the potential catastrophic effects of the collapse of ecosystems and society due to the current emergency of climate change.

Building the World Wide Hypermap(WWH)with a RESTful architecture

The hypermap concept was introduced in 1992 as a way to hyperlink geospatial features to text,multimedia or other geospatial features.Since then,the concept has been used in several applications,although it has been found to have some limitations.On the other hand,Spatial Data Infrastructures(SDIs)adopt diverse and heterogeneous service oriented architectures(SOAs).They are developed by different standard bodies and are generally disconnected from mass market web solutions.This work expands the hypermap concept to overcome its limitations and harmonise it with geospatial resource oriented architecture(ROA),connecting it to the semantic web and generalising it to the World Wide Hypermap(WWH)as a tool for building a single‘Digital Earth’.Global identifiers,dynamic links,link purposes and resource management capabilities are introduced as a solution that orchestrates data,metadata and data access services in a homogeneous way.This is achieved by providing a set of rules using the current Internet paradigm formalised in the REpresentational State Transfer(REST)architecture and combining it with existing Open Geospatial Consortium(OGC)and International Organization for Standardization(ISO)standards.A reference implementation is also presented and the strategies needed to implement the WWH,which mainly consist in a set of additions to current Geographic Information System(GIS)products and a RESTful server that mediates between the Internet and the local GIS applications.

Small molecules to target tau amyloid aggregation

Protein aggregation has been linked with many neurodegenerative diseases,such as Alzheimer’s disease(AD)or Parkinson’s disease.AD belongs to a group of heterogeneous and incurable neurodegenerative disorders collectively known as tauopathies.They comprise frontotemporal dementia,Pick’s disease,or corticobasal degeneration,among others.The symptomatology varies with the specific tau protein variant involved and the affected brain region or cell type.However,they share a common neuropathological hallmark-the formation of proteinaceous deposits named neurofibrillary tangles.Neurofibrillary tangles,primarily composed of aggregated tau(Zhang et al.,2022),disrupt normal neuronal functions,leading to cell death and cognitive decline.

An acute coronary syndrome can compromise blood flow to certain areas of the heart and cause ischemia and myocardial cell death,which can lead to ventricular dysfunction.If this myocardial injury is large and the ventricular dysfunction is severe,cardiogenic shock(CS)may develop,which results in a life-threatening state of hypoperfusion and critical hypoxia of vital organs.Acute myocardial infarction(AMI)with left ventricular dysfunction is the most common cause of CS(80%of the cases).Other possible causes of CS include myocarditis and acute decompensation of chronic heart failure(HF).

Five-year outcomes of laparoscopic sleeve gastrectomy as a primary procedure for morbid obesity:A prospective study

AIM To prospectively evaluate the postoperative morbimortality and weight loss evolution of patients who underwent a laparoscopic sleeve gastrectomy(LSG) as a primary bariatric procedure during 5 years of follow-up. METHODS Since 2006,data from patients undergoing a highly restrictive primary LSG have been prospectively registered in a database and analysed. Preoperative co-morbid conditions,operating time,hospital stay,early and late complications rate and evolution of weight loss after 5 years of follow-up were analysed.RESULTS A total of 156 patients were included,74.3% of whom were women. The mean age was 43.2 ± 13.1 years and the mean body mass index(BMI) was 41.5 ± 7.9 kg/m^2. Seventy patients(44.8%) presented a BMI under 40 kg/m^2. The mortality rate was 0%. The leakage rate was 1.2%,and the total 30-d morbidity rate was 5.1%(8/156). With a mean follow-up of 32.7 ± 28.5(range 6-112) mo,the mean percent of excess of weight loss(%EWL) was 82.0 ± 18.8 at 1 year,76.7 ± 21.3 at 3 years and 60.3 ± 28.9 at 5 years. The mean percent of excess of BMI loss(%EBMIL) was 94.9 ± 22.4 at 1 year,89.4 ± 27.4 at 3 years and 74.8 ± 29.4 at 5 years. Patients with preoperative BMI less than 40 kg/m^2 achieved greaterweight loss than did the overall study population. Diabetes remitted in 75% of the patients and HTA improved in 71.7%. CPAP masks were withdrawn in all patients with obstructive sleep apnoea.CONCLUSION LSG built with a narrow 34 F bougie and starting 3 cm from the pylorus proved to be safe and highly effective in terms of weight loss as a stand-alone procedure,particularly in patients with a preoperative BMI lower than 40 kg/m^2.

In recent years,researchers of different communities have increased their efforts in formalizing a set of measurements regularly collected for analysing changes in Drivers,States,Impacts and Responses of a given discipline.In some cases,different actors have converged in a minimum set of Essential Variables(EVs),such as for Climate,Biodiversity or Oceans.The definition of such EVs is an ongoing evolution and in extension(e.g.EVs for water)although some communities have not even started(e.***.agriculture and energy).This paper characterizes the Earth Observation(EO)networks and creates a graph representation of their relations.Secondly,this graph is enriched with the EVs produced by each network creating a knowledge base.Finally,an effort has been done to identify links between EVs and Sustainable development Goals(SDG)indicators in a way that they indirectly connect the EO.An analysis to detect gaps in EO variables due to a lack of observational networks is performed.Several suggestions for improving SDG indicators framework by considering EVs are exposed,as well as proposing new necessary EVs and suggesting new EO based indicators.The complete graph is available in the ENEON website(http://***/graphev-sdg/).

developing spatially and thematically detailed backdated maps for land cover studies

Global or regional land cover change on a decadal time scale can be studied at a high level of detail using the availability of remote sensing data such as that provided by Landsat.However,there are three main technical challenges in this goal.First,the generation of land cover maps without reference data is problematic(backdating).Second,it is important to maintain high accuracies in land cover change map products,requiring a reasonably rich legend within each map.Third,a high level of automation is necessary to aid the management of large volumes of data.This paper describes a robust methodology for processing time series of satellite data over large spatial areas.The methodology includes a retrospective analysis used for the generation of training and test data for historical periods lacking reference information.This methodology was developed in the context of research on global change in the Iberian Peninsula.In this study we selected two scenes covering geographic regions that are representative of the Iberian Peninsula.For each scene,we present the results of two classifications(1985-1989 and 2000-2004 quinquennia),each with a legend of 13 categories.An overall accuracy of over 92%was obtained for all 4 maps.
