Study on the Mechanism and Molecular Docking Verification of Buyang Huanwu Decoction in Treating Diabetic Foot

Objective:The objective of this study was to investigate the molecular mechanism of Buyang Huanwu decoction(BYHWD)in the treatment of diabetic foot(DF).Methods:The TCMSP,BATMAN,PubChem,PharmMapper,UniProt,GeneCards,Webgestalt,and Kobas databases were used to obtain the structures,targets,main biological functions,and pathways of the active ingredients of BYHWD,and the results were visualized using Cytoscape3.6.1,Ledock,and PyMol ***:A total of 82 active components of BYHWD and 193 targets related to BYHWD were identified,and 5295 genes related to DF were identified using the GeneCards database,including 65 key targets of BYHWD in the treatment of *** and KEGG enrichment analyses of the 65 targets for BYHWD treatment of DF showed that 47 GO items were involved in the *** was mainly involved in biological processes,such as biological regulation,metabolism,and stress *** is primarily involved in protein binding,ion binding,nucleotide binding,and other molecular *** is mainly involved in membrane encapsulation,membrane lumen closure,and other biological components and involved in the VEGF,TNF,RAS,RAP1,PI3K-AKT,MAPK,and IL-17 signaling *** targets were enriched in the PI3K-AKT and MAPK signaling *** docking results showed that the 59 key active components of BYHWD had strong binding activity with 64 key DF ***:The therapeutic effect of BYHWD on DF is based on the pharmacological effects of multiple targets and pathways.

Determination of five neurotransmitters in the rat brain for the study of the hypnotic effects of Ziziphi Spinosae Semen aqueous extract on insomnia rat model by UPLC-MS/MS

Ziziphi Spinosae Semen(ZSS) has been used for treatment of insomnia in China for centuries. To reveal the influence of insomnia on the levels of the neurotransmitters including serotonin(5-HT), glutamic acid(Glu),γ-aminobutyric acid(GABA),noradrenaline(NE) and dopamine(DA), and to study the role of ZSS aqueous extract in the treatment of insomnia, an UPLC-ESIMS/MS method was developed and validated for simultaneous determination of five neurotransmitters in the rat brain. The brain samples were pretreated by one-step direct protein precipitation with acetonitrile. The analytes were detected in positive mode with multiple reaction monitoring(MRM) and the procedure was completed in less than 10 min. The method showed a good linearity(R^2 >0.9967) with the other validation parameters were within acceptance range. The results indicated that the concentration of 5-HT, GABA and DA is significantly lower(P treatment with ZSS aqueous extract(4 or 8 g·kg^1·d^-1 for seven days) could ameliorate the symptoms of insomnia by significantly changing the levels of the neurotransmitter parameters mentioned above. The data obtained in this study demonstrate that ZSS aqueous extract could ameliorate the symptoms of insomnia by modulating the levels of monoamines and amino acid neurotransmitters in the brain.

Ge Gen Decoction(GGD),a traditional chinese medicine prescription,is mainly used to treat infectious respiratory diseases and can relieve the symptoms of influenza A virus(IAV)***,the underlying mechanism of GGD against IAV infection remains *** this study,we found that GGD had moderate anti-IAV activity in *** was more effective when given before the viral infection and targeted the viral attachment and replication stages rather than the internalization sta *** vivo,GGD treatment reduced the virus titers of lung tissue significantly and improved the survival rate,lung index,and pulmonary hist opathological changes in H1N1-infected *** observed the changes in several key immuno-related indexes in GGD administrated H1N1-infected mice with anti-IAV drug oseltamivir phosphate as the *** treatment decreased the expression of TNF-αand improved Th1/Th2 immune balance to reduce the excessive immune response in H1N1-infected ***,the expression of the toll-like receptor 7 signaling pathway in H1N1-infected mice decreased after GGD *** results showed that GGD has anti-IAV activity and can modulate the immune system to relieve lung inflammation.

Protective mechanism of Coprinus comatus polysaccharide on acute alcoholic liver injury in mice,the metabolomics and gut microbiota investigation

Coprinus comatus polysaccharide(CCP)has significant hepatoprotective *** explore hepatoprotective mechanism of CCP,the study analyzed preventive effect of CCP on acute alcoholic liver injury in mice by histopathological examination and biochemical ***,hepatoprotective mechanism was also analyzed in conjunction with metabolomics and proliferation of gut *** results showed that CCP significantly decreased alanine aminotransferase(ALT),aspartate aminotransferase(AST)and triglyceride(TG)levels in serum of alcoholic liver disease(ALD)*** examination showed that CCP can significantly improve liver *** results showed that there were significant differences in the level of metabolites in liver tissue of control group,ALD group and CCP group,including taurine,xanthosine,fumaric acid and arachidonic acid,among *** pathways analysis showed that hepatoprotective effect of CCP was related to energy metabolism,biosynthesis of unsaturated fatty acids,amino acids metabolism and lipid ***,CCP inhibited an increase in the number of Clostridium perfringens,Enterobacteriaceae and Enterococcus,and a decrease in the number of Lactobacillus and Bifidobacterium in the gut of ALD *** these findings suggested that CCP treatment reversed the phenotype of ethanol-induced liver injury and the associated metabolites pathways.

Effect of Yanggan Jiedu Sanjie formula on human hepatocellular carcinoma Bel-7402 cells

OBJECTIVE: To observe the effect of Yanggan Jiedu Sanjie(YGJDSJ) formula on human hepatocellular carcinoma Bel-7402 ***: Bel-7402 cells were treated with YGJDSJ. Cell proliferation was detected by cell counting kit-8 assay. Cell apoptosis was identified by Hoechst 33258 staining and flow cytometric analysis. Cell cycle distribution was quantified by flow cytometric analysis. Caspase activities were measured by commercial kit. Cell senescence was detected by senescence-associated β-galactosidase(SA-β-gal)staining. Protein expression and phosphorylation were identified by Western blot. Protein expression was knocked-down by ***: YGJDSJ inhibited proliferation of Bel-7402 cells in a dose-and time-dependent *** induced apoptosis and activated caspase-3, 8, and 9 in Bel-7402 cells. YGJDSJ-induced apoptosis was completely abrogated by a pan caspase inhibitor, Z-VAD-FMK. YGJDSJ also induced cell senescence, up-regulated cyclin-dependent kinase inhibitor 1 a(CDKN1 a) and CDKN2 a expression and down-regulated retinoblastoma protein(RB) phosphorylation in Bel-7402 cells. Specific knockdown of CDKN1 a and CDKN2 a significantly reduced YGJDSJ-induce cell senescence in Bel-7402 ***: YGJDSJ inhibited cell proliferation,induced caspase-dependent apoptosis and CDKN1 a/CDKN2 a-RB signalling mediated cell senescence in Bel-7402 cells. Our findings suggest that YGJDSJ might be potential for hepatocellular carcinoma treatment.

Bloodletting puncture in the treatment of acute ischemic stroke:protocol for a mixed-method study of a multi-center randomized controlled trial and focus group

Microanalysis of Multi-Element in Juncus effusus L. by LIBS Technique

Laser-induced breakdown spectroscopy （LIBS） was used to decipher the unique multi-elemental characteristics of Juncus effusus L. The spectral fingerprints of Juncus effusus L. were established based on elemental microanalysis via LIBS. Microanalysis and multimode sampling methodologies were designed in this study. The relative standard deviation （RSD） approach was performed to optimize the multi-shot measurements. Taking advantage of the capability with no or minimal sample pre-treatment of LIBS, a thermodynamic chart of four elements （Mg, Ca, Ba, and Na） was created from twelve collection regions. The diagram of elemental distribution on a micro-scale was generated to explore the nature of Juncus effusus L. by LIBS. The results demon- strated that LIBS is a promising technique for rapid elemental microanalysis of heterogeneous samples.
