Identifying and Validating a Novel mirna-mrna Regulatory Network Associated with Prognosis in Lung Adenocarcinoma

Objective Many studies have revealed the crucial roles of mirna in multiple human cancers,including lung adenocarcinoma(LUAD).In this study,we sought to explore new mirna-mrna pairs that are associated with LUAD *** A novel mirna-mrna regulatory network associated with prognosis in LUAD was identified and validated using the bioinformatic tools including OncomiR database,StarBase,miRnet,GEPIA2,*** Twenty key mirnas were compiled after the analysis of the expression and prognostic value in OncomiR and *** mrnas of these key mirnas were predicted in miRnet,and the resulting mrnas were also analyzed for their prognostic values and expression patterns in GEPIA2 and UALCAN,*** expression correlation analysis was performed in ***,a new mirna-mrna network was built,of which each rna pair showed negative expression correlation,opposite expression pattern,and prognostic ***-protein interaction network was under construction for the mrnas,and 19 hub genes were *** analysis showed that“Cell Cycle,Mitotic”was the most significantly enriched ***,a mirna-hub gene sub-network was *** selected and validated the regulatory relationship of some mirna-hub pairs,including hsa-miR-1976/RFC2,hsa-let-7c-5p/RFC2,hsa-let-7c-5p/ESPL1,hsa-let-7c-5p/CDC25A,and hsa-miR-101-3p/***,over-expression of hsa-miR-1976 and hsa-let-7c-5p resulted in significant cell cycle *** Our results determined new prognosis-associated mirna-mrna pairs and might shed further light on the mechanism via which mirna-mrna network influences prognosis in LUAD.

鲍曼不动杆菌双组分系统bfmR基因缺失株构建及其生物学特性分析

为研究双组分系统bfmR基因对鲍曼不动杆菌生物学特性的影响,利用RecA同源重组技术构建鲍曼不动杆菌临床分离株AB43双组分系统bfmR基因缺失突变株并观察其生物学性状的变化,采用ExPASY-ProtParam、SMART、SOPMA和SWISS-MODEL等软件对bfmR进行生物信息学分析以及系统进化树构建。结果表明:与鲍曼不动杆菌野生株AB43相比,bfmR基因缺失突变株(AB43ΔbfmR)生长速度明显降低,细菌的自聚合能力和胞内超氧化物歧化酶(SOD)活性明显下降,生物被膜形成能力、黏附和入侵能力无显著性变化,而对头孢曲松的敏感性明显提高。生物信息学分析结果显示,bfmR蛋白为较稳定的亲水性蛋白,二级结构中α-螺旋含量最多,占总数的36.13%;鲍曼不动杆菌AB43株bfmR基因与标准株ATCC 17978和ATCC 19606的亲缘关系较近。综上,鲍曼不动杆菌AB43ΔbfmR的构建为进一步探讨双组分系统bfmR基因的调控机制提供试验基础。

Ticagrelor Protects against Sepsis-Induced Acute Kidney Injury through an Adenosine Receptor-Dependent Pathway

目的:本研究旨在探究超保守rna uc.102-uc.106基因簇及其宿主基因Obg样ATP酶1(OLA1)在C57BL/6J小鼠不同组织器官中的表达差异。方法:选取5只8~10周龄的健康雄性C57BL/6J小鼠,在正常生理条件下采集不同组织器官(包括肝脏、睾丸、骨髓、大脑、肾脏、血液、肺、结肠、胸腺、脾脏、胃、心脏、淋巴结和膀胱)。同时使用磁珠分选技术提取骨髓血细胞,包括B细胞、巨噬细胞、有核红细胞和成熟红细胞。通过RT-qPCR技术,以GAPDH作为内参照,检测uc.102-uc.106和OLA1 mrna的表达量。结果:OLA1 mrna在C57BL/6J小鼠大脑、淋巴结、睾丸和胸腺中呈高水平表达,在肝脏、脾脏、肺、结肠和外周血中呈低水平表达(Prna表达基本相反,但在睾丸组织中两者均呈高水平表达(P<0.05)。在血细胞中,OLA1和uc.102-uc.106在B细胞中的表达水平最高,在成熟红细胞中的表达水平最低(P<0.05)。结论:OLA1基因和uc.102-uc.106基因簇在C57BL/6J小鼠的不同组织器官及血细胞中均有表达且存在差异。值得注意的是,OLA1基因和uc.102-uc.106基因簇在小鼠睾丸中均呈高水平表达,表明OLA1基因和uc.102-uc.106基因簇可能与雄性小鼠的生殖功能密切相关。
