Unveiling the predictive power of bacterial response-related genes signature in hepatocellular carcinoma: with bioinformatics analyses and experimental approaches

Background: Despite progress in therapeutic strategies, treatment failure in hepatocellular carcinoma (HCC) remains a major challenge, resulting in low survival rates. The presence of bacteria and the host ' s immune response to bacteria can in fl uence the pathogenesis and progression of HCC. We developed a risk model based on bacterial response-related genes (BRGs) using gene sets from molecular signature databases to identify new markers for predicting HCC outcomes and categorizing patients into different risk groups. Methods: The data from the Cancer Genome Atlas (TCGA) portal was retrieved, and differentially expressed BRGs were identi fi ed. Uni- and multivariate Cox regression and least absolute shrinkage and selection operator (LASSO) LASSO analyses were executed to develop the prognostic risk model. Key contributor to the prognostic model was identi fi ed, and the results were tested by using experimental assays in HCC cell lines. Results: Multivariate analysis demonstrated an independent prognostic factor of 12-BRG signature in HCC patients. The low-risk group had better overall survival with signi fi cantly lower tumor mutation burden (TMB). The risk scores were negatively correlated with the presence of tumor-in fi ltrating immune cells. In an effort to fi nd the key contributor of the 12-BRG signature, we found polo like kinase1 (PLK1) had the best accuracy with 1-, 3-, and 5-year AUC of 0.72, 0.66, and 0.65, respectively. Both PLK1 inhibitor Volasertib and the knockdown of the PLK1 gene resulted in diminished viability in HCC cell lines. The combination of PLK1 inhibition with low-dose chemotherapy exhibited an ampli fi ed effect of the treatment. Conclusion: To date, there have been no reports of BRG biomarkers in HCC, and this study represents for the fi rst time that a 12-BRG signature has the potential to predict the survival of HCC.

Biomechanics of knee joint during forward-walking and drop-landing after anterior cruciate ligament reconstruction: finite element and gait analysis

The anterior cruciate ligament plays a crucial role in maintaining stability within the knee joint, particularly for athletes who frequently experience its rupture. This study presents a novel approach using personalized three-dimensional (3D) parametric finite element modeling of the knee joint to simulate the treatment following anterior cruciate ligament reconstruction (ACLR) in both forward walking (FW) and drop landing (DL) tasks. The study encompasses two distinct cohorts: five healthy athletes and five ACLR patients. Biomechanical motion analysis was conducted on both cohorts, with the ACLR patient group evaluated at 6 and 9 months post-surgery. A comprehensive 3D parametric model of the knee joint was meticulously crafted. The findings reveal a notable reduction in stress on crucial knee structures such as the autograft, meniscus, and cartilages over time for both FW and DL tasks following ACLR, with a reduction in tissue tension of approximately 9.5% and 37% for FW and DL, respectively. This personalized model not only facilitates the investigation of knee joint tissue biomechanics post-ACLR but also aids in estimating the return-to-sports timeline for patients. By accommodating individual tissue geometries and incorporating patient-specific kinetic data, this model enhances our comprehension of post-ACLR biomechanics across various functional tasks, thereby optimizing rehabilitation strategies.

Ginger (Zingiber officinale) essential oil (GEO) was loaded into nanoliposome (NLP) for smart release in simulated in-vitro digestion. Optimal conditions for the fabrication of NLp-GEO were determined according to physical stability, size distribution and particle size. Morphological analyses (atomic force microscopy (AFM) and scan-ning electron microscopy (SEM)) confirmed the results of the dynamic light scattering (DLS). Structural evalu-ation (differential Scanning Calorimetry (DSC) and Fourier-transform infrared spectroscopy (FTIR)) demonstrated the interaction between GEO and NLP. Additionally, the approximate size and loading capacity of optimized NLp-GEO (100 nm) was 100 nm and 66.24%. NLp-GEO release under oral, gastric, and intestinal conditions showed that over 90% of the nutraceutical was released after exposure to model gastrointestinal conditions. Finally, NLp-GEO structure showed an improved antioxidant, UV stability and reduced cytotoxicity on cell lines (human colon cancer (HT-29) and normal human umbilical vein endothelial cells (HUVEC). As an innovative smart sustained-release system in simulated in-vitro digestion, NLP-GEO would be a potential practical option in the food and pharmaceutical industries.

Enhancing Sound Absorption in Micro-Perforated Panel and Porous Material Composite in Low Frequencies: A Numerical Study Using FEM

Mitigating low-frequency noise poses a significant challenge for acoustic engineers, due to their long wavelength, with conventional porous sound absorbers showing limitations in attenuating such noise. An effective strategy involves combining porous materials with micro-perforated plates (MPP) to address this issue. Given the significant impact of structural variables like panel thickness, hole diameter, and air gap on the acoustic characteristics of MPP, achieving the optimal condition demands numerous sample iterations. The impedance tube's considerable expense for sound absorption measurement and the substantial cost involved in fabricating each sample using a 3D printer underscore the advantage of utilizing simulation methods to attain the optimal state. This study focuses on optimizing low-frequency enhancement by investigating key parameters. Using the Finite Element Numerical Method (FEM) in COMSOL software, a composite panel was constructed comprising date palm fiber layers, an intervening air layer, and MPP. The study explored the arrangement of these layers and the impact of parameters like hole diameter, plate thickness, and perforation ratio on acoustic behavior. The selected optimal parameter at each stage was consistently maintained for subsequent steps. Results revealed that layer arrangement significantly influenced acoustic characteristics. Placing the MPP layer before the porous material yielded superior low-frequency performance. Optimizing low-frequency behavior involved reducing hole diameter and perforation ratio while increasing plate thickness. Elevating the porous material's thickness relative to the air layer behind the MPP enhanced absorption peak and resonance frequency. In conclusion, halving the porous layer's thickness while incorporating an air layer and single MPP proved more effective than using a thick porous material. Analysis of the Relationships between Noise Exposure and Stress/Arousal Mood at Different Levels of Workload

Noise is one of the environmental factors with mental and physical effects. The workload is also the multiple mental and physical demands of the task. Therefore, his study investigated the relationship between noise exposure and mood states at different levels of workload. The study recruited 50 workers from the manufacturing sector (blue-collar workers) as the exposed group and 50 workers from the office sector (white-collar workers) as the control group. Their occupational noise exposure was measured by dosimetry. The Stress -Arousal Checklist (SACL) and the NASA Task Load Index (NASA-TLX) were used to measure mood and workload, respectively. The equivalent noise exposure level of the exposed group at high and very high workload levels was 85 and 87 dBA, respectively. The mean mood score of the exposed group was 76 at very high workload. The correlation coefficient between noise exposure level and mood state based on workload levels ranged from 0.3 at medium workload to 0.57 at very high workload. Noise exposure at high workload levels can increase its adverse effects, so controlling and optimizing the multiple demands of the task in the workplace can be used as a privative measure to reduce the adverse effects of noise.

Gelatin-maltodextrin microcapsules as carriers of vitamin D3 improve textural properties of synbiotic yogurt and extend its probiotics survival

Food fortification is one of the most essential processes to compensate for the deficiency of minerals and vitamins in customers' body. Hence, this study aimed to fortify low-fat stirred yogurt via encapsulated vitamin D3 (VD3) in maltodextrin and gelatin matrix, characterize obtained microcapsules (encapsulation efficiency, zeta potential, particle size, differential scanning calorimetry (DSC), scanning electron microscopy (SEM), Fourier trans-formation infrared (FTIR) spectroscopy, release features, and explore the properties of resultant yogurt including its syneresis, prebiotic activity, sensory properties, and rheology. Encapsulation efficiency and loading efficiency of microcapsules were satisfactorily high (Up to 97% and 25%, respectively). Microscopy images of micro -encapsulated VD3 showed a smooth surface as well as spherical and slight cracks in the microcapsules. The microcapsules had strong negative zeta potentials (-27.29 mV), implying a considerable repulsive force between microcapsules and their good stability. The FTIR result showed strong interactions between VD3 and wall ma-terials, confirming the presence of VD3 in the microcapsules. VD3 had an almost slow release in the gastric, while it was quickly released from microcapsules under simulated intestinal conditions. Fortification of yogurts with encapsulated VD3 led to an increase in textural parameters where syneresis value decreased and viscosity increased. The number of probiotics in fortified yogurts was also considerably higher than in the control sample. The sensory analyses revealed no major differences between the fortified and control samples, hinting that protein-polysaccharide microcapsules were suitable wall materials for VD3 microencapsulation, and these mi-crocapsules could potentially have numerous applications in the food industry.

Interferon Regulatory Factors (IRFs), a family of transcription factors, profoundly influence the immune system, impacting both physiological and pathological processes. This review explores the diverse functions of nine mammalian IRF members, each featuring conserved domains essential for interactions with other transcription factors and cofactors. These interactions allow IRFs to modulate a broad spectrum of physiological processes, encompassing host defense, immune response, and cell development. Conversely, their pivotal role in immune regulation implicates them in the pathophysiology of various diseases, such as infectious diseases, autoimmune disorders, metabolic diseases, and cancers. In this context, IRFs display a dichotomous nature, functioning as both tumor suppressors and promoters, contingent upon the specific disease milieu. Post-translational modifications of IRFs, including phosphorylation and ubiquitination, play a crucial role in modulating their function, stability, and activation. As prospective biomarkers and therapeutic targets, IRFs present promising opportunities for disease intervention. Further research is needed to elucidate the precise mechanisms governing IRF regulation, potentially pioneering innovative therapeutic strategies, particularly in cancer treatment, where the equilibrium of IRF activities is of paramount importance.

The levels of thyroxine (T3, T4) and thyroid stimulating hormone (TSH) in rat serum were measured by radioimmunoassay, and also ultrastructure of the thyroid gland and adenohypophyseal cell were examined by transmission electronic microscopy (TEM) after low concentration (0. 05 mg/kg) of samarium chloride (SmCl3) administration by intraperitoneal, oral and tail intravenous routes. The results showed that the level of T4 in rat serum increased and the level of TSH unchanged by the intraperitoneal administration, and the levels of T3, T4 and TSH did not increase by the oral and tail intravenous administration. The evidences mentioned above were in accord with the morphological changes of thyroid follicular epithelial cells andadenohypophysis thyrotroph observed under electronic microscope in a sense.

Design of DLC/Titanium Alloy Bio-Functionally Gradient Coat

The material design is used to direct the magnetron cosputtering process. At first, according to the particularity of functionally gradient coat (FGC) the thermal elastic stress analysis for FGC was carried out based on the plane stress hypothesis. It is obtained that the peak value of plane thermal stress within FGC is only determined by the physical properties of materials of FGC and substrate, the composition distribution coefficient only influences the distribution and trend of plane thermal stress. And the plane thermal stress criterion for design of FGC was presented. Then the plane thermal stress of diamond like carbon/titanium alloy FGC was calculated.

Effect of Low Dose Samarium Chloride on Serum Hormone Level in Rat

Low dose of samarium chloride was injected into rat(0.05 mg/kg) every other day through three different routes i.e. peritioneal injection, gastric slysis and caudal vein injection. One month later. Serum GH, THT3, THT4 and insulin were determined by using radio-immunity assay. The results demonstrated that both serum GH and insulin raised remarkably in experimental groups. THT4 raised only in peritioneal injection group. Besides these, the four different serum hormones showed no obvious change when the dose of samarium chloride decreased to 0.025 mg/kg with caudal vein injection.
