Background:Diabetes mellitus is a chronic metabolic disease that is a risk factor for epidemic *** hyperglycemic conditions,the enzyme aldose reductase catalyzes the formation of sorbitol in the metabolism of glucose via polyols,leading to the development of diabetic ***,inhibitors of this enzyme are therapeutic targets for the prophylaxis and treatment of these ***:In this study,a generalized linear regression model was developed to analyze flavonoids-obtained from a database-that have been tested as inhibitors of aldose *** this sense,the molecular descriptors implemented in DRAGON and MATLAB software were used to determine the correlation between the chemical structure of the inhibitors and their pharmacological *** model was validated according to the Organisation for Economic Co-operation and development Standards and subsequently used for the virtual screening of the flavonoids identified in Jatropha gossypiifolia ***:The proposed model showed a good fit for its statistical parameters(R2=0.95).In addition,it showed good predictive power(R2 ext=0.94)and robustness(Q2 LOO=0.92).The experimental chemical space wherein the predictions were reliable(domain of application)was also ***,the model was used to identify 10 flavonoids from Jatropha gossypiifolia *** candidates for natural *** with a low probability of oral absorption were identified,among which the elagic acid biflavonoid showed the greatest promise(pIC50 predicted=9.75).Conclusion:The Jatropha gossypiifolia *** harbors flavonoids with high potential as inhibitors of the aldose reductase enzyme,in which the biflavonoid ellagic acid was shown to be the most promising inhibitor of the aldose reductase enzyme,suggesting its possible use in the treatment of the late complications of diabetes mellitus.

Effect of BA Treatments on Morphology and Physiology of Proliferated Shoots of <i>Bambusa vulgaris</i>Schrad. Ex Wendl in Temporary Immersion

Axillary buds, collected from greenhouse-grown plants of Bambusa vulgaris Schrad. ex Wendl (B. vulgaris), were incubated on a static liquid culture medium, Murashige and Skoog (MS) medium with 2% (w/v) sucrose, and supplemented with 12.0 μM 6-benzyladenine (BA). They were transferred to a temporary immersion system (TIS) using liquid MS medium supplemented with 0 (CK-free medium), 6.0, 12.0, 18.0 μM BA. The morphological and anatomical indicators were measured. The BA influenced in vitro multiplication of B. vulgaris. The best results were achieved in the SIT with a concentration of 6.0 μM of BA, which increased the number of shoots (5.1 shoots/explant) in the absence of hyperhydric shoots. Results demonstrated that the water content in the sprouts increased with 12.0 and 18.0 μM BA every four hours. Furthermore, these high levels of BA contributed to a lower accumulation of phenolic compounds and lignin content. The total chlorophyll significantly increased when using 6.0 uM BA, but decreased both parameters with other treatments. These results favor to increase the number of shoots/explants during in vitro multiplication. They will also optimize the in vitro culture conditions, leading to an improvement of in vitro propagation methods for this species.

Stereological Evaluation of Precipitates within Ferrite Grains in Heat-Treated 1.25Cr0.5Mo Steel Previously Exposed to Service

The objective of this work was to study the effect of the heat treatment and further operation aging on the stereological parameters(size,quantity and volume fraction)of precipitates within ferrite grains of a creep-resistant 1.25Cr0.5Mo steel after long-term *** heat treatment was similar to the treatment that is carried out in industrial steam pipelines after welding(post weld heat treatment-PWHT)during installation and/or *** operation aging corresponds to a subsequent long in-service operation after *** determine the stereological parameters,SEM digital images were taken from samples of this material after conditions of in-service-aged(after long-term operation),in-service-aged and heat-treated(simulating repairing)and in-service-aged,heat-treated and in-laboratory aged(simulating subsequent long-term operation after repairing).The results indicate that the changes in the stereological parameters of the precipitates within ferrite grains after PWHT is associated with the coarsening process of the within-ferrite-grain precipitates as well as stabilizing the microstructure,since the material aging after PWHT of an in-serviced aged material would not change the stereological parameters.

Nonlinear Model of Image Noise: An Application on Computed Tomography including Beam Hardening and Image Processing Algorithms

This paper proposes a more inclusive statistical model for predicting image noise in Computed Tomography (CT), associated with scanning factors, considering the effect of beam hardening and image processing filters. It is based on power functions where the levels of the parameters will determine the rate of noise variation with respect to a given scanning factor. It includes the influence of tube potential, tube current, slice thickness, Field of View (FOV), reconstruction methods and post-processing filters. To validate the model, tomographic measurements were made by using a PMMA phantom that simulates paediatric head and adult abdomen, a PET bottle was used to simulate the head of the new-born. The influence of ROI (Region Of Interest) size over nonlinear model parameters was analysed, and high variations of powers of attenuation and FOV were found depending on ROI size. A nonlinear robust regression method was used. The validation was performed graphically by weighted residual analysis. A nonlinear noise model was obtained with an adjusted coefficient of determination for ROI sizes between 10% and 70% of the phantom diameter or FOV. The model confirms the significance of the tube current, slice thickness and beam hardening effect on image. The process of estimation of the parameters of the model by Nonlinear Robust Regression turned out to be optimal.

Aim:It remains unclear what the best therapeutic option for recurrent glioma patients after Stupp treatment ***(BVZ)is commonly administered in progression,but it appears that only some patients *** would be useful to find biomarkers that determine beforehand who these patients ***:The protocol included 31 high-risk progressing glioma patients after Stupp treatment who received BVZ 5-10 mg/kg every 14 days and temozolomide(3-19 cycles,150-200 mg five days each 28-day cycle)during a mean of eight cycles of BVZ or until tumor progression or unacceptable *** analyzed the clinical outcome values of inflammatory indices measured before BVZ ***:Lymphocyte level before BVZ administration was the best independent predictor of overall survival(HR=0.34;95%CI:0.145-0.81;P=0.015).The area under the receiver operating characteristic(ROC)curve was 0.823,with 1.645 being the optimal cut-off value,and 0.80 and 0.85 the sensitivity and specificity values,*** and non-responder survival curves were also significantly different,considering the first and second tertiles as cut-off *** number of BVZ cycles was not related to *** neutrophil platelet levels,platelet-to-lymphocyte ratio(PLR),and neutrophil-to-lymphocyte ratio(NLR)did not have independent predictive *** variables were not correlated with each ***,patients with high NLR and PLR simultaneously(double positive PLR-NLR)showed a worse clinical outcome than the rest(P=0.043).Conclusion:Pretreatment lymphocyte levels and double positive PLR-NLR could be used as non-invasive hematological prognostic markers for recurrent gliomas treated with bevacizumab.A close relationship emerged between inflammation and angiogenesis.

Post-translational modifications of prostaglandin-endoperoxide synthase 2 in colorectal cancer:An update

The biosynthesis of prostanoids is involved in both physiological and pathological processes. The expression of prostaglandin-endoperoxide synthase 2(PTGS2; also known as COX-2) has been traditionally associated to the onset of several pathologies, from inflammation to cardiovascular, gastrointestinal and oncologic events. For this reason, the search of selective PTGS2 inhibitors has been a focus for therapeutic interventions. In addition to the classic non-steroidal anti-inflammatory drugs, selective and specific PTGS2 inhibitors, termed coxibs, have been generated and widely used. PTGS2 activity is less restrictive in terms of substrate specificity than the homeostatic counterpart PTGS1, and it accounts for the elevated prostanoid synthesis that accompanies several pathologies. The main regulation of PTGS2 occurs at the transcription level. In addition to this, the stability of the mRNA is finely regulated through the interaction with several cytoplasmic elements, ranging from specificmicroR NAs to proteins that control mR NA degradation. Moreover, the protein has been recognized to be the substrate for several post-translational modifications that affect both the enzyme activity and the targeting for degradation via proteasomal and non-proteasomal mechanisms. Among these modifications, phosphorylation, glycosylation and covalent modifications by reactive lipidic intermediates and by free radicals associated to the proinflammatory condition appear to be the main changes. Identification of these post-translational modifications is relevant to better understand the role of PTGS2 in several pathologies and to establish a correct analysis of the potential function of this protein in diseases progress. Finally, these modifications can be used as biomarkers to establish correlations with other parameters, including the immunomodulation dependent on molecular pathological epidemiology determinants, which may provide a better frame for potential therapeutic interventions.

The aim of this work is to establish the influence of the relative position of the alloy charge C-Cr-Mn in the structure of the coating of rutile electrodes for hardfacing,on the operational behavior(arc stability).For this,three variants of electrodes with similar chemical composition are elaborated in the metallic core and the coating,differentiating only in the relative position that occupies the alloy charge(C-Cr-Mn)in the structure of the coating:internal,external and *** the development of the research,a completely random design is *** operative characterization of the electrodes is performed in terms of the arc *** conclusion,it can be seen that the position of the alloy load influences the operation behavior(stability of the arc).The variant of internal alloy charge in the coating presents the better arc stability performance.
