To metabolomics and beyond:a technological portfolio to investigate cancer metabolism

Tumour cells have exquisite flexibility in reprogramming their metabolism in order to support tumour initiation, progression,metastasis and resistance to therapies. These reprogrammed activities include a complete rewiring of the bioenergetic, biosyntheticand redox status to sustain the increased energetic demand of the cells. Over the last decades, the cancer metabolism field hasseen an explosion of new biochemical technologies giving more tools than ever before to navigate this complexity. Within a cell ora tissue, the metabolites constitute the direct signature of the molecular phenotype and thus their profiling has concrete clinicalapplications in oncology. Metabolomics and fluxomics, are key technological approaches that mainly revolutionized the fieldenabling researchers to have both a qualitative and mechanistic model of the biochemical activities in cancer. Furthermore, theupgrade from bulk to single-cell analysis technologies provided unprecedented opportunity to investigate cancer biology at cellularresolution allowing an in depth quantitative analysis of complex and heterogenous diseases. More recently, the advent offunctional genomic screening allowed the identification of molecular pathways, cellular processes, biomarkers and noveltherapeutic targets that in concert with other technologies allow patient stratification and identification of new treatmentregimens. This review is intended to be a guide for researchers to cancer metabolism, highlighting current and emergingtechnologies, emphasizing advantages, disadvantages and applications with the potential of leading the development ofinnovative anti-cancer therapies.

Abstract Recently a, monotone generalized directional derixrative has been introduced for Lipschitz functions. This concept has been applied to represent and optimize nonsmooth functions. The second *** result,ed relevant for parallel computing, by allowing to define minimization algorithms with high degree of inherent parallelism. The paper presents first the theoretical background, namely the notions of monotone generalized directional derivative and monotone generalized subdifferential. Then it defines the tools for the procedures, that is a necessary optimality condition and a steel>est descent direction. Therefore the minimization algorithms are outlined. Successively the used architectures and the performed numerical expertence are described, by listing and commenting the *** functions and the obtained results.

Clinical implications of endogenous testosterone density on prostate cancer progression in patients with very favorable low and intermediate risk treated with radical prostatectomy

We tested the association between endogenous testosterone density(ETD;the ratio between endogenous testosterone[ET]and prostate volume)and prostate cancer(PCa)aggressiveness in very favorable low-and intermediate-risk PCa patients who underwent radical prostatectomy(RP).Only patients with prostate-specific antigen(PSA)within 10 ng ml^(-1),clinical stage T1c,and International Society of Urological Pathology(IsUP)grade group 1 or 2 were *** ET levels up to 350 ng dl^(-1)were classified as *** quantitation density factors were evaluated as the ratio between percentage of biopsy-positive cores and prostate volume(biopsy-positive cores density,BPCD)and the ratio between percentage of cancer invasion at final pathology and prostate weight(tumor load density,TLD).disease upgrading was coded as ISUP grade group>2,and progression as recurrence(biochemical and/or local and/or distant).Risk associations were evaluated by multivariable Cox and logistic regression *** 320 patients,151(47.2%)had intermediate-risk ***(median:402.3 ng dl^(-1))resulted abnormal in 111(34.7%)cases(median ETD:9.8 ng dl^(-1)ml^(-1)).Upgrading and progression occurred in 109(34.1%)and 32(10.6%)cases,*** was predicted by ISUP grade group 2(hazard ratio[HR]:2.290;P=0.029)and upgrading(HR:3.098;P=0.003),which was associated with ISUP grade group 2(odds ratio[OR]:1.785;P=0.017)and TLD above the median(OR:2.261;P=0.001).After adjustment for PSAdensity and bodymass index(BMI),ETDabovethemedianwas positivelyassociatedwithBPCD(OR:3.404;P<0.001)and TLD(OR:5.238;P<0.001).Notably,subjects with abnormal ET were more likely to have higher BPCD(OR:5.566;P=0.002),as well as TLD(OR:14.998;P=0.016).Independently by routinely evaluated factors,as ETD increased,BPCD and TLD increased,but increments were higher for abnormal ET *** very favorable cohorts,ETD may further stratify the risk of aggressive PCa.

Liver grafts from hepatitis B surface antigen-positive donors: A review of the literature

The scarcity of available organs and the gap between supply and demand continue to be the main limitations of liver transplantation. To relieve the organ shortage, current transplant strategies have implemented extended criteria, which include the use of liver from patients with signs of past or present hepatitis B virus(HBV) infection. While the use of liver grafts from donors with evidence of past HBV infection is quite limited, some data have been collected regarding the feasibility of transplanting a liver graft from a hepatitis B surface antigen(HBs Ag) positive donor. The aim of the present work was to review the literature regarding liver transplants from HBs Ag-positive donors. A total of 17 studies were identified by a search in Medline. To date, HBs Ag positive grafts have preferentially been allocated to HBs Ag positive recipients. The large majority of these patients continue to be HBs Ag positive despite the use of immunoglobulin, and infection prevention can only be guaranteed by using antiviral prophylaxis. Although serological persistence is evident, no significant HBV-related disease has been observed, except in patients coinfected with delta virus. Consistently less data are available for HBs Ag negative recipients, although they are mostly promising. HBs Agpositive grafts could be an additional organ source for liver transplantation, provided that the risk of reinfection/reactivation is properly prevented.

The aim of this survey paper is to provide the state of the art of the research on control and optimal control of Boolean control networks,under the assumption that all the state variables are accessible and hence available for *** and sufficient conditions for stabilisability to a limit cycle or to an equilibrium point are ***,it is shown that when such conditions are satisfied,stabilisation can always be achieved by means of state *** results are obtained for the safe control problem that is investigated for the first time in this *** and infinite horizon optimal control are subsequently considered,and solution algorithms are provided,based on suitable adaptations of theRiccati difference and algebraic ***,an appropriate definition of the cost function allows to restate and to solve both stabilisation and safe control as infinite horizon optimal control problems.

Performance of Slope Behavior Indicators in Unsaturated Pyroclastic Soils

Landslide risk is increasing in many parts of the world due to growth of population and infrastructures. Therefore, an effort has to be made in developing new and cheap sensors for areas susceptible of landslides to continuously control the slope behaviour, until approaching failure conditions. The paper reported experimental data from smallscale physical models about the performance of Time Domain Reflectometry(TDR) and optical fibres, which act as the indicators of the incoming failure of slopes covered by unsaturated granular soils. Obtained results appear encouraging, since both sensors provide continuous information about the state of the slope, in terms of water content profiles and ongoing deformations, induced by rainwater infiltration, even immediately before the triggering of a fast landslide.

Impact of hepatitis C virus infection on lifestyle

AIM: To evaluate the impact of the diagnosis of hepatitis C infection on lifestyle habits such as smoking, drinking, sports activity and ***: A self-administered, anonymous question-naire was offered to out-patients with HCV infection consecutively attending three clinical centres in ***: Of the 275 respondents, 62.2% (171) were male. Mean age was 51 (range 20-80) years. Overall, after the diagnosis of hepatitis C, 74.5% of drinkers had modified (giving up or reducing) alcohol consumption, 21.3% of smokers had modifi ed their habits and 32.3% of subjects who reported sports activity had either stopped or reduced frequency of activity or chose a less fatiguing sport. Sixty-four percent of the respondents reported having modified their diet, most of them on physician's advice. CONCLUSION: After the diagnosis of hepatitis C many patients correctly modif ied their alcohol consumption and a minority their smoking habits. The reason for reported changes in diet and physical activity needs further investigations.

Colonic diverticular disease (diverticulosis) is one of the most common gastrointestinal disorders in Western countries. This disorder is strictly related to aging and fibre intake, and still bears a discrete amount of morbidity. Numerous etiological co-factors have to date been implicated in the pathogenesis of the disease, yet the supporting evidence is still far from absolute. The present review considers the pathophysiology of colonic diverticular disease, with a special emphasis on factors related to abnormal colonic motility.

In recent years,the contamination of water by arsenic reached alarming levels in many countries of the world,attracting the interest of many researchers engaged in testing methodologies able to remove this harmful *** important aspect that must be taken into consideration is the possibility to find arsenic in different chemical forms which could require different approaches for its *** this aim,a speciation analysis appears to be crucial for better understanding the behavior of arsenic species in aqueous solutions,especially in presence of compounds with marked chelating *** can be identified as good sequestering agents and,at this purpose,this manuscript intends to investigate the interaction of As(Ⅲ)with three phosphonic acids derived from nitrilotriacetic acid(NTA)by replacements of one(N-(Phosphonomethyl)iminodiacetic acid,NTAP),two(N,N-Bis-(phosphonomethyl)glycine,NTA2P)and three(Nitrilotri(methylphosphonic acid),NTA3P)carboxylic groups with the same number of phosphonate *** in-depth potentiometric and calorimetric investigation allowed to determine speciation models featured by simple ML,MLH i and ML(OH)species.A complete thermodynamic characterization of the systems is reported together with the definition of coordination mode by mass spectrometry *** the light of the speciation models,the possibility of using these ligands in arsenic removal techniques was assessed by determining the pL 0.5(the concentration of ligand able to remove the 50%of metal ion present in trace).All ligands show a good sequestering ability,in particular under the conditions of fresh water,following the trend NTA3P>NTA2P>NTAP.
