In vitro gastrointestinal digestion study of two wheat cultivars and evaluation of xylanase supplementation

Background： The filamentous fungus Talaromyces versatifis is known to improve the metabolizable energy of wheat- based poultry diets thanks to its ability to produce a pool of CAZymes and particularly endo-β（1,4）-xylanases. In order to appreciate their in vivo mode of action, the supplementation effect of two of its xylanases, XynD and XynB from families GH10 and GHll respectively, have been evaluated on two different wheat cultivars Caphorn and Isengrain, which were chosen amongst 6 varieties for their difference in non starch polysaccharides content and arabinoxylan composition. results： Polysaccharides digestion was followed during 6 h along the digestive tract using the TNO gastrointestinal model-1, to mimic monogastric metabolism. Polysaccharide degradation appeared to occur mainly at the jejunal level and was higher with Isengrain than with Caphorn. For both cultivars, XynD and XynB supplementation increased notably the amount of reducing end sugars into the jejuno-ileal dialysates, which has been confirmed by a valuable increase of the soluble glucose into the jejunal dialysates. Conclusions： The amounts of arabinose and xylose into the dialysates and ileal deliveries increased consequently mainly for Caphorn, suggesting that XynD and XynB supplementation in wheat-based diet could alleviate the anti-nutritional effects of arabinoxylans by limiting the physical entrapment of starch and could increase the available metabolizable energy.

The effect of two fermented milks （FMI, FM2） with lactic acid bacteria and Bifidobacteria on the intestine mucosa was studied. BALB/c mice were divided in groups of non-sensitized or sensitized by oral route to cow＇s milk or colonized by FM1 [10^7 cfu/mL ofBifidobacterium longum （Bfl） and 10^8 cfu/mL of Streptococcus thermophillus （Stl）] and sensitized by oral route to cow＇s milk or colonized by FM2 [10^7 cfu/mL of Bfl and 10^7 cfu/mL ofLactobacillusplantarum （LbO）] and sensitized to cow＇s milk. Blood was sampled and the amount of anti-β-Lactoglobulin （β-Lg） IgG was measured. Mice were sacrificed, fragments of their intestines were isolated to inspect the structural changes of intestinal mucosa. A significant anti-β-Lg response was elicited by oral sensitization in positive control compared to other groups. Inspection of villi structural changes reveal signs of inflammation in challenged group compared with FM1 and FM2 groups, which conserved long villi characteristic of negative controls. The colonization of intestines by BfI-StI and BfI-LbO, and the evaluation of the residual antigenicity of β-Lg in mice sensitized to bovine milk by oral route followed by histological studies, revealed that FM1 and FM2 play protective role and reduce the histological lesions typical of bovine milk allergy.

Cinnamoyl CoA reductase （CCR） and cinnamyl alcohol dehydrogenase （CAD） catalyze the last steps of mono- lignol biosynthesis. In Arabidopsis, one CCR gene （CCR1, Atlg15950） and two CAD genes （CAD C At3g19450 and CAD D At4g34230） are involved in this pathway. A triple cad c cad d ccrl mutant, named ccc, was obtained. This mutant displays a severe dwarf phenotype and male sterility. The lignin content in ccc mature stems is reduced to 50% of the wild-type level. In addition, stem lignin structure is severely affected, as shown by the dramatic enrichment in resistant inter-unit bonds and incorporation into the polymer of monolignol precursors such as coniferaldehyde, sinapaldehyde, and ferulic acid. Male sterility is due to the lack of lignification in the anther endothecium, which causes the failure of anther de- hiscence and of pollen release. The cc~ hypolignified stems accumulate higher amounts of flavonol glycosides, sinapoyl malate and feruloyl malate, which suggests a redirection of the phenolic pathway. Therefore, the absence of CAD and CCR, key enzymes of the monolignol pathway, has more severe consequences on the phenotype than the individual absence of each of them. Induction of another CCR （CCR2, Atlg80820） and another CAD （CAD1, At4g39330） does not compensate the absence of the main CCR and CAD activities. This lack of CCR and CAD activities not only impacts lignification, but also severely affects the development of the plants. These consequences must be carefully considered when trying to reduce the lignin content of plants in order to facilitate the lignocellulose-to-bioethanol conversion process.

With the analysis of the sources and formation mechanism of the clay minerals in the sediment core from the Dalianhai lake in the Gonghe Basin,northeastern Tibet-Qinghai Plateau,clay mineral composition proxies,grain-size and carbonate contents have been employed for high-resolution study in order to reconstruct East Asian Summer Monsoon （EASM） over the northeastern Tibet-Qinghai Plateau during the *** study also extended to establish a relationship between vegetation cover changes and erosion during the last 14.5 ka with pollen record and clay mineral ***/kaolinite and smectite/（illite＋chlorite） ratios allow us to assess hydrolysis conditions in lowlands and/or physical erosion process in highlands of the Gonghe *** 12.9 Cal ka BP,both mineralogical ratios show low values indicative of strong physical erosion in the basin with a dominant cold and dry *** 12.9 Cal ka BP,an increase in both mineralogical ratios indicates enhanced chemical weathering in the basin associated with a warm and humid *** beginning of Holocene is characterized by high smectite/（illite＋chlorite） and smectite/kaolinite ratios that is synchronous as with deposition of many peat laminae,implying the best warm and humid conditions specifically between 8.0 to 5.5 Cal ka *** time interval after 5.0 Cai ka BP is characterized by a return to high physical erosion and low chemical weathering with dry climate conditions in the *** variations of clay mineral assemblages with previous pollen results,we observe a rapid response in terms of chemical weathering and physical erosion intensity to a modification of the vegetation cover in the basin.

Studies on large-sample gut microbial sequencing data indicate that gut microbiota can be divided into multiple community types;different community types may influence the community function and ecosystem ***,the knowledge on the classification,diversity,interaction,and assembling of microbial community types in the gut of wild animals is still ***,we used pika gut microbiota data as an example to study the microbial community types in large-sample sequencing *** microbial communities from 118 wild plateau pika(Ochotona curzoniae)individuals at 5 elevational regions on the Qinghai–Tibet Plateau were *** results show that pika gut microbiota can be separated into 2 community types(Cluster I and Cluster II).Cluster I was mainly distributed on the high-elevation regions with more than 3694 m and was most dominated by *** II was from the low-elevation areas(lower than 3580 m),and was predominated by *** I had a higher community alpha-diversity and predicted functional diversity than Cluster II,and the betadiversity and predicted functional profiles of these 2 clusters were significantly *** analysis revealed that there were more complex interactions between Cluster I,which had enhanced influence on the co-occurrences of other microbes in the bacterial community when compared to Cluster *** analysis found that the environmental filtering in the Cluster I was stronger than Cluster *** assemblages of pika gut bacterial communities were determined mainly by deterministic processes,while the relative importance of deterministic processes accounted for more percentages in the Cluster I than Cluster *** results demonstrated that 2 gut microbial community types in pikas had distinct diversity patterns and ecological *** methods are also helpful for identifying gut community types and the related mechanisms behind gut microbiota types in large-sample sequen

对近35年来数值模拟方法,特别是经典分子动力学方法和相关的优先采样技术在生物体系研究中的应用作了回顾.由于生物体系研究对象的特点是体系空间尺度大且细胞机制时间跨度长,因此所涉及的结构生物学和生物物理学方面的研究构成了分子动力学模拟的最大挑战.从生物学的角度对分子动力学的基本理论、算法发展以及在生物体系中的应用进行综述,重点阐释在生理活动相关的时间尺度上生物体系的模拟是如何逐步发展的.另一方面,回顾了生物模拟体系在空间和时间尺度上得益于计算机硬件和算法的飞速发展而急速扩张的历程.最后,基于最近生物体系分子动力学模拟领域的尖端研究成果,对该领域未来发展的趋势进行了思考和展望.
