Identification of cytokines involved in hepatic differentiation of mBM-MsCs under liver-injury conditions

AIM: To identify the key cytokines involved in hepatic differentiation of mouse bone marrow mesenchymal stem cells (mBM-MsCs) under liver-injury conditions. METHODs: Abdominal injection of CCl4 was adopted to duplicate a mouse acute liver injury model. Global gene expression analysis was performed to evaluate the potential genes involved in hepatic commitment under liver-injury conditions. The cytokines involved in hepatic differentiation of mBM-MsCs was function-ally examined by depletion experiment using specifi c antibodies, followed by rescue experiment and direct inducing assay. The hepatic differentiation was characterized by the expression of hepatic lineage genes and proteins, as well as functional features. REsULTs: Cytokines potentially participating in hepatic fate commitment under liver-injury conditions were initially measured by microarray. Among the up-regulated genes determined, 18 cytokines known to closely relate to liver growth, repair and development, were selected for further identif ication. The f ibroblast growth factor-4 (FGF-4), hepatocyte growth factor (HGF) and oncostatin M (OsM) were fi nally found to be involved in hepatic differentiation of mBM-MsCs under liver-injury conditions. Hepatic differentiation could be dramatically decreased after removing FGF-4, HGF and OsM from the liver-injury conditioned medium, and could be rescued by supplementing these cytokines. The FGF-4, HGF and OsM play different roles in the hepatic differentiation of mBM-MsCs, in which FGF-4 and HGF are essential for the initiation of hepatic differentiation, while OsM is critical for the maturation of hepatocytes. CONCLUsION: FGF-4, HGF and OsM are the key cytokines involved in the liver-injury conditioned medium for the hepatic differentiation of mBM-MsCs.

Determination of Melatonin in the Mian Erkang Capsules by HPLC

The Mian Erkang capsules made by us is a high potency calcium preparation with actions of hypnosis and anti-decrepitude. Melatonin is one of the main effective components which can hypnotize, calm, relieve pain, increase the body's immunity, be used for cancer therapy, and delaydecrepitude. A rapid, sensitive, and accurate HPLC method for the determination of melatonin in Mian Erkang capsules is described in this paper.

Objectives Because pulmonary thromboendarterectomy (PTE) can result in an imme diate reduction in pulmonary artery (PA) pressure, we sought to evaluate the eff ect of PTE on severe tricuspid regurgitation (TR) without tricuspid annuloplasty . Background Few data exist regarding the frequency and magnitude of functional TR improvement after reduction in PA pressure. Methods We identified 27 patients with severe TR, defined by a regurgitant index (RI)>33%, who underwent PTE. The RI, tricusp id annular diameter (TAD), apical displacement of leaflet coaptation, and estima ted PA systolic pressure were determined on pre-and post-PTE echocardiograms. Patients were stratified based on resolution (RI ≤33%) or persistence (RI >33 %) of severe TR. Results Comparing pre-and post-PTE echocardiography results, severe TR resolved in 19 of 27 (70%) patients. This group had a more effective PA systolic pressure reduction after PTE (49 ±20 mm Hg vs. 32 ±16 mm Hg by ec hocardiography, p=0.075, and 37 ±16 mm Hg vs. 16 ±13 mm Hg by catheter measure ment, p=0.004). No difference was observed in TAD, apical displacement of the tr icuspid valve, or other features compared with the group with persistent severe TR. There was a trend toward longer hospital stays in the group with persistent severe TR (19 ±15 days vs. 14 ±9 days; p=0.55). Conclusions After significant PA pressure reduction by PTE, severe functional TR with a dilated annulus may im prove without annuloplasty despite dilated tricuspid annulus diameters.

Objective: In slovenia the opportunistic cervical cancer (CC)screening programme has been performed within the regular gynaecological practice since 1960. The incidence rate 28.8/105 decreased to 16.1/105 in 1982, and increased to 23/105 in 1996. To explain the increased CC incidence the patientsscreening histories, occurrence of symptoms, and details of preventive measures were studied on the basis of medical records and questionnaire. Errors of cytological screening were analysed by reviewing previous smears. Material: The answers obtained from 324 women, treated for CC in the period 1995-2000, were analysed in relation to age, stage and histology. The mean womens age was 46.76 ±13.07 (s.D.) years (range 23-85 years). After therapy, 271 patients were followedup by the end of January 2004, (mean followup time 80.3 ±20 s.D. months, range 29.7-110.3 months). statistical analysis was performed using Chisquare test. Results: In 208 (80%) cases, CC was detected in stage 1, in 43 (17%) in stages 2A to 3B. squamouscell carcinoma (192, 74.1%) was followed by adenocarcinoma (61, 23.5%) and (5, 1.9%) other malignancies. The screening interval ranged from 6 months to 4 years. statistically significant differences existed between clinical and screening variables. CC was diagnosed in higher stages in women who were ignorant of the Pap test. Rescreened smears (n=126) showed 27.8%of false negative results. Conclusions: The reasons for high incidence of CC are poor performance of cytological screening, failures in gynecological examinations and diagnostic procedures, and the patientsnegligence of attending regular screening.

Background The receptor tyrosine kinase-like orphan receptor 2 （ROR2） gene has been recently shown to play important roles in palatal development in animal models and resides in the chromosomal region linked to non syndromic cleft lip with or without cleft palate in *** aim of this study was to investigate the possible association between ROR2 gene and non-syndromic oral *** Here we tested 38 eligible single-nucleotide polymorphisms （sNPs） in ROR2 gene in 297 non-syndromic cleft lip with or without cleft palate and in 82 non-syndromic cleft palate case parent trios recruited from Asia and *** Based Association Test was used to test for deviation from Mendelian *** software was used to test potential parent of origin *** maternally mediated in utero effects were assessed using the TRlad Multi-Marker approach under an assumption of mating symmetry in the *** significant evidence of linkage and association was shown for 3 sNPs （rs7858435,rs10820914 and rs3905385） among 57 Asian non-syndromic cleft palate trios in Family Based Association Tests.P values for these 3 sNPs equaled to 0.000068,0.000115 and 0.000464 respectively which were all less than the significance level （0.05/38=0.0013） adjusted by strict Bonferroni *** odds ratios for the risk allele were 3.42 （1.80-6.50）,3.45 （1.75-6.67） and 2.94 （1.56-5.56）,*** evidence of linkage and association was not shown for study groups other than non-syndromic cleft *** evidence for parent-of-origin nor maternal genotypic effect was shown for any of the ROR2 markers in our analysis for all study *** Our results provided evidence of linkage and association between the ROR2 gene and a gene controlling risk to non-syndromic cleft palate.

Association of NOD1 (CARD4) insertion/deletion polymorphism with susceptibility to IBD: A meta-analysis

AIM: To find evidences about whether NOD1/CARD4 insertion/deletion polymorphism is associated with inflammatory bowel disease by meta-analysis. METHODs: We surveyed the studies on the association of NOD1/CARD4 insertion/deletion polymorphism with inflammatory bowel disease in Pubmed. Meta-analysis was performed for genotypes GG/T vs T/T, GG/GG vs T/T, GG/T + GG/GG vs T/T, GG/GG vs T/T + GG/T, and GG allele vs T allele in a fixed/random effect model. REsULTs: We identified 8 studies (6439 cases and 4798 controls) in Caucasian populations using Pubmed search. We found no association between NOD1/CARD4 insertion/deletion polymorphism and inflammatory bowel disease, Crohn's disease, and ulcerative colitis. stratification of cases by age showed that NOD1/CARD4 insertion/deletion polymorphism was associated with inflammatory bowel disease in younger age group at onset (s) (GG vs T: OR = 0.68, 95% CI: 0.50-0.93, P = 0.02; GG/T + GG/GG vs T/T: OR = 0.71, 95% CI: 0.59-0.85, P = 0.0003). CONCLUsION: This meta-analysis demonstrates an association between NOD1/CARD4 insertion/deletion polymorphism and inflammatory bowel disease in the younger age group at onset (s) in Caucasian populations.
