Hypothalamic circuits and aging:keeping the circadian clock updated

Over the past century,age-related diseases,such as cancer,type-2 diabetes,obesity,and mental illness,have shown a significant increase,negatively impacting overall quality of life.Studies on aged animal models have unveiled a progressive discoordination at multiple regulatory levels,including transcriptional,translational,and post-translational processes,resulting from cellular stress and circadian derangements.The circadian clock emerges as a key regulator,sustaining physiological homeostasis and promoting healthy aging through timely molecular coordination of pivotal cellular processes,such as stem-cell function,cellular stress responses,and inter-tissue communication,which become disrupted during aging.Given the crucial role of hypothalamic circuits in regulating organismal physiology,metabolic control,sleep homeostasis,and circadian rhythms,and their dependence on these processes,strategies aimed at enhancing hypothalamic and circadian function,including pharmacological and non-pharmacological approaches,offer systemic benefits for healthy aging.Intranasal brain-directed drug administration represents a promising avenue for effectively targeting specific brain regions,like the hypothalamus,while reducing side effects associated with systemic drug delivery,thereby presenting new therapeutic possibilities for diverse age-related conditions.

Background High dietary carbohydrates can spare protein in rainbow trout(Oncorhynchus mykiss)but may affect growth and health.Inulin,a prebiotic,could have nutritional and metabolic effects,along with anti-inflammatory properties in teleosts,improving growth and welfare.We tested this hypothesis in rainbow trout by feeding them a 100%plant-based diet,which is a viable alternative to fishmeal and fish oil in aquaculture feeds.In a two-factor design,we examined the impact of inulin(2%)as well as the variation in the carbohydrates(CHO)/plant protein ratio on rainbow trout.We assessed the influence of these factors on zootechnical parameters,plasma metabolites,gut microbiota,production of short-chain fatty acids and lactic acid,as well as the expression of free-fatty acid receptor genes in the mid-intestine,intermediary liver metabolism,and immune markers in a 12-week feeding trial.Results The use of 2%inulin did not significantly change the fish intestinal microbiota,but interestingly,the high CHO/protein ratio group showed a change in intestinal microbiota and in particular the beta diversity,with 21 bacterial genera affected,including Ralstonia,Bacillus,and 11 lactic-acid producing bacteria.There were higher levels of butyric,and valeric acid in groups fed with high CHO/protein diet but not with inulin.The high CHO/protein group showed a decrease in the expression of pro-inflammatory cytokines(il1b,il8,and tnfa)in liver and a lower expression of the genes coding for tight-junction proteins in mid-intestine(tjp1a and tjp3).However,the 2%inulin did not modify the expression of plasma immune markers.Finally,inulin induced a negative effect on rainbow trout growth performance irrespective of the dietary carbohydrates.Conclusions With a 100%plant-based diet,inclusion of high levels of carbohydrates could be a promising way for fish nutrition in aquaculture through a protein sparing effect whereas the supplementation of 2%inulin does not appear to improve the use of CHO when combined with a 100%plant-based diet.

Comparison of Ashes Produced in a Biomass Moving Grate Boiler by Wood Chips and Sewage Sludge

One option to fight global warming is to convert our use of fossil energy into renewables such as biomass energy.However,the forest preservation and the quality of the ambient air are also two major issues.Therefore,the use of biomass waste without any supplementary emissions could represent a part of the solution.In this study,two fuels were considered for a 200 kW moving grate boiler.A multicyclone and a bag filter were fitted on the boiler.The first fuel consisted of classical wood chips whereas the second was a mixture of wood chips with sewage sludge.This second fuel presented a high ashes mass ratio compared to wood chips.The aim was to verify the possibility to burn this kind of fuel without any modification of the installation.The first relevant result is that the conventional pollutants,i.e.,CO and NOx,remained under the emissions limits even with the sewage sludge combustion.The Total Suspended Particles emissions at the exhaust were always under 5.4 mg·Nm-3 dry based corrected at 6%of O_(2),which is low with respect to the standard limitation.The majority of the ashes remained on the combustion room.However,with both fuels,about 5%of ashes mass remained in the heat exchanger.Nevertheless,the heat exchanger was more clogged with the second fuel,which produced five time more ashes.This may lead to a yield loss.Thus,sewage sludge can be used in a wood boiler without any issue if an automatic exchanger sweep is fitted on the installation.

Strain engineering,as a powerful strategy to tune the optical and electrical properties of two-dimensional(2D)materials by deforming their crystal lattice,has attracted significant interest in recent years.2D materials can sustain ultra-high strains,even up to 10%,due to the lack of dangling bonds on their surface,making them ideal brittle solids.This remarkable mechanical resilience,together with a strong strain-tunable band structure,endows 2D materials with a broad optical and electrical response upon strain.However,strain engineering based on 2D materials is restricted by their nanoscale and strain quantification troubles.In this study,we have modified a homebuilt three-points bending apparatus to transform it into a four-points bending apparatus that allows for the application of both compressive and tensile strains on 2D materials.This approach allows for the efficient and reproducible construction of a strain system and minimizes the buckling effect caused by the van der Waals interaction by adamantane encapsulation strategy.Our results demonstrate the feasibility of introducing compressive strain on 2D materials and the potential for tuning their optical and physical properties through this approach.

Climate Change-Related Disaster Risk Events in Togo: A Systematic Review

Togo is facing significant climate challenges that have profound consequences for its environment, economy, and population. This study provides an overview of various climate phenomena affecting Togo and highlights potential adaptation strategies. We used the inclusion and exclusion criteria (PRISMA) to search both French and English articles on climate change-related disaster risk events in Togo through Google Scholar, Directory of Open Access Journals (DOAJ), and PubMed databases using the keywords “Climate Change”, “Floods”, “Drought”, “Coastal erosion”, “High winds”, “Epidemy”, Heatwaves”, and “Air pollution”. Twenty-five articles from 2000-2023 were included in this study after applying different criteria. Droughts, floods, coastal erosion, food and crop productivity loss, heatwaves, spread of vector-borne diseases, air pollution, and high winds are among the climate phenomena discussed. These challenges are driven by climate change, altering precipitation patterns, increasing temperatures, and rising sea levels. Drought, floods, coastal erosion, loss of food and crop productivity, spread of vector-borne diseases, air pollution and heatwaves are the most climate risks experienced by Togo. Drought contributes to decreased plant cover, water scarcity, and changes in the water and energy balance. Floods cause property damage, health risks, and disruptions to livelihoods. Coastal erosion threatens coastal communities, infrastructure, and ecosystems. Adaptation strategies include early warning systems, improved water management, sustainable agriculture, urban and health planning, and greenhouse gas emissions reduction. Drought-resistant crops, mosquito control, and clean energy adoption are essential.

Aim: Sub-Saharan Africa is undergoing an epidemiological transition responsible for a change in the metabolic profile in favour of insulin resistance. The aim of this study was to assess the dynamics of the prevalence of insulin resistance and associated risk factors in diabetic patients in the democratic Republic of Congo between 2005 and 2023. Method: We measured fasting blood glucose and insulin levels and looked for metabolic syndrome parameters (2009 criteria) in type 2 diabetes patients in 2005-2008 (n = 176) and in 2018-2023 (n = 303). The HOMA model was used to measure insulin sensitivity and islet β-cell secretory function. Results: Between 2005 and 2013, the trend was towards an increase in the prevalence of insulin resistance (from 13.1% to 50.8%;p Conclusion: This present study shows an increase in insulin resistance in Congolese urban areas and a persistence of atypical diabetes mellitus in Congolese rural areas, confirming the particularity of the pathophysiology of the disease in African areas currently influenced by the epidemiological transition. Further studies using an appropriate methodology are required.

Background: The role of vitamin D and parathyroid hormone in the metabolic profile of type 2 diabetes mellitus in sub-Saharan Africa has not been adequately assessed. The aim of this study was to determine the prevalence of low vitamin D level and secondary hyperparathyroidism and their association with insulin sensitivity and β-cell secretory function among Congolese type 2 diabetics. Methodology: Fasting glycaemia, fasting insulin, 25OH D3 and human parathyroid hormone (hPTH) were measured in one hundred and eighty-four type 2 diabetic patients followed as outpatients in South Kivu. Levels of 25OH D3 65 pg/ml defined low vitamin D and elevated parathyroid hormone levels, respectively. The HOMA model was used to measure insulin sensitivity and β-cell secretory function. Results: Medians (IQR) were 25.3 (20.4 - 32.4) ng/ml for 25OH D3 and 53.7 (38.4 - 115.7) pg/ml for hPTH. 58.7% of diabetics had insulin resistance, 126 (68.5%) had low vitamin D and 80 (43.5%) had hyperparathyroidism. In multivariate analysis, hPTH (partial r = −0.28;p = 0.0002) and 25OH D3 (partial r = 0.16;p = 0.03) showed an independent association with insulin sensitivity after adjustment for body mass index and waist circumference. Finally, hPTH (partial r = 0.27;p = 0.0002) was the sole determinant of β-cell secretory function. Conclusions: This study confirms the high prevalence of low vitamin D level and secondary hyperparathyroidism and their association with insulin resistance and impaired islet β-cell secretory function among Congolese with type 2 diabetes mellitus. Vitamin D and calcium supplementation should be envisaged for cases of deficiency in this region.

Kinematic analysis of flexible bipedal robotic systems

In spite of its intrinsic complexities,the passive gait of bipedal robots on a sloping ramp is a subject of interest for numerous researchers.What distinguishes the present research from similar works is the consideration of flexibility in the constituent links of this type of robotic systems.This is not a far-fetched assumption because in the transient(impact)phase,due to the impulsive forces which are applied to the system,the likelihood of exciting the vibration modes increases considerably.Moreover,the human leg bones that are involved in walking are supported by viscoelastic muscles and ligaments.Therefore,for achieving more exact results,it is essential to model the robot links with viscoelastic properties.To this end,the Gibbs-Appell formulation and Newton's kinematic impact law are used to derive the most general form of the system's dynamic equations in the swing and transient phases of motion.The most important issue in the passive walking motion of bipedal robots is the determination of the initial robot configuration with which the system could accomplish a periodic and stable gait solely under the effect of gravitational force.The extremely unstable nature of the system studied in this paper and the vibrations caused by the impulsive forces induced by the impact of robot feet with the inclined surface are some of the very serious challenges encountered for achieving the above-mentioned goal.To overcome such challenges,an innovative method that uses a combination of the linearized equations of motion in the swing phase and the algebraic motion equations in the transition phase is presented in this paper to obtain an eigenvalue problem.By solving this problem,the suitable initial conditions that are necessary for the passive gait of this bipedal robot on a sloping surface are determined.The effects of the characteristic parameters of elastic links including the modulus of elasticity and the Kelvin-Voigt coefficient on the walking stability of this type of robotic systems are also studied.The findings o

Reproductive biology of harvestmen(Arachnida:Opiliones):a review of a rapidly evolving research field

Harvestmen are a major arachnid order that has experienced a dramatic increase in biological knowledge in the 21st century.The publication of the book Harvestmen:The Biology of Opiliones in 2007 stimulated the development of many behavioral studies.Although the book is relatively recent,our understanding of the reproductive biology of harvestmen is already outdated due to the fast accumulation of new data.Our goal is to provide an updated review of the subject to serve as a benchmark for the following years.In the pre-copulatory phase,we explore the evolu-tion of facultative parthenogenesis,the factors that may affect the types of mating system,and the role of nuptial gifts in courtship.Regarding the copulatory phase,harvestmen are unique arachnids because they have aflagellate spermatozoa and a penis with complex morphology.We discuss the implications of these two features for sperm competition and cryptic female choice.In the post-copulatory phase,we connect ovi-position site selection and climate conditions to the widespread occurrence of resource defense polygyny,alternative reproductive tactics,and sexual dimorphism in several clades of tropical harvestmen.Finally,we present the different forms of parental care in the order,and discuss the benefits and costs of this behavior,which can be performed either by females or males.Throughout the review,we indicate gaps in our knowl-edge and subjects that deserve further studies.Hopefully,the information synthesized here will stimulate researchers worldwide to embrace harvestmen as a study system and to improve our effort to unravel the mysteries of their reproductive biology.

The potential for an old-growth forest to store carbon in the topsoil:A case study at Sasso Fratino,Italy

There is considerable interest devoted to oldgrowth forests and their capacity to store carbon(C)in biomass and soil.Inventories of C stocks in old-growth forests are carried out worldwide,although there is a lack of information on their actual potential for C sequestration.To further understand this,soil organic carbon(SOC)was measured in one of Italy's best-preserved old-growth forests,the Sasso Fratino Integral Nature Reserve.This reserve is on the World Heritage List along with other ancient beech forests of Europe,and it is virtually untouched due to the steepness of the terrain,even before legal constraints were imposed.Although the sandstone-derived soils are often shallow,they are rich in organic matter.However,no quantification had been carried out.By systematically sampling the topsoil across the forest,we accurately determined the average amount of SOC(62.0±16.9 Mg ha^(–1))and nitrogen(4.0±1.2 Mg ha^(–1))in the top 20 cm.Using the CENTURY model,future dynamics of SOC stocks were predicted to 2050 according to two climate scenarios,A1F1 and B2,the first of high concern and the second more optimistic.The model projected an increase of 0.2 and 0.3 Mg ha^(–1)a^(–1)by 2030 under the A1F1 and B2 scenarios,respectively,suggesting that the topsoil in old-growth forests does not reach equilibrium but continues accumulating SOC.However,from 2030 to 2050,a decline in SOC accumulation is predicted,indicating SOC net loss at high altitudes under the worst-case scenario.This study confirms that soils in oldgrowth forests play a significant role in carbon sequestration.It also suggests that climate change may affect the potential of these forests to store SOC not only in the long term but also in the coming years.
