Background:Platinum chemotherapy(CT)remains the backbone of systemic therapy for patients with smallcell lung cancer(SCLC).The nucleotide excision repair(NER)pathway plays a central role in the repair of the DNA damage exerted by platinum *** in this repair mechanism may affect patients’*** and Methods:We conducted a retrospective analysis of data from 38 patients with extensive disease(ED)-SCLC who underwent platinum-CT at the Clinical Oncology unit,Careggi University Hospital,Florence(Italy),from 2015 to *** expression analysis and single nucleotide polymorphism(SNP)characterization of three NER pathway genes—namely ERCC1,ERCC2,and ERCC5—were performed on patient tumor ***:Overall,elevated expression of ERCC genes was observed in SCLC patients compared to healthy *** with low ERCC1 and ERCC5 expression levels exhibited a better median progression-free survival(mPFS=7.1 vs.4.9 months,p=0.39 for ERCC1 and mPFS=6.9 vs.4.8 months,p=0.093 for ERCC5)and overall survival(mOS=8.7 vs.6.0 months,p=0.4 for ERCC1 and mOS=7.2 vs.6.2 months,p=0.13 for ERCC5).Genotyping analysis of five SNPs of ERCC genes showed a longer survival in patients harboring the wild-type genotype or the heterozygous variant of the ERCC1 rs11615 SNP(p=0.24 for PFS and p=0.14 for OS)and of the rs13181 and rs1799793 ERCC2 SNPs(p=0.43 and p=0.26 for PFS and p=0.21 and p=0.16 for OS,respectively)compared to patients with homozygous mutant ***:The comprehensive analysis of ERCC gene expression and SNP variants appears to identify patients who derive greater survival benefits from platinum-CT.

It is difficult to track multiple maneuvering targets of which the number is unknown and timevarying,especially when there is range *** random finite sets(RFS)based probability hypothesis density filter(PHDF)is an effective solution to the problem of multiple targets ***,when tracking multiple targets via the range ambiguous radar,the problem of range ambiguity has to be *** this paper,a multiple model PHDF and data association(MMPHDF-DA)based method is proposed to address multiple maneuvering targets tracking with range ambiguous radar in ***,by introducing the turn rate of target and the discrete pulse interval number(PIN)as components of target state vector,and modeling the incremental variable of the PIN as a three-state Markov chain,the problem of multiple maneuvering targets tracking with range ambiguity is converted into a hybrid state filtering ***,by implementing a novel"track-estimate"oriented association with the filtering results of the hybrid filter,target tracks are provided at each time *** results demonstrate that the MMPHDF-DA can estimate target state as well as the PIN simultaneously,and succeeds in multiple maneuvering target tracking with range ambiguity in *** results also demonstrate that the MMPHDF-DA can overcome the limitation of the Chinese Remainder Theorem for range ambiguity resolving.

The middle pulse repetition frequency（MPRF）and high pulse repetition frequency（HPRF）modes are widely adopted in airborne pulse Doppler（PD）radar systems,which results in the problem that the range measurement of targets is *** existing data processing based range ambiguity resolving methods work well on the condition that the signal-to-noise ratio（SNR）is high *** this paper,a multiple model particle flter（MMPF）based track-beforedetect（TBD）method is proposed to address the problem of target detection and tracking with range ambiguous radar in low-SNR *** introducing a discrete variable that denotes whether a target is present or not and the discrete pulse interval number（PIN）as components of the target state vector,and modeling the incremental variable of the PIN as a three-state Markov chain,the proposed algorithm converts the problem of range ambiguity resolving into a hybrid state fltering *** last,the hybrid fltering problem is implemented by a MMPF-based TBD method in the Bayesian *** results demonstrate that the proposed Bayesian approach can estimate target state as well as the PIN simultaneously,and succeeds in detecting and tracking weak targets with the range ambiguous *** results also show that the performance of the proposed method is superior to that of the multiple hypothesis（MH）method in low-SNR environment.

The online diagnosis for aircraft system has always been a difficult problem. This is due to time evolution of system change, uncertainty of sensor measurements, and real-time requirement of diagnostic inference. To address this problem, two dynamic Bayesian network(DBN) approaches are proposed. One approach prunes the DBN of system, and then uses particle filter(PF) for this pruned DBN(PDBN) to perform online diagnosis. The problem is that estimates from a PF tend to have high variance for small sample sets. Using large sample sets is computationally expensive. The other approach compiles the PDBN into a dynamic arithmetic circuit(DAC) using an offline procedure that is applied only once, and then uses this circuit to provide online diagnosis recursively. This approach leads to the most computational consumption in the offline procedure. The experimental results show that the DAC, compared with the PF for PDBN, not only provides more reliable online diagnosis, but also offers much faster inference.

Irisin is a myokine that is generated by cleavage of the membrane protein fibronectin type Ⅲ domain-containing protein 5(FNDC5) in response to physical exercise. Studies reveal that irisin/FNDC5 has neuroprotective functions against Alzheimer's disease, the most common form of dementia in the elderly, by improving cognitive function and reducing amyloid-β and tau pathologies as well as neuroinflammation in cell culture or animal models of Alzheimer's disease. Although current and ongoing studies on irisin/FNDC5 show promising results, further mechanistic studies are required to clarify its potential as a meaningful therapeutic target for alleviating Alzheimer's disease. We recently found that irisin treatment reduces amyloid-β pathology by increasing the activity/levels of amyloid-β-degrading enzyme neprilysin secreted from astrocytes. Herein, we present an overview of irisin/FNDC5's protective roles and mechanisms against Alzheimer's disease.

Our previous studies have reported that activation of the NLRP3(NOD-,LRR-and pyrin domain-containing protein 3)-inflammasome complex in ethanol-treated astrocytes and chronic alcohol-fed mice could be associated with neuroinflammation and brain *** stem cell-derived extracellular vesicles(MSC-EVs)have been shown to restore the neuroinflammatory response,along with myelin and synaptic structural alterations in the prefrontal cortex,and alleviate cognitive and memory dysfunctions induced by binge-like ethanol treatment in adolescent *** the therapeutic role of the molecules contained in mesenchymal stem cell-derived extracellular vesicles,the present study analyzed whether the administration of mesenchymal stem cell-derived extracellular vesicles isolated from adipose tissue,which inhibited the activation of the NLRP3 inflammasome,was capable of reducing hippocampal neuroinflammation in adolescent mice treated with binge *** demonstrated that the administration of mesenchymal stem cell-derived extracellular vesicles ameliorated the activation of the hippocampal NLRP3 inflammasome complex and other NLRs inflammasomes(e.g.,pyrin domain-containing 1,caspase recruitment domain-containing 4,and absent in melanoma 2,as well as the alterations in inflammatory genes(interleukin-1β,interleukin-18,inducible nitric oxide synthase,nuclear factor-kappa B,monocyte chemoattractant protein-1,and C–X3–C motif chemokine ligand 1)and miRNAs(miR-21a-5p,miR-146a-5p,and miR-141-5p)induced by binge-like ethanol treatment in adolescent *** analysis further revealed the involvement of miR-21a-5p and miR-146a-5p with inflammatory target genes and NOD-like receptor signaling *** together,these findings provide novel evidence of the therapeutic potential of MSC-derived EVs to ameliorate the hippocampal neuroinflammatory response associated with NLRP3 inflammasome activation induced by binge drinking in adolescence.
