Current Tracking on Effectiveness and Mechanisms of Acupuncture Therapy: A Literature Review of High-Quality Studies

Effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on chemotactic migration of alveolar macrophages and inters regions macrophages in rats

OBJECTIVE: To observe the effects of serum containing Mahuang(Herba Ephedra Sinica) or Wuweizi(Fructus Schisandrae Chinensis) on the migration of alveolar macrophages(AM) and interstitial macrophages(IM) from normal rats, and to analyze and compare the mechanisms leading to cell migration ***: Rats were randomly divided into three groups: Mahuang(Herba Ephedra Sinica), Wuweizi(Fructus Schisandrae Chinensis), and blank serum. After treatment with the herbs, serum was extracted from the rats. AM and IM were isolated from normal rats and cultured. The effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on normal rat AM and IM chemotactic migration were determined by transwell assays. The CC chemokine receptor(CCR) 2, CCR5, voltage-gated Kvl. 3 K^+channel(Kv1. 3), and voltage-gated Kvl. 5 K^+channel(Kv1. 5) protein levels were analyzed by western ***: The migration quantities of AM and IM in the Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum groups were significantly higher than those in the blank serum group(P < 0.01). Compared with the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group, the migration quantity of cultured rat AM in the Mahuang(Herba Ephedra Sinica) medicated serum group was significantly increased(P <0.01). Meanwhile, compared with the Mahuang(Herba Ephedra Sinica) medicated serum group, the migration quantity of cultured rat IM in the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group was significantly increased(P < 0.01).CCR2, CCR5, Kv1. 3, and Kv1. 5 proteins were expressed on the AM cell surface, and showed significantly higher expression in the Mahuang(Herba Ephedra Sinica) medicated serum group compared with the Wuweizi(Fructus Schisandrae Chinensis)medicated serum group. Effect of Guizhi Fuling Capsule on Apoptosis of Myeloma Cells Through Mitochondrial Apoptosis Pathway

Objective: To observe the effects of Guizhi Fuling Capsule(GZFLC) on myeloma cells and explore the mechanisms. Methods: MM1S and RPMI 8226 cells were co-cultured with different concentrations of serum and the cell experiments were divided into negative(10%, 20% and 40%) groups, GZFLC(10%, 20%, and 40%)groups and a control group. Cell counting kit-8(CCK-8) assays and flow cytometry were used to detect the viability and apoptosis levels of myeloma cells. The effects on mitochondria were examined by reactive oxygen specie(ROS) and tetrechloro-tetraethylbenzimidazol carbocyanine iodide(JC-1) assays. Western blot was used to detect the expression of B cell lymphoma-2(Bcl-2), Bcl-2-associated X(Bax), cleaved caspase-3,-9, cytochrome C(Cytc)and apoptotic protease-activating factor 1(Apaf-1). RPMI 8226 cells(2×107) were subcutaneously inoculated into 48 nude mice to study the in vivo antitumor effects of GZFLC. The mice were randomly divided into four groups using a completely randomized design, the high-, medium-, or low-dose GZFLC(840, 420, or 210 mg/kg per day,respectively) or an equal volume of distilled water, administered daily for 15 days. The tumor volume changes in and survival times of the mice in the GZFLC-administered groups and a control group were observed. Cytc and Apaf-1 expression levels were detected by immunohistochemistry. Results: GZFLC drug serum decreased the viability and increased the apoptosis of myeloam cells(P<0.05). In addition, this drug increased the ROS levels and decreased the mitochondrial membrane potential(P<0.01). Western blot showed that the Bcl-2/Bax ratios were decreased in the GZFLC drug serum-treated groups, whereas the expression levels of cleaved caspase-3,-9,Cytc and Apaf-1 were increased(all P<0.01). Over time, the myeloma tumor volumes of the mice in the GZFLCadministered groups decreased, and survival time of the mice in the GZFLC-administered groups were longer than that of the mice in the control group. Systematic review and Meta-analysis of acupuncture and acupoint catgut embedding for the treatment of abdominal obesity

OBJECTIVE:To systematically review and analyze the effect of acupuncture and acupoint catgut embedding in treatment of abdominal obesity to provide a more reasonable clinical treatment ***:Ten databases were searched as of August 2022:the English databases Pub Med,Embase,Cochrane Library,Web of Science,Wiley,and Scopus and the chinese databases China National Knowledge Infrastructure Database,China Science and Technology Journal Database,Wanfang,and Sino Med/chinese Biomedical Literature *** controlled trials(RCTs)of acupuncture and acupoint catgut embedding as the main interventions to treat abdominal obesity were *** investigators imported the citations into End Note version X9.1 for deduplication,screening,extraction,and *** risk of bias in the included RCTs was assessed according to the Cochrane *** Man 5.4 software was used to conduct a Meta-analysis of RCTs that met the inclusion ***:Thirteen RCTs(1069 patients)were included in this study,and the data of eleven RCTs(966 patients)were include in the *** results showed that acupoint catgut embedding can significantly change the weight and waist circumference of patients with abdominal obesity when compared to sham acupuncture or no treatment[mean difference(MD)=2.32,95%confidence interval(CI)(1.88,2.76),P<0.00001],[MD=3.47,95%CI(1.99,4.94),P<0.00001].The change in hip circumference after acupuncture was also significant[MD=0.89,95%CI(0.12,1.66),P=0.02].CONCLUSION:This study found that acupuncture and acupoint catgut embedding can effectively treat abdominal obesity,therefore,these interventions can be used as clinical supplements and alternative *** diagnostic criteria of the existing studies and the intervention measures of the control group are not *** will be necessary to improve the clinical study protocols and expand the sample size to further validate the reliability of the results obtained of this study.

Jixuecao(Herba Centellae Asiaticae) alleviates mesangial cell proliferation in IgA nephropathy by inducing mitofusin 2 expression

Icariin protects vascular endothelial cells from oxidative stress through inhibiting endoplasmic reticulum stress

Objective: To investigate the protective effect and underlying mechanism(s) of icariin(ICA) in preventing hydrogen peroxide(H_2O_2)-induced vascular endothelial cell injury via endoplasmic reticulum stress(ERS).Methods: To study the effects of ICA on H_2O_2-induced damage, we used the cell counting kit-8 assay to detect cell viability and the terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling assay to determine cell adhesion and apoptosis, respectively. Spectrophotometry and enzyme-linked immunosorbent assay were used to measure the expression levels of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px). Subsequently, glucose-regulated protein 78(GRP78), activating transcription factor-4(ATF4) and eukaryotic initiation factor-2 a(eIF2 a) were detected using Western ***: In human umbilical vein endothelial cells, different concentrations of ICA exhibited multiple effects, including reduced H_2O_2 damage, improved cell viability and adhesion, reduced cell apoptosis and increased SOD and GSH-Px activity. Among the ICA concentrations used, only the H_2O_2+ 100 lmol/L ICA group had significant differences compared to the H_2O_2 group. ERS activators H_2O_2 and DL-dithiothreitol(DTT) significantly increased GRP78, ATF4 and eIF2 a expressions, decreased cell activity and reduced SOD and GSH-Px activity. In contrast, the H_2O_2+ 100 lmol/L ICA and H_2O_2+ 100 lmol/L ICA + DTT groups had significant inhibitory effects on the expressions of GRP78,ATF4 and eIF2 a proteins, showing enhanced cell viability and SOD and GSH-Px ***: The results showed the dose-dependent effects of ICA against H_2O_2-induced injury in vascular endothelial cells. The inhibition of GRP78, ATF4 and eIF2 a protein expressions in the ERS, and the subsequent alleviation of oxidative stress damage, might be the molecular mechanism.

Effect of Acupuncture on the Notch Signaling Pathway in Rats with Brain Injury

Objective： To observe the effect of acupuncture on the Notch signaling pathway in rats with traumatic brain injury and to explore the pathogenesis of acupuncture intervention on traumatic brain injury. Methods： Feeney＇s freefall epidural impact method was used to establish a traumatic brain injury model in rats; the rats were randomly divided into a normal group, sham operation group, model group and acupuncture group. Acupuncture was performed in the Baihui（DU 20）, Shuigou（DU 26）, Fengfu（DU 16）, Yamen（DU 15） and Hegu（LI 4） acupoints in the rat, and Yamen was punctured via Fengfu. Then, the rats in each group were randomly divided into three subgroups, namely the day 3 subgroup, day 7 subgroup and day 14 subgroup according to treatment duration. The modified neurological severity scores（m Nss） method was used to perform neurobehavioral scoring for evaluating the degree of injury in the rats. The hematoxylin-eosin（HE） staining method was used to observe the pathological change in the brain tissue of rats in each group. Real-time fluorescent quantitative polymerase chain reaction（Q-PCR） technology was used to detect changes in the Notch1, Hes1 and Hes5 gene expression levels in the cortex on the injured side. Western blot was used to detect the protein expression changes. Results： One day after modeling, the m Nss scores in the model group and in the acupuncture group were significantly higher than those in the normal and sham operation groups（P〈0.01）; there was no statistically significant difference between the normal group and the sham operation group.
