Human intestinal Caco-2 cell model to evaluate the absorption of 7-ketophytosterols and their effects on cholesterol transport

7-Ketophytosterols are the major oxidation products of phytosterols in foods, which have been associated with atherosclerosis. However, their absorption mechanism remains unclear. The aim of our work was to investigate the absorption mechanism of 7-ketophytosterols and their effects on the cholesterol transport using Caco-2 cell model. The absorption percentage of 7-ketositosterol and 7-ketocampesterol was 1.16%-1.68% and 1.18%-2.23% respectively in the Caco-2 model, which is higher than that of their parent phytosterols, but lower than cholesterol-d7. The apparent permeability of 7-ketositosterol and 7-ketocampesterol at 30 μmol/L in the basolateral(BL)-to-apical(AP)direction were 0.42-and 0.55-fold of that in the AP-to-BL direction, indicating an active intake in the permeation mechanism of 7-ketophytosterols. Ezetimibe could significantly inhibit the transport of 7-ketophytosterols(P < 0.05), which means that their transport depends on niemann-pick c1-like 1(NPC1L1)protein. The transport of cholesterol-d7 was significantly inhibited by 7-ketophytosterols(P < 0.05). Taken together, this study deepened our understanding of the absorption mechanism of common food-born 7-ketophytosterols and provides useful information on the inhibition of 7-ketophytosterols absorption.

A time fractional model to represent rainfall process

This paper deals with a stochastic representation of the rainfall process. The analysis of a rainfall time series shows that cumulative representation of a rainfall time series can be modeled as a non-Gaussian random walk with a log-normal jump distribution and a time-waiting distribution following a tempered a-stable probability law. Based on the random walk model, a fractional Fokker-Planck equation （FFPE） with tempered a-stable waiting times was obtained. Through the comparison of observed data and simulated results from the random walk model and FFPE model with tempered a-stable waiting times, it can be concluded that the behavior of the rainfall process is globally reproduced, and the FFPE model with tempered a-stable waiting times is more efficient in reproducing the observed behavior.

Synergism of Zinc Oxide/Organoclay-Loaded Poly(lactic acid) Hybrid Nanocomposite Plasticized by Triacetin for Sustainable Active Food Packaging

The synergistic effect of organoclay(OC)and zinc oxide(ZnO)nanoparticles on the crucial properties of poly(lactic acid)(PLA)nanocompositeﬁlms was systematically investigated *** their incorporation into PLA via the solvent casting technique,the water vapor barrier property of the PLA/OC/ZnOﬁlm improved by a maximum of 86%compared to the neat PLAﬁlm without the deterioration of Young’s modulus or the tensile ***,theﬁlm’s self-antibacterial activity against foodborne pathogens,including gram-negative(Escherichia coli,***)and gram-positive(Staphylococcus aureus,***)bacteria,was enhanced by a max-imum of approximately 98–99%compared to the neat PLAﬁ***,SEM images revealed the homo-geneous dispersion of both nano-ﬁllers in the PLA ***,the thermal stability of theﬁlm decreased slightly after the addition of the OC and ***ﬁlm exhibited notable light barrier properties in the UV-Vis ***,the incorporation of a suitable biodegradable plasticizer signiﬁcantly decreased the Tg and notably enhanced theﬂexibility of the nanocompositeﬁlm by increasing the elongation at break approxi-mately 1.5-fold compared to that of the neat PLAﬁ*** contributes to its feasibility as an active food packa-ging material.

Three-dimensional reconstruction and comparison of vacuolar membranes in response to viral infection

The vacuole is a unique plant organelle that plays an important role in maintaining cellular homeostasis under various environmental stress conditions. However, the effects of biotic stress on vacuole structure has not been examined using three-dimensional(3D) visualization. Here, we performed 3D electron tomography to compare the ultrastructural changes in the vacuole during infection with different viruses. The 3D models revealed that vacuoles are remodeled in cells infected with cucumber mosaic virus(CMV) or tobacco necrosis virus A Chinese isolate(TNV-AC), resulting in the formation of spherules at the periphery of the vacuole. These spherules contain neck-like channels that connect their interior with the cytosol. Confocal microscopy of CMV replication proteins 1 a and 2 a and TNV-AC auxiliary replication protein p23 showed that all of these proteins localize to the *** microscopy revealed that the expression of these replication proteins alone is sufficient to induce spherule formation on the tonoplast, suggesting that these proteins play prominent roles in inducing vacuolar membrane remodeling. This is the first report of the 3D structures of viral replication factories built on the tonoplasts. These findings contribute to our understanding of vacuole biogenesis under normal conditions and during assembly of plant(+) RNA virus replication complexes.

The foxtail millet Si69 gene is a Wali7 (wheat aluminum-induced protein 7) homologue and may function in aluminum tolerance

We isolated a clone, named Si69, from a foxtail millet immature seed cDNA library. The protein encoded by Si69 contains a conserved Wali7 (wheat aluminum induced protein 7) domain and shares high-level homology with aluminum-induced proteins from other species including rice and Arabidopsis. The Si69 gene presents as a single locus in foxtail millet genome and is globally expressed in all tissues examined. Its expression is up-regulated by aluminum. The sequence feature and expression pattern suggest that the Si69 gene is involved in aluminum tolerance or detoxification. To confirm its biological functions, Si69 controlled by the CaMV35S promoter was introduced into Arabidopsis. Transgenic plants did not show any visible morphological changes compared to wild-type plants under normal growth conditions. However, when treated with 20 or 50 μmol/L Aluminum (Al), the root apices of wild-type plants were heavily stained by hematoxylin, whereas those of Si69 transgenic plants were not stained when treated with 20 μmol/L Al and slightly stained when treated with 50 μmol/L Al. Scanning electron microscopy (SEM) results further demonstrated that the damage of the root apices was se- verer in wild-type plants than in transgenic plants. Inhibition of root growth and accumulation of malondialdehyde (MDA), an indicator of lipid peroxidation, were lower in transgenic plants than in wild-type plants. The results show that overexpression of Si69 may increase Al tolerance in transgenic plants, indicating that a series of Wali7-containing genes may play similar roles in Al tolerance/detoxification.

The Green Revolution （GR） based on semi-dwarf breeding has greatly increased cereal crop yields since the 1960＇*** to appropriate plant height, the semi-dwarf crop plants are beneficial to increase light utilization and improve the lodging-resistance, and eventually increase yield potential （Hedden, 2003）. During the past 50 years, the worldwide application of semi-dwarf crop breeding has produced plenty of elite varieties. The characters of Green Revolution varieties （GRVs） are caused by the mutant growth-repressing genes.

Enrichment of Cd^(2＋) from water with a calcium alginate hydrogel filtration membrane

A new method was developed for effective enrichment of Cd2+ ions from water with a calcium alginate(CaAlg) hydrogel filtration membrane. First, the CaAlg hydrogel filtration membrane was prepared without a pore-forming agent. This membrane was used to remove Cd^(2+) via ion exchange with Ca^(2+), and the Cd^(2+) was preserved in the CaAlg hydrogel. Then, the CaAlg hydrogel containing Cd^(2+) was soaked in a sodium citrate solution, and the hydrogel was fully dissolved. The removal rate of the CaAlg filtration membrane reached almost 100% within 120 min when the Cd^(2+) concentration was under 1 mg/L. Factors affecting the removal rate were investigated, such as NaAlg concentration, operating pressure, operating time and the initial concentration of Cd^(2+). The effects of initial Cd^(2+) concentration, pressure and filtration time on the enrichment factor were also investigated. The results show that the enrichment factor reached 87.3 when the pressure was 0.18 MPa and the filtration time was 240 min. Different enrichment factors could be achieved by adjusting the operating pressure and filtration time.

Xinjiang is the main producing area of raisins and the largest green raisins production base in *** browning of Thompson seedless grape raisin is extremely serious during the drying process,and has become the key issue in the development of Xinjiang raisin *** studies have shown that dehydration speed has a great impact on the browning of Thompson seedless grape,but few relevant mechanisms have been ***,we demonstrate the effect of dehydration speed on lipid metabolism and its relation to the browning of the Thompson seedless grape during *** to slow dehydration treatment,rapid dehydration treatment of the Thompson seedless grape exhibited a lower degree of browning and activities of lipoxygenase(LOX),a higher index of unsaturated fatty acids and degree of unsaturated fatty ***,the Thompson seedless grape treated with rapid dehydration resulted in a lower rate of superoxide anion production,hydrogen peroxide content,membrane permeability,and malondialdehyde *** findings demonstrate that rapid dehydration inhibiting the browning of Thompson seedless grapes might be due to the inhibiting activities of LOX and the lower accumulation of reactive oxygen *** activities can inhibit lipid peroxidation and slow the decomposition of unsaturated fatty acid in the membrane in Thompson seedless grapes,protecting the cellular membrane structural integrity,which may result in less contact of polyphenol oxidase with phenolic substrates and less enzymatic browning during *** results provide a theoretical basis for the application of rapid dehydration in drying Thompson seedless grapes.

NBO Analysis by ONIOM and DFT (B3LYP) Calculations of Intramolecular and Intermolecular Interactions of Artemisinin, Quinine and Thirteen Manzamenones with H₂O or Alanine

This work was undertaken to analyze intramolecular and intermolecular interactions of Manzamenones from natural bond orbitals (NBO method). For their use in the treatment of malaria, the results of these molecules are compared to those of Artemisinin and Quinine. Manzamenones are a class of atypical fatty acids. They are isolated from a marine sponge of the genus Plakortis kenyensis. The analysis of intramolecular interactions compares the results of each molecule (Manzamenones, Artemisinin and Quinine) in the non-complexed state with those of its complex with a water molecule. Thus, for the same electron donors (i) and associated acceptors (j), the electron density (ED), stabilization energy E2 related to the delocalization of i to j, the energies of the NBO orbitals εi and εj of the donor and acceptor, respectively, and element of the Fock matrix Fi,j are determined and compared. The change in E2 is used to deduce whether or not the molecule is stabilized after complex formation. These analyses allowed to match each Manzamenone to one of the two antimalarials. The intermolecular interactions were analyzed, for each molecule (Manzamenones, Artemisinin and Quinine), in two complexes. These complexes are obtained with a water molecule on the one hand and with an alanine molecule on the other hand. For these interactions, the electron donor and its electron density, the electron acceptor and its electron density as well as the donor—acceptor stabilization energy have been calculated. The ONIOM 2 method is used to study Manzamenones. Theoretical calculations were done using density functional theory (B3LYP) by combining one of the two function bases 6-31++G(d,p) and 6-31+G(d,p).

Genome-wide analysis of nucleotide-binding site disease resistance genes in Medicago truncatula

The class of nucleotide-binding site(NBS)-Leucine-rich repeat(LRR) disease resistance genes play an important role in defending plants from a variety of pathogens and insect pests. Consequently, many NBS-LRR genes have been identified in various plant species. In this study, we identified 617 NBS-encoding genes in the Medicago truncatula genome(Mt3.5v5) and divided them into two groups, regular(490) and non-regular(127) NBSLRR genes. The regular NBS-LRR genes were characterized on the bases of structural diversity, chromosomal location, gene duplication, conserved protein motifs, and EST expression profiling. According to N-terminal motifs and LRR motifs, the 490 regular NBS-LRR genes were then classified into 10 types: CC-NBS(4), CC-NBS-LRR(212), TIR-NBS(20), TIR-NBS-LRR(160), TIR-NBS-TIR(1), TIR-NBS-TIR-LRR(2), NBS-TIR(7), NBS-TIR-LRR(1), NBS(10), and NBS-LRR(73). Analysis of the physical location and duplications of the regular NBS-LRR genes revealed that the M. truncatula genome is similar to rice. Interestingly, we found that TIR-type genes are more frequently expressed than non-TIR-type genes in M. truncatula, whereas the number of non-TIR-type regular NBSLRR genes was greater than TIR-type genes, suggesting the gene expression was not associated with the total number of NBS-LRR genes. Moreover, we found that the phylogenetic tree supported our division of the regular NBS-LRR genes into two distinct clades(TIR-type and non-TIR-type), but some of the non-TIR-type lineages contain TIR-type genes. These analyses provide a robust database of NBS-LRR genes in M. truncatula that will facilitate the isolation of new resistance genes and breeding strategies to engineer disease resistance in leguminous crop.
