The effects of sample pretreatments on the determination of raw materials of Mei-ting in the grass carp muscle was explored by high performance liquid *** show that the pretreatment methods of methanol as extraction agents,methylene chlorideas a purifying agent,oscillation centrifugal,n-hexane reducing fat,30℃rotation concentratedevaporation,is rapid,sensitive,economical,safe and strong *** minimumdetectable amount of raw materials of Mei-ting were1.0μg/*** averaged recovery rangedfrom 67.72%～95.52%at spiking levels of1.0～100μg/*** relative standard deviation was7.76%～37.81%.It is suitable to measure the residues of raw materials of Mei-ting in fish.

A synthetic method was developed to chemically attach few molecules of simple sugars like glucose, mannose, galactose, maltose and xylose (0.09 - 0.37 wt%) and with quaternary nitrogen pendants (0.42 - 0.46 atomic%) along the polybutadiene section of polystyrene-block-polybutadiene-block-polystyrene (sBs) block copolymer. These functionalized sBs copolymers were evaluated for biodegradation using the fungal culture Aspergillus niger NCIM 1025 (ATCC 9642) and bacterial culture Pseudomonas sp. NCIM 2220, and for antimicrobial properties using bacteria E. coli DH5α and Bacillus subtilis and yeasts Pichia stipitis NCIM 3497 and P. stipitis NCIM 3499. It was conclusively demonstrated that these modified sBs block copolymers were significantly more biodegradable than the unmodified sBs;the observed weight loss after biodegradation was ~4 - 14-fold for bacterial and ~7 - 36-fold for fungal cultures with respect to the sugar content of modified sBs. Preliminary studies on antimicrobial properties of these biodegradable polymers showed a 4% - 24% decrease in growth of the microorganisms E. coli and Bacillus subtilis studied.

Host genes involved in lipid metabolism are differentially affected during the early stages of hepatitis C virus （HCV） infection. Here we demonstrate that artificial up-regulation of fatty acid biosynthesis has a positive effect on the replication of the HCV full-length replicon when cells were treated with nystatin. Conversely, the HCV RNA replication was decreased when fatty acid biosynthesis was inhibited with 25-hydroxycholesterol and PDMP（D-threo-l-phenyl-2-decanoylamino-3- morpholino-l-propanol）. In agreement with these results, the expression level of GlcT-l（ceramide glucosyltransferase）, a host glucosyltransferase in the first step of GsL （glycosphingolipid） biosynthesis, was found to be closely associated with the expression and replication of HCV RNA. On the other hand, the viral RNA can also activate GlcT-1 in the early stage of viral RNA transfection in vitro. To identify viral factors that are responsible for GlcT-1 activation, we constructed ten stable Vero cell lines that express individual HCV proteins. Based on the analyses of these cell lines and transient transfection assay of the GlcT-1 promoter regions, we conclude that HCV proteins, especially Ns5A and Ns5B, have positive effects on the expression of GlcT-1. It is possible that Ns5A and Ns5B stimulate transcription factor（s） to activate the expression of GlcT-1 by increasing its transcription level

Cobalamin(Cbl)-dependent radical s-adenosyl-L-methionine(sAM) proteins constitute the largest collection of the radical sAM superfamily that has hundreds of thousands of individual members. Many of these proteins are involved in the biosynthesis of pharmaceutically important natural products to catalyze chemically demanding reactions. In the biosynthetic pathway of chuangxinmycin(CXM), a unique indole alkaloid antibiotic with potent anti-infective activity, functionalization of the characteristic thiopyrano[4,3,2-cd]indole scaffold by regio-and stereoselective C3-methylation is believed to rely on a Cbl-dependent radical process, which, however, remained to be reconstituted biochemically. We here report the dissection of this enzymatic process, which requires the incorporation of Cxm8, a Cbl-dependent radical sAM protein, with Cxm9, a DuF5825family protein that shares no homology to any proteins of known functions. Cxm8 and Cxm9 function together by forming an unexpected heterodimeric complex that selectively catalyzes C3-methylation of the tricyclic indole-s-hetero ring system in a successive manner, achieving CXM and a recently identified, C3-dimethylated congener. Detailed biochemical characterization,isotope labeling, structural simulation and bioinformatics analysis rationalized the catalysis of the Cxm8/Cxm9 complex and particularly the necessity of the DuF5825 protein for C3-methylase activity. This is the first example that a Cbl-dependent protein acts with a partner to exhibit radical sAM activity.

Anthracnose disease caused by the Colletotrichum gloeosporioides species complex is a major problem *** this study,we investigated the phylogenetic diversity of 207 Indian Colletotrichum isolates,associated with symptomatic and asymptomatic tissues of mango,belonging to this species *** analyses were performed based on a 6-gene dataset(act,cal,chs1,gapdh,ITs and tub2),followed by ApMat *** ApMat-based phylogeny was found to be superior as it provided finer resolution in most of the species-level ***,the ApMat marker identified seven lineages within *** sensu lato,including ***-sambac,***,***,*** sensu stricto and three undesignated,potentially novel *** this study,*** sensu stricto,***,***-sambac,*** and five undesignated,potentially novel lineages were found to be associated with mango *** is a need to develop a consensus among mycologists as to which genes should be used to define and delimit a Colletotrichum species and in the mean time mycologists should voluntarily restrain from describing new species based on inadequate datasets.

[ Objective] This study aimed to develop a kit to rapidly choose an appropriate medicine for maricultural bactedosis. [ Method] By screening enriched bacteria, solid medium and medicine and comparing effects of rapidly choosing medicine, a kit that could pick up the right medicine quickly for mariculture bacteriosis was created. [Result] The pilot test in some regions showed the kit had ideal yesalts and was easy to operate. [ Conclusion] The kit, a convenient and useful kit to detect drug sensitivity, can help users select effective drugs from many common medicines rapidly and accurately, and it is mainly suitable to use in the rapid selection of an appropriate medicine for maricultural bacteriosis.

Baculoviruses are insect-specific viruses with a circular double-stranded DNA genome ranging in size from 80-180 kb （Lu et al., 2012）. Two distinct types of viri- ons have been identified during the infectious cycle of baculoviruses, namely budded virions （BVs） and occlu- sion-derived virions （ODVs）. BVs mediate infection from cell to cell, while ODVs initiate oral infection in the insect midgut （Braunagel and summers, 2007）.

Numerical simulation of meso-β-scale convective cloud systems associated with a PRE-sTORM MCC case has been carried out using a 2-D version of the Csu Regional Atmospheric Modeling system (RAMs) nonhydrostatic model with parameterized microphysics. It is found that the predicted meso-r-scale convective phenomena arc basically unsteady under the situation of strong shear at low-levels, while the meso-β-scale convective system is maintained up to 3 hours or more. The meso -β- scale cloud system exhibits characteristics of a multi-celled convective storm in which the meso-r-scale convective cells have lifetime of about 30 min. Pressure perturbation depicts a meso-low after a half hour in the low levels. As the cloud system evolves, the meso-low intensifies and extends to the upshear side and covers the entire domain in the mid-lower levels with the peak values of 5-8 hPa. Temperature perturbation depicts a warm region in the middle levels through the entire simulation period. The meso-r-scale warm cores with peak values of 4-8 ℃ are associated with strong convective cells. The cloud top evaporation causes a stronger cold layer around the cloud top *** of microphysics exhibits that graupel is primarily concentrated in the strong convective cells forming the main source of convective rainfall after one hour of simulation time. Aggregates are mainly located in the stratiform region and decaying convective cells which produce the stratiform rainfall. Riming of the ice crystals is the predominant precipitation formation mechanism in the convection region, whereas aggregation of ice crystals is the predominant one in the stratiform region, which is consistent with observations. sensitivity experiments of ice-phase mierophysical processes show that the microphysical structures of the convective cloud system can be simulated better with the diagnosed aggregation collection efficiencies.

Activating protein-1(AP-1)composed of Jun and Fos family proteins,which are frequently activated by pathogens to induce cellular inflammation and immune *** this study,by using RNA sequencing(RNA-seq)technology,we found that FosB,Fosl2,and JunD expression was co-induced 12 and 20 h in grass carp(Ctenopharyngodon idella)kidney cell line(CIK cells)after grass carp reovirus(GCRV)***-length cDNA of the three genes were cloned and phylogenetic analysis indicated that the corresponding proteins shared high homology with those in higher *** tissue distribution analysis of the three genes showed they were ubiquitously expressed in various tissues of healthy grass *** infection assay,FosB,Fosl2,and JunD were up-regulated and co-expressed 8 h after GCRV infection in CIK *** interaction and co-expression network analysis revealed that FosB,Fosl2,and JunD showed similar functions and were co-expressed in mammals and *** together,these results indicated that the co-expression of the AP-1 family proteins in grass carp uniformly responds to GCRV *** findings provided insights into the further study of AP-1 transcription factors function during grass carp reovirus infection.

Comparative Transcriptome Analysis Reveals Different Host Cell Responses to Acute and Persistent Foot-and-Mouth Disease Virus Infection

Foot-and-mouth disease virus(FMDV) rapidly causes cytopathic effects in susceptible cells. Incomplete viral clearance during the acute infection leads to persistent infection. The relationship between host gene expression and the persistent infection remains unclear. In this study, we analyzed the transcriptome profiles of BHK-21 cells acutely and persistently infected with FMDV to identify differences in gene expression. GO and KEGG enrichment analyses showed that the 8,378 differentially expressed genes were significantly enriched in categories including metabolism, biosynthesis, ribosome function, and endocytosis. In persistently infected BHK-21 cells, ribosome-and translation-related genes were significantly down-regulated. There were more differentially expressed immune-related genes during persistent infection than during acute infection. Two hundred and seventy-four genes were differentially expressed in both acutely and persistently infected BHK-21 cells. Among these genes, heat shock protein family B member 1(Hspb1) knockdown significantly inhibited FMDV replication. Our research provides a basis for further research to understand the mechanisms of persistent FMDV infection including the genes involved in FMDV replication.
