Background: Dietary supplement use(protein/amino acids, weight-loss supplements, performance enhancers) is common among u.s. military members. Reported dietary supplement use in deployed troops is limited and is of concern in settings where troops are exposed to high ambient temperatures, increased physical demands, and dehydration. Our objective was to describe dietary supplement use and adverse events(AEs) among deployed u.s. service members compared with their pre-deployment ***: We conducted an institutional review board(IRB) approved, descriptive study in Afghanistan using a written qu.s.ionnaire and collected demographic information, dietary supplement use before and during deployment, AEs associated with supplement use, and physical workout routines. Participants were u.s. military personnel of all branches of service deployed to Afghanistan. They were recruited in high-traffic areas in the combat theater. We analyzed the data with descriptive statistics. Paired t-test/Wilcoxon signed-rank test was conducted to examine the before/during deployment changes for continuous data, and Mc Nemar's chi-square test was conducted for categorical data. We constructed separate logistic regression models to determine the best predictors of increases or decreases in dietary supplement use, with demographic information, reasons for using supplements, and education requ.s.ed/received as covariates in each model. All statistical tests were two-sided at a significance level of 5%(Psults: Data were collected on 1685 participants. Ninety-seven of the participants were in the army or Air Force. The participants were more likely to work out daily or more than once a day during deployment. Thirty-five percent of the participants reported no supplement use before or during deployment. Novel serum microRNAs panel on the diagnostic and prognostic implications of hepatocellular carcinoma

AIM To determine a panel of serum micro RNAs(mi RNAs) that could be used as novel biomarkers for diagnosis of hepatocellular carcinoma(HCC).METHODs We initially screened 9 out of 754 serum mi RNAs by Taq Man Low Density Array in two pooled samples respectively from 35 HCC and 35 normal controls, and then validated individually by RT-qP CR in another 114 patients and 114 controls arranged in two phases. The changes of the selected mi RNAs after operation and their prognostic value were *** miR-375, miR-10 a, miR-122 and miR-423 were found to be significantly higher in HCC than in controls(P under the receiver-operating-characteristic curve for the 4-miR NA panel was 0.995(95%CI: 0.985-1). All the four mi RNAs were significantly reduced after surgical removal of the tumors(P still higher than normal controls(at least P usION The four serum miR NAs(miR-375, miR-10 a, miR-122 and miR-423) could potentially serve as novel biomarkers for the diagnostic and prognostic of HCC.

Although DNA mutation drives stem cell aging,how mutation-accumulated stem cells obtain clonal advantage during aging remains poorly ***,using a mouse model of irradiation-induced premature aging and middle-aged mice,we show that DNA mutation accumulation in hematopoietic stem cells(HsCs)during aging upregulates their surface expression of major histocompatibility complex class II(MHCII).MHCII upregulation increases the chance for recognition by bone marrow(BM)-resident regulatory T cells(Tregs),resulting in their clonal expansion and accumulation in the HsC *** the basis of the establishment of connexin 43(Cx43)-mediated gap junctions,BM Tregs transfer cyclic adenosine monophosphate(cAMP)to aged HsCs to diminish apoptotic priming and promote their survival via activation of protein kinase A(PKA)***,targeting the HsC–Treg interaction or depleting Tregs effectively prevents the premature/physiological aging of *** findings show that aged HsCs use an active self-protective mechanism by entrapping local Tregs to construct a prosurvival niche and obtain a clonal advantage.

Background:ultrasound-triggered microbubble destruction(uTMD) is a widely used noninvasive technology in both military and civilian medicine,which could enhance radiosensitivity of various ***,little information is available regarding the effects of uTMD on radiotherapy for glioblastoma or the underlying *** study aimed to delineate the effect of uTMD on the radiosensitivity of glioblastoma and the potential involvement of ***:GL261,u251 cells and orthotopic glioblastoma-bearing mice were treated with ionizing radiation(IR) or IR plus *** was observed by confocal microscopy and transmission electron *** blotting and immunofluorescence analysis were used to detect progesterone receptor membrane component 1(PGRMC1),light chain 3 beta 2(LC3B2) and sequ.s.osome 1(sQsTM1/p62) *** vectors or siRNAs transfection,and fluorescent probes staining were used to explore the underlying ***:uTMD enhanced the radiosensitivity of glioblastoma in vitro and in vivo(PuTMD inhibited autophagic flux by disrupting autophagosome-lysosome fusion without impairing lysosomal function or autophagosome synthesis in IR-treated glioblastoma *** of autophagy by 3-methyladenine,bafilomycin A1 or ATG5 siRNA had no significant effect on uTMD-induced radiosensitization in glioblastoma cells(Psimilar results were found when autophagy was induced by rapamycin or ATG5 overexpression(P>0.05).Furthermore,uTMD inhibited PGRMC1expression and binding with LC3B2 in IR-exposed glioblastoma cells(PsiRNA pretreatment enhanced uTMD-induced LC3B2 and p62 accumulation in IR-exposed glioblastoma cells,thereby promoting uTMD-mediated radiosensitization(Pssion abolished uTMD-caused blockade of autophagic degradation,su.s.quently inhibiting uTMD-induced radiosensitization of glioblastoma ***,compared with IR plus uTMD group

目的　观察不同浓度哌拉西林/他唑巴坦在体外诱导铜绿假单胞菌释放DNA的情况。方法　选择体外对铜绿假单胞菌菌株敏感的哌拉西林/他唑巴坦为本研究所用抗生素。制备铜绿假单胞菌菌液后,测定其浓度并检测哌拉西林/他唑巴坦最低抑菌浓度(MIC)和最低杀菌浓度(MBC).将不同稀释度的哌拉西林/他唑巴坦加入铜绿假单胞菌菌液中,于37℃下培养4、24h,取其上清液,采用加入sYBRGold染料的DNA电泳法检测其DNA释放情况。　结果　无论有无哌拉西林/他唑巴坦的作用,体外培养4h的铜绿假单胞菌均未测得DNA释放。无哌拉西林/他唑巴坦时,铜绿假单胞菌在体外培养24h可自发释放一定量的DNA分子,其碱基对数目分布在>2 000bp和<100bp两个区域;以第3次测定的MIC(0. 008g/L)为例,哌拉西林/他唑巴坦浓度在亚MIC(0. 002、0. 004g/L)时,诱导铜绿假单胞菌释放的DNA分子碱基对数目仍分布在上述两个区域, 但<100bp的DNA分子明显增多。哌拉西林/他唑巴坦在大于MIC时,仅诱导铜绿假单胞菌释放少量<400bp的DNA.结论　自然生长情况下铜绿假单胞菌可释放一定量的DNA分子。不同浓度的哌拉西林/他唑巴坦诱导铜绿假单胞菌释放的DNA分子,在碱基对数目和分子数量方面均有明显差别。

Low back pain,mainly caused by intervertebral disc degeneration(IVDD),is a common health problem;however,current surgical treatments are less than ***,it is essential to develop novel non-invasive surgical methods for IVDD ***,we describe a therapeutic strategy to inhibit IVDD by injecting hydrogels modified with the extracellular matrix of costal cartilage(ECM-Gels)that are loaded with cartilage endplate stem cells(CEsCs).After loaded with CEsCs overexpressing sphk2(Lenti-sphk2-CEsCs)and injected near the cartilage endplate(CEP)of rats in vivo,ECM-Gels produced sphk2-engineered exosomes(Lenti-sphk2-Exos).These exosomes penetrated the annulus fibrosus(AF)and transported sphk2 into the nucleus pulposus cells(NPCs).sphk2 activated the phosphatidylinositol 3-kinase(PI3K)/p-AKT pathway as well as the intracellular autophagy of NPCs,ultimately ameliorating *** study provides a novel and efficient non-invasive combinational strategy for IVDD treatment using injectable ECM-Gels loaded with CEsCs that express sphk2 with sustained release of functional exosomes.
