Mechanomedicine:From Mechanogenomics to Mechano-Immunoengineering

Mechanobiology is a rapidly growing field that has expanded significantly over the past 40 years. There have been great examples of mechanobiology research on the cardiovascular, musculoskeletal, and respiratory systems. The field has evolved from the system level to the organ and tissue levels, and down to the cellular and molecular levels, benefiting from scientific progress and technological developments in molecular and cell biology, biomaterials, imaging techniques, and computational tools. Additionally, the interdisciplinary integration of mechanobiology with cell engineering, tissue engineering, and drug delivery has led to promising mechanomedicine applications in both therapeutics and ***, I will discuss two examples at the molecular and cellular levels:(1) mechanical regulation of epigenomics for cell reprogramming, and (2) engineering the mechanical properties of artificial antigen-presenting cells (APCs) for immune cell modulation and cancer *** memory of its identity is determined by the epigenetic state of the cells. However, how cells control the epigenetic state and thus cell fate is not well understood, and how mechanical factors such as surface topography, cell morphology,mechanical properties of the extracellular matrix (ECM), and the mechanical deformation of cells regulate the epigenetic state is not clear. In an early study, we showed that biophysical cues, in the form of parallel microgrooves on the surface of cell-adhesive substrates, can replace the effects of small-molecule epigenetic modifiers and significantly improve reprogramming efficiency from fibroblasts to induced pluripotent stem cells. The mechanism relies on the mechanomodulation of the cells’ epigenetic state through the activity of histone deacetylase and H3 methyltransferase. DiffGR:Detecting Differentially Interacting Genomic Regions from Hi-C Contact Maps

Recent advances in high-throughput chromosome conformation capture(Hi-C)techniques have allowed us to map genome-wide chromatin interactions and uncover higher-order chromatin structures,thereby shedding light on the principles of genome architecture and ***,statistical methods for detecting changes in large-scale chromatin organization such as topologically associating domains(TADs)are still ***,we proposed a new statistical method,DiffGR,for detecting differentially interacting genomic regions at the TAD level between Hi-C contact *** utilized the stratum-adjusted correlation coefficient to measure similarity of local TAD *** then developed a nonparametric approach to identify statistically significant changes of genomic interacting *** simulation studies,we demonstrated that DiffGR can robustly and effectively discover differential genomic regions under various ***,we successfully revealed cell type-specific changes in genomic interacting regions in both human and mouse Hi-C datasets,and illustrated that DiffGR yielded consistent and advantageous results compared with state-of-the-art differential TAD detection *** DiffGR R package is published under the GNu General Public License(GPL)≥2 license and is publicly available at https://***/wmalab/DiffGR.

Stagnant lid planets are characterized by a globe-encircling, conducting lid that is thick and strong, which leads to reduced global surface heat flows. Consequently, the mantles of such planets can have warmer interiors than Earth, and interestingly, a pyrolitic mantle composition under warmer conditions is predicted to have a distinctly different mantle transition zone compared to the present-day Earth(Hirose, 2002; Stixrude and Lithgow-Bertelloni, 2011; Ichikawa et al., 2014; Dannberg et al, 2022). Instead of olivine primarily transforming into its higher-pressure polymorphs such as wadsleyite and then ringwoodite, at pressures corresponding to410 km and 520 km depth in Earth, respectively, it instead transforms into a mineral assemblage of wadsleyite, majorite, and ferropericlase(WMF), and then to majorite + ferropericlase(MF), before finally transforming into bridgmanite at pressures corresponding to 660 km depth in Earth(Stixrude and Lithgow-Bertelloni, 2011; Ichikawa et al., 2014). Convective motions in stagnant lid planets are dominated by small-scale instabilities(cold drips) forming within the mobile rheological sublayer under the rigid lid. using ASPECT and a thermodynamic model of a pyrolitic mantle composition generated by HeFESTo, we show that under certain conditions, the small drips can pond atop the WMF-MF mineral phase transition. The barrier to convective flow arises from the WMF mineral phase assemblage having an effective negative thermal expansivity(Stixrude and Lithgow-Bertelloni, 2022). Although large-scale downwellings that typically occur within mobile lid planets are able to pass through the WMF zone without difficulty(Dannberg et al., 2022; Li RP et al.,2024), the smaller and less negatively buoyant nature of downwelling drips in stagnant lid planets are more susceptible to these effects,which leads to an ephemeral layering of the mantle. upon infecting a host cell,the reticulate body(RB)form of the Chlamydia bacteria simply proliferates by binary fission for an extended *** data show only RB units in the infected cells 20 hours post infection(hpi),spanning nearly half way through the development *** data collected every 4 hpi,conversion to the elementary body(EB)form begins abruptly at a rapid rate sometime around 24 *** modeling proliferation and conversion as simple birth and death processes,it has been shown that the optimal strategy for maximizing the total(mean)EB population at host cell lysis time is a bang-bang control qualitatively replicating the observed conversion ***,the simple birth and death model for the RB proliferation and conversion to EB deviates in a significant way from the available data on the evolution of the RB population after the onset of RB-to-EB *** working with a more refined model that takes into account a small size threshold eligibility requirement for conversion noted in the available data,we succeed in removing the deficiency of the previous models on the evolution of the RB population without affecting the optimal bang-bang conversion strategy.
