Inhibition of human cytochrome CYP1B1 by anthraquinone compounds,chrysophanol and physcion

The in vitro inhibitory effects of chrysophanol and physcion on CYP1B1 were explored,utilizing ethoxyresorufin as the *** inhibition kinetics of CYP1B1 by these compounds were assessed with escalating doses of *** chrysophanol(IC_(50)(0.47±0.01)μmol·L^(-1))and physcion(IC_(50)(0.35±0.02)μmol·L^(-1))significantly reduce the catalytic efficiency of *** V_(max)and K_(m)values are determined to be(51.9912±10.0547)pmol·μg^(-1)(protein)·min^(-1) and(0.9663±0.2987)nmol·L^(-1)for chrysophanol,and(45.4227±1.9978)pmol·μg^(-1)(protein)·min^(-1) and(0.4367±0.0386)nmol·L^(-1)for physcion,*** analysis reveals that chrysophanol and physcion exert mixed inhibitory effects on *** mixed inhibition is primarily characterized by the compounds’ability to competitively bind to the active sites of CYP1B1,as well as potentially through non-competitive mechanisms,thereby reducing the enzyme’s catalytic *** docking studies are conducted to elucidate the interaction between anthraquinone derivatives and CYP1B1,indicating that these compounds may inhibit CYP1B1 activity by binding to their active *** demonstrated capacity of chrysophanol and physcion to inhibit CYP1B1 enzymatic function unveils a potential anticancer mechanism,advancing our comprehension of how the structure of anthraquinone derivatives correlates with CYP1B1 inhibition and paving the way for developing innovative cancer treatments.

Objective To analyze the clinical application of manual therapy(MT)to tumor-related adverse reactions via summarizing the research at home and abroad,in order to provide more theoretical evidence for the clinical promotion of *** We searched 7 chinese and English databases,including China National Knowledge Infrastructure(CNKI),Wanfang Academic Journal Full-text Database(Wanfang),Chongqing VIP Database(CQVIP),PubMed,Excerpta Medica Database(EMBASE),Ovid and *** publication date was between the establishment date of the database and December 31,*** screened the literature according to the inclusion and exclusion criteria,and then sorted and analyzed the selected *** A total of 46 papers were *** studies focused on the adverse reactions in breast cancer *** interventions demonstrated the best efficacy for fatigue,followed by pain,depression and *** different MT interventions,Tuina(chinese therapeutic massage)was mainly adopted for fatigue,pain,anxiety,depression,and limb *** pressing was mainly adopted for gastrointestinal and psychological problems such as abdominal bloating,insomnia,depression and *** application of reflexotherapy was similar to that of *** MT can alleviate various adverse reactions by effectively relieving patients’somatic symptoms and improving their psychological states and overall *** can be popularized as a significant non-drug ***,however,the clinical application of MT is neither extensive nor has sufficient basic ***,we should attach importance to this application.

Objective:To investigate the protective effects and its possible mechanism of Wuzi Yanzong Pill(WYP) on Parkinson’s disease(PD) model ***:Thirty-six C57BL/6 male mice were randomly assigned to 3 groups including normal,PD,and PD+WYP groups,12 mice in each *** week of intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) was used to establish the classical PD model in ***,mice in the PD+WYP group were administrated with 16 g/kg WYP,twice daily by *** 14 days of administration,gait test,open field test and pole test were measured to evaluate the movement *** hydroxylase(TH) neurons in substantia nigra of midbrain and binding immunoglobulin heavy chain protein(GRP78) in striatum and cortex were observed by *** levels of TH,GRP78,p-PERK,p-elF2α,ATF4,p-IRE1α,XBP1,ATF6,CHOP,ASK1,p-JNK,Caspase-12,-9 and-3 in brain were detected by Western ***:Compared with the PD group,WYP treatment ameliorated gait balance ability in PD mice(P<0.05).Similarly,WYP increased the total distance and average speed(P<0.05or P<0.01),reduced rest time and pole time(P<0.05).Moreover,WYP significantly increased TH positive cells(P<0.01).Immunofluorescence showed WYP attenuated the levels of GRP78 in striatum and ***,WYP treatment significantly decreased the protein expressions of GRP78,p-PERK,p-elF2α,ATF4,p-IRE1α,XBP1,CHOP,Caspase-12 and Caspase-9(P<0.05or P<0.01).Conclusions:WYP ameliorated motor symptoms and pathological lesion of PD mice,which may be related to the regulation of unfolded protein response-mediated signaling pathway and inhibiting the endoplasmic reticulum stress-mediated neuronal apoptosis pathway.

BACKGROUND Post-transarterial chemoembolization(TACE)liver failure occurs frequently in hepatocellular carcinoma(HCC)*** identification of predictors for post-TACE liver failure is of great importance for clinical decision-making in this *** To investigate the occurrence rate and predictive factors of post-TACE liver failure in this retrospective study to provide clues for decision-making regarding TACE procedures in HCC *** The clinical records of HCC patients treated with TACE therapy were *** clinical characteristics and laboratory parameters of these patients were *** models were used to identify candidates to predict post-TACE liver *** A total of 199 HCC patients were enrolled in this study,and 70 patients(35.2%)developed post-TACE liver *** and multivariate logistic models indicated that microspheres plus gelatin embolization and main tumor size>5 cm were risk predictors for post-TACE liver failure[odds ratio(OR):4.4,95%confidence interval(CI):1.2-16.3,P=0.027;OR:2.3,95%CI:1.05-5.3,P=0.039,respectively].Conversely,HCC patients who underwent tumor resection surgery before the TACE procedure had a lower risk for post-TACE liver failure(OR:0.4,95%CI:0.2-0.95,P=0.039).CONCLUSION Microspheres plus gelatin embolization and main tumor size might be risk factors for post-TACE liver failure in HCC patients,while prior tumor resection could be a favorable factor reducing the risk of post-TACE liver failure.
