Evaluation of a user-friendly CSDS cage apparatus for studying depressive-like behaviors in rodents

Objective:To explore the efficacy and safety of Juan Bi Pill(JBP)in treatment of active rheumatoid arthritis(RA).Methods:From February 2017 to May 2018,115 participants from 4 centers were randomly divided into JBP group(57 cases)and placebo group(58 cases)in a 1:1 ratio using a random number table *** received a dose of JBP(4 g,twice a day,orally)combined with methotrexate(MTX,10 mg per week)or placebo(4 g,twice a day,orally)combined with MTX for 12 *** were required with follow-up visits at 24 and 48 weeks,attending 7 assessment *** were undergo disease activity assessment 7 times(at baseline and 2,4,8,12,24,48 weeks)and safety assessments 6 times(at baseline and 4,8,12,24,48 weeks).The primary endpoint was 28-joint Disease Activity Score(DAS28-ESR and DAS28-CRP).The secondary endpoints included American College of Rheumatology(ACR)criteria for 20%and 50%improvement(ACR20/50),Health Assessment Questionnaire Disability Index(HAQ-DI),clinical disease activity index(CDAI),visual analog scale(VAS),Short Form-36(SF-36)score,Medial Outcomes Study(MOS)sleep scale score,serum erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),tender joint count,swollen joint count,and morning *** adverse reactions were observed during the ***:After 12 weeks of treatment,DAS28-ESR and DAS28-CRP scores in both groups were lower than before treatment(both P<0.01),while the remission rate of DAS28-ESR and DAS28-CRP and low disease activity of JBP group were higher than those in the placebo group(both P<0.01).JBP demonstrated better efficacy on ACR20 and ACR50 compliance rate at 12 and 48 weeks comparing to placebo(all P<0.05).The CDAI and HAQ-DI score,pain VAS and global VAS change of RA patients and physicians,the serum ESR and CRP levels,and the number of tenderness and swelling joints were lower than before treatment at 4,8,12,24,48 weeks in both groups(P<0.05 or P<0.01),while the reduction of above indices in

Zhongfeng Xingnao Liquid ameliorates post-stroke cognitive impairment through sirtuin1(SIRT1)/nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase 1(HO-1) pathway

The activation of the sirtuin1(SIRT1)/nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase 1(HO-1) pathway has been shown to mitigate oxidative stress-induced apoptosis and mitochondrial damage by reducing reactive oxygen species(ROS) levels. Clinical trials have demonstrated that Zhongfeng Xingnao Liquid(ZFXN) ameliorates post-stroke cognitive impairment(PSCI). However, the underlying mechanism, particularly whether it involves protecting mitochondria and inhibiting apoptosis through the SIRT1/Nrf2/HO-1 pathway, remains unclear. This study employed an oxygen-glucose deprivation(OGD) cell model using SHSY5Y cells and induced PSCI in rats through modified bilateral carotid artery ligation(2VO).The effects of ZFXN on learning and memory, neuroprotective activity, mitochondrial function, oxidative stress, and the SIRT1/Nrf2/HO-1 pathway were evaluated both in vivo and in vitro. Results indicated that ZFXN significantly increased the B-cell lymphoma 2(Bcl2)/Bcl2-associated X(Bax) ratio, reduced terminal deoxynucleotidyl transferase-mediated d UTP nickend-labeling(TUNEL)+ cells, and markedly improved cognition, synaptic plasticity, and neuronal function in the hippocampus and cortex. Furthermore, ZFXN exhibited potent antioxidant activity, evidenced by decreased ROS and malondialdehyde(MDA) content and increased superoxide dismutase(SOD), catalase(CAT), and glutathione(GSH) levels. ZFXN also demonstrated considerable enhancement of mitochondrial membrane potential(MMP), Tom 20 fluorescence intensity, adenosine triphosphate(ATP) and energy charge(EC) levels, and mitochondrial complex Ⅰ and Ⅲ activity, thereby inhibiting mitochondrial damage. Additionally,ZFXN significantly increased SIRT1 activity and elevated SIRT1, nuclear Nrf2, and HO-1 ***, these effects were substantially counteracted when SIRT1 was suppressed by the inhibitor EX-527 in vitro. The paper is an introduction to the front-end pulse acquisition and the back-end pulse biomimetic reproduction system. This system is capable of faithfully replicating the complete pulse waveform collected at the front end. traditional Chinese medicine(TCM) practitioners analyze and diagnose the pulse patterns at the replication end. Meanwhile, the obtained pulse waveforms are analyzed and learnt by a neural network based on key diagnostic points in TCM pulse taking, which enables the determination of the corresponding relationships between different pulse waveforms and various pulse patterns in TCM pulse taking. With the support of clinical samples, an auxiliary diagnostic system for TCM pulse patterns ensures the accuracy of pulse pattern replication.

Curvularin derivatives from hydrothermal vent sediment fungus Penicillium ***-50 guided by molecular networking and their antiinflammatory activity

Guided by molecular networking, nine novel curvularin derivatives（1-9） and 16 known analogs（10-25） were isolated from the hydrothermal vent sediment fungus Penicillium sp. HL-50. Notably, compounds 5-7 represented a hybrid of curvularin and purine. The structures and absolute configurations of compounds 1-9 were elucidated via nuclear magnetic resonance（NMR） spectroscopy, X-ray diffraction, electronic circular dichroism（ECD） calculations,13C NMR calculation, modified Mosher's method, and chemical derivatization. Investigation of anti-inflammatory activities revealed that compounds 7-9, 11, 12, 14, 15, and 18 exhibited significant suppressive effects against lipopolysaccharide（LPS）-induced nitric oxide（NO）production in murine macrophage RAW264.7 cells, with IC50values ranging from 0.44 to 4.40μmol·L-1. Furthermore, these bioactive compounds were found to suppress the expression of inflammation-related proteins, including inducible NO synthase（i NOS）, cyclooxygenase-2（COX-2）, NLR family pyrin domain-containing protein 3（NLRP3）, and nuclear factor kappa-B（NF-κB）. Additional studies demonstrated that the novel compound 7 possessed potent antiinflammatory activity by inhibiting the transcription of inflammation-related genes, downregulating the expression of inflammation-related proteins, and inhibiting the release of inflammatory cytokines, indicating its potential application in the treatment of inflammatory diseases.

Effect of add-on therapy with traditional Chinese medicine on the survival of patients with anemic lower-risk myelodysplastic syndromes in the real-world setting:a retrospective study

OBJECTIVE: To illustrate the effect of add-on therapy of traditional Chinese medicine(TCM) on the long-term survival of anemic lower-risk myelodysplastic syndromes(LR-MDS) patients. METHODS: In this study, we conducted a retrospective analysis of the data of patients with LR-MDS who were identified from Shuguang Hospital between January 2006 and June 2020. The Kaplan-Meier method was used to calculate overall survival(OS), while the Cox model was employed to analyze prognostic factors of long-term survival. A total of 162 patients who met the inclusion criteria were divided into two groups: the TCM group(n = 78) or the non-TCM group(n = 84). RESULTS: There were no significant differences in baseline characteristics. The cumulative survival rate of the TCM group was significantly higher than that of the non-TCM group [log-rank P = 0.029, hazard ratio(HR) = 0.484, 95% confidence interval(CI)(0.249, 0.942), P medicine may be a protective factor affecting the long-term survival of LR-MDS patients [HR = 0.509, 95% CI,(0.261, 0.993), P = 0.048]. The hematologic improvement erythroid response(HI-E) rate of the TCM group was significantly higher than that of the non-TCM group(69.23% vs 52.38%; P = 0.028). However, the duration of HI-E and cases of disease progression after HI-E did not significantly differ from the non-TCM group(P > 0.05). CONCLUSIONS: The integration of TCM and Western medicine has the potential to extend the OS of LR-MDS patients when compared to the use of Western medicine alone.
