The ameliorative effects of ginger against heat-induced oxidative stress and mitochondrial dysfunction in Caenorhabditis elegans

Ginger， rich in gingerols and shogaols， exhibits multiple biological properties. However， the mechanisms underlying its thermotolerance remain unclear. The study employed network pharmacology and experimental validation in Caenorhabditis elegans to investigate how gingerol-related compounds within ginger extract (GE) mitigated damage caused by heat stress (HS). A total of 18 types of gingerol analogues were identified in GE， among which 6-， 8-， and 10-gingerol， as well as 6-， 8-， and 10-shogaol were quantified. Collectively， these six compounds accounted for 54.4% of the total composition. Supplementation with 15 μg/mL GE significantly extended heat-stress lifespan by 20.30%， while the combination of the six major gingerols and shogaols at the same concentration prolonged lifespan by 18.93%. Additionally， pretreatment with GE and the combination alleviated HS-induced oxidative damage by eliminating reactive oxygen species (ROS) and upregulating antioxidant enzymes. Network pharmacology analysis suggested that the MAPK pathway may play a crucial role in thermotolerance. Experimental findings confirmed that ginger attenuated oxidative damage through the activation of SKN-1/Nrf2 and DAF-16/FOXO via the MAPK pathway. Moreover， GE stabilized mitochondrial membrane potential and restored ATP levels， thus preserving mitochondrial function during heat exposure. Further investigations using molecular docking and molecular dynamics simulations revealed that shogaols， with more stable binding affinities for KEAP1 protein， exhibited more potent effects than gingerols in prolonging lifespan and reducing ROS levels under HS conditions. In short， gingerol analogues from ginger conferred thermal resistance to nematodes by mitigating oxidative damage and mitochondrial dysfunction.

The possible impacts on nitrogen-cycle in a p-nitrophenol （PNP） polluted soil and the effectiveness of wastewater sludge amendments in restoring nitrification potential and urease activity were evaluated by an incubation study. The results indicated that PNP at 250 mg/kg soil inhibited urease activity, nitrification potential, arginine ammonification rate and heterotrophic bacteria counts to some extents. After exposure to PNP, the nitrification potential of the tested soil was dramatically reduced to zero over a period of 30 days. Based on the findings, nitrification potential was postulated as a simple biochemical indicator for PNP pollution in soils. Nitrogen-cycling processes in soils responded positively to the applications of wastewater sludges. A sludge application rate of 200 tons/ha was sufficient for successful biostimulation of these nitrogen processes. The microbial activities in sludge-amended, heavy PNP-polluted soils seemed to recover after 30–45 days, indicating the effectiveness of sludge as a useful soil amendment.

The authors recently defined a new graph invariant denoted by Ω(G) only in terms of a given degree sequence which is also related to the Euler characteristic. It has many important combinatorial applications in graph theory and gives direct information compared to the better known Euler characteristic on the realizability, connectedness, cyclicness, components, chords, loops etc. Many similar classification problems can be solved by means of Ω. All graphs G so that Ω(G) ≤-4 are shown to be disconnected, and if Ω(G) ≥-2, then the graph is potentially connected. It is also shown that if the realization is a connected graph and Ω(G) =-2, then certainly the graph should be a ***, it is shown that if the realization is a connected graph G and Ω(G) ≥ 0, then certainly the graph should be cyclic. Also, when Ω(G) ≤-4, the components of the disconnected graph could not all be cyclic and if all the components of G are cyclic, then Ω(G) ≥ 0. In this paper, we study an extremal problem regarding graphs. We find the maximum number of loops for three possible classes of *** also state a result giving the maximum number of components amongst all possible realizations of a given degree sequence.

We applied the multiple exp-function scheme to the（2＋1）-dimensional Sawada-Kotera（SK） equation and（3＋1）-dimensional nonlinear evolution equation and analytic particular solutions have been deduced. The analytic particular solutions contain one-soliton, two-soliton, and three-soliton type solutions. With the assistance of Maple, we demonstrated the efficiency and advantages of the procedure that generalizes Hirota＇s perturbation scheme. The obtained solutions can be used as a benchmark for numerical solutions and describe the physical phenomena behind the model.

Since 194os, Mount uludag （Bithynian Olympus） has been considered famous with winter sports center in Turkey. In addition, it is one of the rare places because of its plant diversity and is also one of the important plant areas of Turkey. The ski run constructions are rapidly increased in the past decades in the alpine and subalpine belts of the mountain. Rate of nitrogen mineralization and some properties of soft were investigated in the soils of the three ski runs and undisturbed neighborhood forest sites. These ski runs are at the upper part of the forest belt in the mountain. Abies bornmuelleriana forest community is the postclimax and very sensitive to destruetion. The rates of the nitrogen mineralization in the soils were determined in controlled conditions （60% water-holding capacity, 20℃）. Nitrification and nitrogen mineralization were in the ski run inside where the natural recovery started, increased compared with the outside of the ski runs. Waterholding capacity, organic carbon, and total nitrogen were decreased in the massive damaged ski run, and due to these changes, the rates of mineralization and nitrification were also decreased.

The effect of size on the biaxial flexural strength(BFS)of Portland cement mortar was investigated by using the recently proposed triangular plate method(TPM).An experimental program was conceived to study the size effect by keeping a constant water-cement ratio of 0.485,cement-sand ratio of 1:2.75,and using unreinforced triangular mortar plates of five different thicknesses and seven different side *** BFS of the produced specimens was tested,and variations of BFS depending on specimen thickness and side length were *** results indicated that increases in triangular plate specimen side length and specimen thickness led to a decrease in the BFS of Portland cement *** effect of specimen length increase on BFS was more significant than on the effect of the specimen *** variations in specimens’thickness indicated a deterministic Type I size effect,while the variations in specimens’length showed an energetic-statistical Type I size effect.

Objective To determine the in vitro possible clastogenic and cytotoxic activities of Ulva rigida crude extracts （URE）, and identify their antigenotoxic and protective effects on chemotherapeutic agent mitomycine-C （MMC）. Methods Anti-clastogenic and anti-genotoxic activities of Ulva rigida crude extracts （URE） were studied using chromosome aberration （CA）, sister chromatid exchange （SCE）, and micronuclei （MN） tests in human lymphocytes cultured in vitro. Results The chromosome aberration, sister chromatid exchange or micronuclei tests showed that URE at concentrations of 10, 20, and 40 lag/mL had no clastogenic activity in human lymphocyte cell culture. Three doses of URE significantly decreased the number of chromosomal aberrations and the frequencies of SCE and MN when compared with the culture treated with MMC （P〈0.0001）. Conclusion Although URE itself is not a clastogenic or cytotoxic substance, it possesses strong antigenotoxic, anti-clastogenic, and protective effects on MMC in vitro.

In this paper,the effect of usage of the permeability reducing admixture(PRA)having different action mechanisms on hardened state properties of cementitious systems containing mineral additives is *** this aim,three commercial PRAs were used during *** effective parameters in the first and third PRAs were air-entraining and high-rate air-entraining,*** second one contained the insoluble calcium carbonate residue and had a small amount of the air-entraining *** mixes with binary and ternary cementitious systems were prepared by partially replacing cement with fly ash and *** hardened state properties of mortar mixtures such as compressive strength,ultrasonic pulse velocity,water absorption,drying shrinkage and freeze-thaw resistance were *** ternary cement-based mixture having both fly ash and metakaolin was selected as the most successful mineral-additive bearing mix in regard to hardened state *** this sense,PRA-B,with both insoluble residues and a small amount of air-entraining properties,showed the best performance among the mixtures containing *** combined use of mineral additive and PRA had a more positive effect on the properties of the mixes.
