Genetic characterization and potential molecular dissemination mechanism of tet(31) gene in Aeromonas caviae from an oxytetracycline wastewater treatment system

Recently, the rarely reported tet(31) tetracycline resistance determinant was commonly found in Aeromonas salmonicida, Gallibacterium anatis, and Oblitimonas alkaliphila isolated from farming animals and related environment. However, its distribution in other bacteria and potential molecular dissemination mechanism in environment are still unknown. The purpose of this study was to investigate the potential mechanism underlying dissemination of tet(31) by analysing the tet(31)-carrying fragments in A. caviae strains isolated from an aerobic biofilm reactor treating oxytetracycline bearing wastewater. Twenty-three A. caviae strains were screened for the tet(31) gene by polymerase chain reaction(PCR). Three strains(two harbouring tet(31), one not) were subjected to whole genome sequencing using the PacBio RSII platform. Seventeen A. caviae strains carried the tet(31) gene and exhibited high resistance levels to oxytetracycline with minimum inhibitory concentrations(MICs)ranging from 256 to 512 mg/L. tet(31) was comprised of the transposon Tn6432 on the chromosome of A. caviae, and Tn6432 was also found in 15 additional tet(31)-positive A. caviae isolates by PCR. More important, Tn6432 was located on an integrative conjugative element(ICE)-like element, which could mediate the dissemination of the tet(31)-carrying transposon Tn6432 between bacteria. Comparative analysis demonstrated that Tn6432 homologs with the structure ISCR2-ΔphzF-tetR(31)-tet(31)-ΔglmM-sul2 were also carried by A. salmonicida, G. anatis, and O. alkaliphila, suggesting that this transposon can be transferred between species and even genera. This work provides the first report on the identification of the tet(31) gene in A. caviae, and will be helpful in exploring the dissemination mechanisms of tet(31) in water environment.

Effect of stocking density on performances of juvenile turbot (Scophthalmus maximus) in recirculating aquaculture systems

Limited information has been available about the influence of loading density on the performances of Scophthalmus maximus, especially in recirculating aquaculture systems （RAS）. In this study, turbot （13.84±2.74 g; average weigh±SD） were reared at four different initial densities （low 0.66, medium 1.26, sub-high 2.56, high 4.00 kg/m^2） for 10 weeks in RAS at 23±1℃ Final densities were 4.67, 7.25, 14.16, and 17.47 kg/m^2, respectively, which translate to 82, 108, 214, and 282 percent coverage of the tank bottom. Density had both negative and independent impacts on growth. The final mean weight, specific growth rate （SGR）, and voluntary feed intake significantly decreased and the coefficient of variation （CV） of final body weight increased with increase in stocking density. The medium and sub-high density groups did not differ significantly in SGR, mean weight, CV, food conversion rate （FCR）, feed intake, blood parameters, and digestive enzymes. The protease activities of the digestive tract at pH 7, 8.5, 9, and 10 were significantly higher for the highest density group, but tended to be lower （not significantly） at pH 4 and 8.5 for the lowest density group. The intensity of protease activity was inversely related to feed intake at the different densities. Catalase activity was higher （but not significantly） at the highest density, perhaps because high density started to induce an oxidative effect in turbot. In conclusion, turbot can be cultured in RAS at a density of less than 17.47 kg/m^2. With good water quality and no feed limitation, initial density between 1.26 and 2.56 kg/m^2 （final： 7.25 and 14.16 kg/m^2） would not negatively affect the turbot cultured in RAS. For culture at higher density, multi-level feeding devices are suggested to ease feeding competition.

In industrial areas, tree leaves contaminated by metals and metalloids could constitute a secondary source of pollutants. In the present study, water extraction kinetics of inorganic elements （IE： Pb, Zn, Cd, As, Fe and Mn）, dissolved organic carbon, pH and biological activity were studied for industrial contaminated poplar leaves. Moreover, the distribution of the IE through the size fractions of the associated top soil was measured. High quantities ofMn, Zn and As and polysaccharides were released in the solution from the strongly contaminated leaves. The kinetic of release varied with time and metal type. The solution pH decreased while dissolved organic contents increased with time after 30 days. Therefore, these contaminated leaves could constitute a source of more available organic metals and metalloids than the initial inorganic process particles. However, the distribution of the IE through the size fractions of the top soil suggested that a great part of the released IE was adsorbed, reducing in consequence their transfers and bioavailability. It＇s concluded that mobility/boioavailability and speciation of metals and metalloids released from the decomposition of polluted tree leaves depends on soil characteristics, pollutant type and litter composition, with consequences for environmental risk assessment.

Rubik's cube as in-situ programmable matter and a reconfigurable mechanical metamaterial

As part of the 4th industrial revolution, programmable mechanical metamaterials exhibit great application potential in flexible robotics, vibration control, and impact protection. However, maintaining a programmed state without sustaining the external stimulus is often challenging and leads to additional energy consumption. Inspired by Rubik's cube, we design and study an insitu programmable and distribution-reconfigurable mechanical metamaterial(IPDR-MM). A matrix model is developed to model IPDR-MMs and describe their morphological transitions. Based on this model, the reinforcement learning method is employed to find the pathways for morphological transitions. We find that IPDR-MMs have controllable stiffness across several orders of magnitude and a wide range of adjustable anisotropies through morphology transformation. Additionally, because of the independence of the directions of morphology transformation and bearing, IPDR-MMs exhibit good stability in bearing and can readily achieve high stiffness. The Rubik's cube-inspired design concept is also instructive for other deformable structures and metamaterials, and the current version of the proposal should be sufficiently illustrative to attract and broaden interdisciplinary interests.

NH_3 Plasma Surface Treatments of Engineering Fluoropolymers: A Way to Enhance Adhesion of Ni or Cu Thin Films Deposited by Electroless Plating

This paper describes the electroless Ni or Cu plating of some fiuoropolymer substrates through a tin-free activation process. Materials subjected to surface metallization are commercial Teflon() FEP, Nafion(), ACLAR() and LaRCTM-CP1 thin films which have recently gained a large scientific and technological interest due to their excellent thermal, chemical, mechanical and dielectric properties. The original approach implemented in the present work involves: (i)the grafting of nitrogen-containing functionalities on the polymer surfaces through plasma treatments in ammonia, (ii) the direct catalysis of the so-modified surfaces via their immersion in a simple acidic PdCl2 solution (i.e. without using a prior surface sensitization in an acidic SnCl2 solution), and finally (iii) the electroless metallization itself. However, prior to the immersion in the industrial plating baths, the chemical reduction of the Pd+2 species (species covalently tethered on the nitrogen-containing groups) to metallic palladium (PdO) is shown to be a key factor in catalyzing the electroless deposition initiation. This is made by immersion in an hypophosphite (H2PO2-) solution. Wettability measurements and X-ray photoelectron spectroscopy (XPS) experiments are used to characterize every surface modification step of the developed process. A cross-hatch tape test was used to asses the adhesion strength of the electroless films that is shown qualitatively good. In addition, a fragmentation test was developed in combination with electrical measurements. Its use allows to distinguish different adhesion levels at the metal/polymer interface and to evidence the influence of some processing parameters.

The exercise-cognition relationship:A virtuous circle

Exercise and health psychology have generated 2 sets of empirical studies guided by separate theory-driven *** first axis focuses on the causal relationship between chronic exercise and cognition and,more particularly,high-level cognitive functions such as executive functions(EFs).The second axis examines factors influencing the adherence process to physical activity(PA).Research conducted during the past decade shows that these 2 topics are closely linked,with EFs and effortful control playing a pivotal role in the bidirectional relationship linking PA and mental/brain *** present article supports the idea that an individual engaged in the regular practice of effortful PA initiates a virtuous circle linking PA and effortful control in a bidirectional *** the one hand,chronic exercise leads to an improvement of EFs and effortful *** the other hand,gains in EFs and effortful control effectiveness lead to a reciprocal facilitation of the maintenance of PA over *** limitations and perspectives to this effort hypothesis are proposed in the last part of the article.

Stochastic Simulation of Saline Intrusion in the Coastal Aquifer of Saloum, Senegal

In the Saloum region of central-western Senegal, water needs are essentially met by tapping an underground aquifer associated with the sandy-clay formations of the Continental Terminal, in contact with both the ocean to the west and the highly saline waters of the Saloum River to the north. In this estuarine and deltaic zone with its very low relief, the hydraulic loads in the water tables are generally close to zero or even negative, creating a reversal of the natural flow and encouraging saline intrusion into this system, which makes it very vulnerable. This study concerns the implementation of a numerical model of saline intrusion to provide a better understanding of the vulnerability of the water table by analyzing the variability of the freshwater/saltwater interface. The Modflow-2005 code is used to simulate saline intrusion using the SWI2 module, coupled with the GRASS (Geographic Resources Analysis Support System) software under the Linux operating system with the steep interface approach. The probable expansion of the wedge is studied in three scenarios, taking into account its position relative to the bedrock at 1 m, 5 m and 10 m. Simulations carried out under imposed potential and river conditions, based on variations in groundwater reserves using two effective porosity values, 10−1 and 10−2, show that the water table is highly vulnerable in the northwest sector. The probable expansion of the wedge increases as the storage coefficient decreases and is more marked with river conditions in the areas surrounding the Saloum River, reaching 6 km with a probability of 1. The probability of the wedge reaching a certain degree of expansion decreases from 1 to 0.5, and then cancels out as it moves inland. The probable position of the wedge is limited to 500 m or even 1 km depending on the corner around the coast to the southwest and in the southern zone. Local Criteria Used by Farmers to Evaluate the Agronomic Performance and the Fertilizing Capacity of Cowpea Varieties: Diversity, Variability and Proximal Relation with Agronomic Measurements in Contrasted Sahelian Locations

When contributing to participatory research, farmers usually appreciate the performance of cowpea varieties using qualitative scores. The score they attribute to each variety are based on local knowledge. The specific criteria they individually use to attribute a score are not well described. The objectives of this work were to: 1) identify and describe exhaustively the local criteria used by farmers to measure the agronomic performance of cowpea;2) assess the variability and statistical structure of these farmer criteria across local contexts;3) and analyze the association between these farmer criteria and the classical agronomic measurement. To achieve these objectives, an augmented block design was implemented across fifteen locations in the regions of Maradi, Dosso and Tillabéri, representing a diversity of local contexts. From a set of 36 cowpea varieties, fifteen varieties were sown per location, including five varieties (controls) common to all locations. In each location, two replicates were sown in randomized Fisher’s blocks. After agronomic measurement and participatory evaluation (scoring of varieties by farmers), a group survey (focus group) was conducted in each location to identify the criteria considered by farmers to found their discretional scoring of varieties during the participatory evaluation. The analysis of the data identified, across locations, thirteen criteria defined by farmers to characterize the agronomic performance of cowpea. Some of these criteria were different according to location. Farmers ranked the three varieties with the best performance for each agronomical trait (Top 3 varieties). A comparison of the farmer ranking with the ranking based on agronomic measurements revealed similarity and complementary between both methods. This study highlighted the importance of considering both local and scientific knowledge in local varietal evaluations.
