Landscape of four different stages of human gastric cancer revealed by single-cell sequencing

BACKGROUND Gastric cancer(GC)poses a substantial risk to human health due to its high prevalence and mortality ***,current therapeutic strategies remain ***-cell RNA sequencing(scRNA-seq)offers the potential to provide comprehensive insights into GC *** To explore the distribution and dynamic changes of cell populations in the GC tumor microenvironment using scRNA-seq *** Cancerous tissues and paracancerous tissues were obtained from patients diagnosed with GC at various stages(I,II,III,and IV).Single-cell suspensions were prepared and analyzed using scRNA-seq to examine transcriptome profiles and cell-cell ***,quantitative real-time polymerase chain reaction(qRT-PCR)and flow cytometry were applied for measuring the expression of cluster of differentiation(CD)2,CD3D,CD3E,cytokeratin 19,cytokeratin 8,and epithelial cell adhesion *** Transcriptome data from 73645 single cells across eight tissues of four patients were categorized into 25 distinct cell clusters,representing 10 different cell *** were observed in these cell type *** adjacent epithelial cells in stages II and III exhibited a degenerative ***,the quantity of CD4 T cells and CD8 T cells were evidently elevated in cancerous *** analysis displayed a remarkable increase in interaction between B cells and other mast cells in stages II,III,and IV of *** findings were further validated through qRT-PCR and flow cytometry,demonstrating elevated T cells and declined epithelial cells within the cancerous *** This study provides a comprehensive analysis of cell dynamics across GC stages,highlighting key interactions within the tumor *** findings offer valuable insights for developing novel therapeutic strategies.

Tianxiangdan(TXD) alleviates myocardial ischemia reperfusion-induced ferroptosis through the activation of estrogen receptor alpha(ERα)

Tianxiangdan（TXD）, a traditional Chinese herbal remedy, demonstrates efficacy in mitigating myocardial ischemia-reperfusion（I/R）-induced damage. This study employed network pharmacology to evaluate the therapeutic targets and mechanisms of TXD in treating I/R. Highperformance liquid chromatography-mass spectrometry（HPLC-MS） identified 86 compounds in TXD. Network pharmacological analysis predicted potential target genes and their modes of action. Cardiac function, ischaemic ST changes, lactate dehydrogenase（LDH）,malondialdehyde（MDA）, superoxide dismutase（SOD） activity, myocardial fiber, and infarct size were assessed using in vivo and in vitro I/R injury models. Estrogen receptor alpha（ERα）protein expression and estradiol（E2） levels were measured to confirm TXD's impact on estrogen levels and ERα expression. To examine if TXD reduces I/R injury through ERα, an AZD group(300 nmol·L-1AZD9496 and 15% TXD serum) was compared to a TXD group（15% TXD serum）. The study hypothesized that TXD upregulates the ERα-mediated iron metamorphosis pathway. I/R injury-induced ferroptosis was identified using a Fer-1 group(1.0 μmol·L-1Fer-1 and 15% TXD serum) to elucidate the potential association between ferroptosis and ERαproteins. A DCFH-DA probe detected reactive oxygen species（ROS） and Fe2+, while Western blotting assessed target protein expression. Both in vitro and in vivo experiments demonstrated that TXD attenuated I/R injury by reducing elevated ST-segment levels, improving cardiac injury biomarkers（LDH, MDA, and SOD）, alleviating pathological features, and preventing I/R-induced loss of cell viability in vitro. The effects and mechanisms of TXD on I/R injury-associated ferroptosis were investigated using I/R-induced H9c2 cells. The TXD group showed significantly decreased ROS and Fe2+levels, while the AZ group（treated with AZD9496） exhibited increased levels. Cohort study on the treatment of BRAF V600E mutant metastatic colorectal cancer with integrated Chinese and western medicine

BACKGROUND Patients with BRAF V600E mutant metastatic colorectal cancer(mCRC)have a low incidence rate,poor biological activity,suboptimal response to conventional treatments,and a poor *** the previous cohort study on mCRC conducted by our team,it was observed that integrated Chinese and Western medicine treatment could significantly prolong the overall survival(OS)of patients with colorectal ***,we further explored the survival benefits in the population with BRAF V600E mutant *** To evaluate the efficacy of integrated Chinese and Western medicine in the treatment of BRAF V600E mutant metastatic colorectal *** A cohort study was conducted on patients with BRAF V600E mutant metastatic colorectal cancer admitted to Xiyuan Hospital of China Academy of Chinese medical Sciences and Traditional Chinese medicine Hospital of xinjiang Uygur Autonomous Region from January 2016 to December *** patients were divided into two *** A total of 34 cases were included,with 23 in Chinese-Western medicine cohort(cohort A)and 11 in Western medicine cohort(cohort B).The median overall survival was 19.9 months in cohort A and 14.2 months in cohort B,with a statistically significant difference(P=0.038,hazard ratio=0.46).The 1-3-year survival rates were 95.65%(22/23),39.13%(9/23),and 26.09%(6/23)in cohort A,and 63.64%(7/11),18.18%(2/11),and 9.09%(1/11)in cohort B,*** analysis showed statistically significant differences in median OS between the two cohorts in the right colon,liver metastasis,chemotherapy,and first-line treatment subgroups(Pmedicine can prolong the survival and reduce the risk of death in patients with BRAF V600E mutant metastatic colorectal cancer,with more pronounced benefits observed in patients with right colon involvement,liver metastasis,combined chemotherapy,and first-line treatment.

Objective:To investigate the distribution of abnormal hilit syndromes in traditional uighur medicine(TUM) among human immunodeficiency virus infection and acquired immune deficiency syndrome(HIV/AIDS)patients,and to find out the clinical characteristics of abnormal savda syndrome type HIV/AIDS ***:Between June and July in 2012,307 eligible HIV/AIDS patients from in-patient department and out-patient clinics of xinjiang uighur Autonomous Region the Sixth People’s Hospital in Urumqi were *** syndrome differentiation was performed by a senior TUM *** participant completed a Sign and Symptom CheckList for Persons Living with HIV/AIDS(SSC-HIV) *** was evaluated by using Hamilton Rating Scale for Depression *** specimen was collected from each participant to test the levels of blood ***:Of 307 HIV/AIDS patients,189(61.6%) were abnormal savda syndrome type,118(38.4%) were non-abnormal-savda syndrome *** CD4 counts of abnormal savda syndrome type patients was(227.61 ±192.93) cells/μL,and the prevalence of anemia,thrombocytopenia,and elevated cystatin C were49.7%,28.6%,and 44.7%,which were significantly higher than those in the non-abnormal-savda syndrome type patients(26.3%,16.0%and 25.0%,P<0.05).In addition,depression(79.9%) and HIV/AIDS-related symptoms such as fatigue(42.3%),back aches(40.7%),lack of appetite(33.9%),night sweats(31.7%) were more common among abnormal savda syndrome patients(P<0.05).Conclusion:Abnormal savda syndrome is the dominant syndrome among HIV/AIDS patients,and they present a more sever clinical manifestation.

Fast electron-hole recombination issues during titanium dioxide(TiO_(2))photocatalysis limit its application in preventing bacterial infection during bone defect *** this study,TiO_(2)@reduced graphene oxide(rGO)composites were synthesized using a hydrothermal method in which rGO,which possesses very high electrical conductivity,promotes the separation of photoelectron-hole pairs of TiO_(2),thus improving the efficiency of photocatalytic production of reactive oxygen species(ROS).Subsequently,TiO_(2)@rGO composites were introduced into poly-L-lactic acid(PLLA)to prepare bone scaffolds with photocatalytic antibacterial function via selective laser *** results showed that TiO_(2)grew on the surface of rGO and formed a covalent bond connection(Ti-O-C)with rGO.A decreased electrochemical impedance of TiO_(2)@rGO composites was observed,and the transient photocurrent intensity increased from 0.05 to 0.5μA/cm^(2).Analysis of electron spin resonance found that the photocatalytic products of TiO_(2)were·OH and·O^(2-),two kinds of ROS capable of killing bacteria via disrupting the structure of the bacterial membrane in *** experiments showed that the PLLA/TiO_(2)@rGO scaffolds had good antibacterial properties against Escherichia coli and Staphylococcus ***,we report that these scaffolds exhibited both enhanced mechanical properties due to the addition of TiO_(2)@rGO as a reinforcement material and good biocompatibility during cell proliferation.

Extracorporeal liver surgery(ELS), also known as liver autotransplantation, is a hybrid(cross-fertilized) surgery incorporating the technical knowledge from extreme liver and transplant liver surgeries, and recently became more embraced and popularized among leading centers. ELS could be summarized into three major categories, namely, ex-situ liver resection and autotransplantation(ELRA), ante-situm liver resection and autotransplantation(ALRA) and auxiliary partial liver autotransplantation(APLA). The successful development of ELS during the past 37 years is definitely inseparable from continuous effort s done by Chinese surgeons and researchers. Especially, the precision liver surgery paradigm has allowed to transform ELS into a modularized, more simplified, and standardized surgery, to upgrade surgical skills, to improve peri-operative outcome and long-term survival, to increase the capability of surgeons to select more complex diseases and to expand the level of medical service to the population. This review highlights the Chinese contributions to the field of ELS, focusing thereby on features of different surgical types, technical innovations, disease selection and surgical indication, patient prognosis and future perspectives.

Background: The available evidence regarding the benefits of percutaneous coronary intervention (PCI) on patients receiving dialysis with coronary artery disease (CAD) is limited and inconsistent. This study aimed to evaluate the association between PCI and clinical outcomes as compared with medical therapy alone in patients undergoing dialysis with CAD in ***: This multicenter, retrospective study was conducted in 30 tertiary medical centers across 12 provinces in China from January 2015 to June 2021 to include patients on dialysis with CAD. The primary outcome was major adverse cardiovascular events (MACE), defined as a composite of cardiovascular death, non-fatal myocardial infarction, and non-fatal stroke. Secondary outcomes included all-cause death, the individual components of MACE, and Bleeding Academic Research Consortium criteria types 2, 3, or 5 bleeding. Multivariable Cox proportional hazard models were used to assess the association between PCI and outcomes. Inverse probability of treatment weighting (IPTW) and propensity score matching (PSM) were performed to account for potential between-group ***: Of the 1146 patients on dialysis with significant CAD, 821 (71.6%) underwent PCI. After a median follow-up of 23.0 months, PCI was associated with a 43.0% significantly lower risk for MACE (33.9% [n = 278]vs. 43.7% [n = 142]; adjusted hazards ratio 0.57, 95% confidence interval 0.45-0.71), along with a slightly increased risk for bleeding outcomes that did not reach statistical significance (11.1%vs. 8.3%; adjusted hazards ratio 1.31, 95% confidence interval, 0.82-2.11). Furthermore, PCI was associated with a significant reduction in all-cause and cardiovascular mortalities. Subgroup analysis did not modify the association of PCI with patient outcomes. BACKGROUND Liver cirrhosis and portal hypertension(PHT)can lead to lymphatic abnormalities and coagulation *** lymphangiogenesis may relieve liver cirrhosis and PHT,the present study investigated the gene expression alterations in the lymphatic system and the effectiveness of platelet-mediated lymphangiogenesis in improving liver cirrhosis and *** To investigate the role of lymphangiogenesis in preclinical PHT *** Immunohistochemistry and transcriptome sequencing of bile duct ligation(BDL)and control lymphatic samples were conducted to reveal the indicated signaling *** enrichment analyses were performed on the differentially expressed genes and hub *** infection of vascular endothelial growth factor C(VEGF-C),plateletrich plasma(PRP),and VEGF3 receptor(VEGFR)inhibitor MAZ-51 was used as an intervention for the lymphatic system in PHT ***,hemodynamic tests and western blot analyses were performed to demonstrate the effects of lymphatic intervention in PHT *** Lymphangiogenesis was increased in the BDL rat *** sequencing analysis of the extrahepatic lymphatic system revealed its close association with platelet adherence,aggregation,and *** role of PHT in the rat model was investigated by activating(PRP)and inhibiting(MAZ-51)the lymphatic *** promoted lymphangiogenesis,which increased lymphatic drainage,alleviated portal pressure,reduced liver fibrosis,inhibited inflammation,inhibited angiogenesis,and suppressed mesenteric artery ***-51 reversed the above *** Via VEGF-C/VEGFR-3,platelets impede fibrosis,angiogenesis,and mesenteric artery remodeling,ultimately alleviating ***,platelet intervention is a therapeutic approach for cirrhosis and PHT.
