Artemisia vulgaris Induces Tumor-Selective Ferroptosis and Necroptosis via Lysosomal Ca^(2+) Signaling

Objective To evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCF-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3T3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/FITC-7AAD,***-1(Fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPTA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCF-7,SKBR-3 and K562 cells(P<0.05).The viability of HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(P<0.05).Treatment of K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-FITC/7-AAD(P<0.05).The use of Nec-1 and Fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(P<0.01).HEAV-induced Ca^(2+)release mainly from lysosomes in K562 cells(P<0.01).Furthermore,BAPTA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(P<0.05).Conclusions HEAV is cytotoxic and activates several modalities of cell death,which are partially dependent on lysosomal release of Ca^(2+).These effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

Subterranean termites （Isoptera： Rhinotermitidae）： Exploitation of equivalent food resources with different forms of placement

Coptotermes gestroi and Heterotermes tenuis have been described as important urban pests in Brazil. The establishment of control technologies using baits that consider the social behavior of termites requires a better knowledge of their foraging behavior. Thus, this study analyzed the feeding behavior of these species with three different forms of food placement： food on the surface of a substrate and food either partially or completely buried in the substrate. Experimental arenas were composed of a central chamber connected to three food chambers. Each central chamber contained 550 foragers of C. gestroi or 517 foragers of 11. tenuis. Blocks of Pinus elliottii were placed in the different food chambers. After 28 days, the consumption of each wood block and the percentage of foraging individuals recruited for food chambers were verified in relation to the total survival rate obtained for each one of the 20 replicates. Results showed that completely buried food was most consumed for H. tenuis and presented a higher recruitment rate of workers and soldiers for both species. Although the consumption had non-significant differences for C. gestroi, these termites exhibited a tendency to prefer completely buried food. In these conditions, it can be concluded that the forms of food placement used in the present research influenced the recruitment of individuals for both species. Data also suggests that when the foraging subterranean termites find food resources in the tunneling substrate, they tend to concentrate their efforts on it, a behavior regime that reduces exposure to external environment.

Near-infrared leaf reflectance modeling of Annona emarginata seedlings for early detection of variations in nitrogen concentration

Nitrogen(N)monitoring is essential in nurseries to ensure the production of high-quality *** spectroscopy(NIRS)is an instantaneous,nondestructive method to monitor *** data such as NIRS can also provide the basis for developing a new vegetation spectral index(VSI).Here,we evaluated whether NIRS combined with statistical modeling can accurately detect early variations in N concentration in leaves of young plants of Annona emargiaata and developed a new VSI for this *** were grown in a hydroponics system with 0,2.75,5.5or 11 mM N for 45 *** we measured gas exchange,chlorophylla fluorescence,and pigments in leaves;analyzed complete leaf nutrients,and recorded spectral data for leaves at 966 to 1685 nm using *** a statistical learning approach,the dimensionality of the spectral data was reduced,then models were generated using two classes(N deficiency,N)or four classes(0,2.75,5.5,11 mM N).The best combination of techniques for dimensionality reduction and classification,respectively,was stepwise regression(PROC STEPDISC)and linear discriminant *** was possible to detect N deficiency in seedlings leaves with 100%precision,and the four N concentrations with93.55%accuracy before photosynthetic damage to the plant ***,NIRS combined with statistical modeling of multidimensional data is effective for detecting N variations in seedlings leaves of ***.

Tree encroachment into savannas alters soil microbiological and chemical properties facilitating forest expansion

Abstract Forests have been expanding over typical savanna sites for the past 3000 years in the Neotropics. Such invasion can produce a series of environmental modifications on typical savanna; however, it remains unclear how modifications in soil properties, caused by the encroachment of woody species, facilitate the expansion of forest ecosystems under dystrophic conditions. Here we examined chemical and microbiological changes associ- ated with tree encroachment in oxisols of a Neotropical Savanna at Assis Ecological Station, Southeastern Brazil. We predicted that tree encroachment caused by typical forest species would cause significant changes in the chemical and microbiological properties of savanna soils. Soils were sampled at Assis Ecological Station, from savanna sites differing in tree encroachment （typical, dense and forested savanna） caused by decades of fire exclusion. We analysed vegetation leaf area index and leaf litter volume deposited in the studied plots and chemical （pH,organic matter, P, K, Ca, Mg, A1, NO3-, NH4＋） and microbiological （microbial C biomass and dehydrogenase activity） properties of soils under distinct encroachment conditions. Most soil chemical properties did not change along the tree encroachment gradient; however, total P, soil organic matter, soil microbial C and dehydrogenase activity increased from typical savanna to forested savanna. The changes in soil organic matter and dehydrogenase activity were correlated with the values of leaf area index and litter volume along the encroachment gradient. Our results demonstrate that forest species can increase carbon and phosphorus supplies in tropical savanna soils.

Current Topics in Acute Abdomen: Causes, Diagnostics, and Emerging Therapies

Acute abdomen is a serious clinical condition that requires immediate attention due to its association with intense pain and rapid progression. This article reviews important aspects of the causes, diagnostics, and treatments of acute abdomen. The research highlights the diversity of etiologies, ranging from the most common, such as acute appendicitis, to the less frequent, such as intestinal perforations by foreign bodies. Complex diagnostic methods, including clinical examinations and imaging such as computed tomography, are essential to guide accurate diagnosis, given the overlap of symptoms among different conditions. Therapeutic approaches range from conservative treatments with clinical observation and antibiotic therapy to urgent surgical interventions, such as appendectomy and cholecystectomy. Laparoscopy stands out as a promising approach, offering advantages in terms of recovery and post-operative morbidity. However, the impact of the COVID-19 pandemic has brought additional challenges to the diagnosis and treatment of acute abdomen, with delays and difficulties in accessing healthcare. This review underscores the importance of a multidisciplinary approach and constant updates among healthcare professionals in addressing acute abdomen, aiming for accurate diagnoses and appropriate therapeutic interventions to improve clinical outcomes and reduce morbidity and mortality associated with this complex clinical condition.

Spatial Quality of Social Housing for Seniors： Village of the Elderly in S^o Paulo （Brazil）

This paper presents the quality assessment of the built environment in the social housing for seniors ＂Village of the Elderly＂, located in the central area of Silo Paulo （Brazil） and it is the case study analyzed here. This multi-method study aimed to evaluate the quality of the built environment through the following indicators： community public facilities in the neighborhood, accessibility, safety and quality of housing. The results showed that ＂Village of the Elderly＂ was designed to house residents with income of up to three minimum wages, however, for lack of specific technical knowledge on human aging, some problems related to accessibility and safety of residents were neglected. The research contributes to studies on the quality of the ambience of constructed space and the results can support public policies, programs and projects of social housing for the elderly.

Effects of functional training on geometric indices of heart rate variability

Background： Geometric methods provide an analysis of autonomic modulation using the geometric properties of the resulting pattern, and represent an interesting tool in the analysis of heart rate variability （HRV）. The aim of this study was to evaluate the impact of functional training on cardiac autonomic modulation in healthy young women using the geometric indices of HRV. Methods： Data were analyzed from 29 women, and were stratified into a functional training group （FTG, n = 13; 23.00 ± 2.51 years; 21.90± 2.82 kg/m2） and a control group （CG, n = 16; 20.56 ± 1.03 years; 22.12±3.86 kg/m2）. The FTG received periodized functional training for 12 weeks. The cardiac autonomic modulation of both groups was evaluated before and after this training, and a qualitative analysis was performed using the Poincar6 plot. Results： There was a significant increase in the difference of the triangular index （RRTri）, SDI, SD2, and RR intervals in the FTG as compared to the CG, and the qualitative analysis from the Poincar6 plot showed an increase in the dispersion of beat-to-beat and long-term RR intervals in the functional group after training. No changes were observed in the triangular interpolation of RR interval histogram （TINN） or SD1/SD2. Conclusion： Functional training had a beneficial impact on autonomic modulation, as characterized by increased parasympathetic activity and overall variability, thus highlighting the clinical usefulness of this type of training.

Posterior partially edentulous jaws, planning a rehabilitation with dental implants

AIM: To discuss important characteristics of the useof dental implants in posterior quadrants and the rehabilitation planning. METHODS: An electronic search of English articles was conducted on MEDLINE(PubM ed) from 1990 up to the period of March 2014. The key terms were dental implants and posterior jaws, dental implants/treatment planning and posterior maxilla, and dental implants/treatment planning and posterior mandible. No exclusion criteria were used for the initial search. Clinical trials, randomized and non randomized studies, classical and comparative studies, multicenter studies, in vitro and in vivo studies, case reports, longitudinal studies and reviews of the literature were included in this review. RESULTS: One hundred and fifty-two articles met the inclusion criteria of treatment planning of dental implants in posterior jaw and were read in their entirety. The selected articles were categorized with respect to their context on space for restoration, anatomic considerations(bone quantity and density), radiographic techniques, implant selection(number, position, diameter and surface), tilted and pterygoid implants, short implants, occlusal considerations, and success rates of implants placed in the posterior region. The results derived from the review process were described under several different topic headings to give readers a clear overview of the literature. In general, it was observed that the use of dental implants in posterior region requires a careful treatment plan. It is important that the practitioner has knowledge about the theme to evaluate the treatment parameters. CONCLUSION: The use of implants to restore the posterior arch presents many challenges and requires a detailed treatment planning.

Relationship between head and neck cancer therapy and some genetic endpoints

Head and neck cancer(HNC) is the sixth most common human malignancy worldwide. The main forms of treat ment for HNC are surgery, radiotherapy(RT) and che motherapy(CT). However, the choice of therapy de pends on the tumor staging and approaches, which are aimed at organ preservation. Because of systemic RT and CT genotoxicity, one of the important side effects is a secondary cancer that can result from the activity of radiation and antineoplastic drugs on healthy cells Ionizing radiation can affect the DNA, causing single and double-strand breaks, DNA-protein crosslinks and oxidative damage. The severity of radiotoxicity can be directly associated with the radiation dosimetry and the dose-volume differences. Regarding CT, cisplatin is stil the standard protocol for the treatment of squamous cell carcinoma, the most common cancer located in theoral cavity. However, simultaneous treatment with cisplatin, bleomycin and 5-fluorouracil or treatment with paclitaxel and cisplatin are also used. These drugs can interact with the DNA, causing DNA crosslinks, double and single-strand breaks and changes in gene expression. Currently, the late effects of therapy have become a recurring problem, mainly due to the increased survival of HNC patients. Herein, we present an update of the systemic activity of RT and CT for HNC, with a focus on their toxicogenetic and toxicogenomic effects.

Intestinal anti-inflammatory activity of Ground Cherry(Physalis angulata L.)standardized CO2 phytopharmaceutical preparation

AIM To investigate the effects of Ground Cherry(Physalis angulata L.)standardized supercritical CO_2 extract in trinitrobenzenesulphonic acid(TNBS)model of rat intestinal *** The animals were divided into groups that received vehicle or *** extract(PACO_2)orally at the doses 25,50 and 100 mg/kg daily by 5 d before TNBS *** effects of PACO_2 were assessed by macroscopic analysis,biochemical determinations of the levels of myeloperoxidase(MPO),alkaline phosphatase(ALP),glutathione and cytokines(such as INF-γ,IL-1β,IL-6,IL-10 and TNF-α),gene expression evaluation(including Hsp70,heparanase,NF-κB,mitogenactivated protein kinases(Mapk)1,3,6 and 9,and the mucins genes Muc 1,2,3 and 4)and histopathological studies using optical,and electronic(transmission and scanning)*** PACO2 extract promoted a significant reduction in MPO and ALP activities,reducing oxidative stress and neutrophil *** effects were accompanied by significant reduction of colonic levels of IFN-γand IL-6 and down-regulation of heparanase,Hsp70,Mapk3,Mapk9,Muc1 and Muc2 genes expression when compared with TNBS-control *** addition,protective effects were also evidenced by reduced neutrophil infiltration,recovery of cell architecture and replacement of mucin by histopathological and ultrastructural *** Physalis angulata supercritical CO2 extract is an intestinal anti-inflammatory product that modulates oxidative stress,immune response and expression of inflammatory mediators,with potentially utility for treating inflammatory bowel disease.
