Role of hypothalamus nociceptin/orphanin FQ in pre-ovulatory luteinizing hormone surge of estrogen and progesterone-primed,ovariectomized rats

Aim:To investigate the role of hypothalamus nociceptin/orphanin FQ（OFQ）andits endogenous receptor,the opioid receptor-likel receptor（ORL1 receptor）in theestrus cycle of female ***:Radioimmunoassay was used to detect theeffect of the intracerebroventricular（icy）administration of OFQ and/or the ORL1receptor antagonist[Nphe1]Nociceptin（1-13）NH2,that is,NC13 on luteinizinghormone（LH）levels of estrogen- and progesterone（EBP）-primed,ovariectomized（OVX）rats（EBP-primed OVX rats）.RT-PCR,Western blotting,and immunohis-tochemistry techniques were adopted to observe the changes of OFQ and theORL1 receptor in the pre-optic area（POA）and the medial basal hypothalamus（MBH）of the estrus cycle of female ***:Pre-ovulatory LH surges in EBP-primed,OVX rats were significantly reduced by icv administration of 20 and 200nmol OFQ（P<0.05）,and the effect of 20 nmol OFQ could be abolished by pretreat-merit with 20 nmol *** OFQ mRNA level in the POA on pro-estrus waslowered markedly compared to diestrus and estrus（P<0.05）,while the mRNA andprotein levels of the ORL1 receptor showed no significant changes in the POAand MBH across the estrus ***,the number of OFQ-immunoreac-tive neurons in the medial POA,ventromedial hypothalamus,and the arcuatenucleus on pro-estrus was significantly decreased compared to diestrus and es-tins（P<0.05）.Conclusion:The inhibitory effect of OFQ on the LH surge of EBP-primed,OVX rats and its downregulation in POA and MBH on pro-estrus sug-gests that it might play a negative modulatory role in the estrus cycle.

It includes those syndromes recorded in the literature of traditional chinese medicine such as 'sensation of gas rushing', 'foreign body sensation in throat', 'syncope' and 'hysteria', and those in modern medicine such as 'anxiety neurosis' and 'depression'.

Clinical study on Tuina plus physical agents for lateral collateral ligament injury of ankle in gymnasts

Objective To compare the efficacy of Tuina(chinese therapeutic massage)plus physical agents and physical agents alone for lateral collateral ligament injury of ankle in gymnasts,and to explore the feasibility of Tuina for injury intervention in competitive *** A total of 64 gymnasts with types Ⅰ-Ⅱ lateral collateral ligament injury of ankle were selected and divided into a control group and an observation group according to a full analysis set based on the intention-to-treat principle,with 32 cases in each *** in the control group received ultrasound and microwave treatment,while those in the observation group received additional Tuina *** efficacy was evaluated by total effective rate,visual analog scale(VAS)score,and American Orthopedic Foot and Ankle Society ankle-hindfoot scale(AOFAS-AHS)*** The total effective rate was 96.9% in the observation group and 90.6% in the control *** was no statistical difference in the total effective rate between the two groups(P>0.05).The markedly effective rate was 75.0% in the observation group and 46.9% in the control *** markedly effective rate in the observation group was higher than that in the control group(P0.05).After three months of treatment,the AOFAS-AHS score in the observation group was higher than that in the control group,indicating statistical significance(P<0.05).There was an interaction between time and group(P<0.05).Con

Objective:To explore the anti-inflammatory effects of ethyl lithospermate in lipopolysaccharide(LPS)-stimulated RAW 264.7 murine-derived macrophages and zebrafish,and its underlying ***:3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide(MTT)assays were performed to investigate the toxicity of ethyl lithospermate at different concentrations(12.5–100μmol/L)in RAW 264.7 *** cells were stimulated with LPS(100 ng/mL)for 12 h to establish an inflammation model in vitro,the production of proinflammatory cytokines interleukin(IL)-6 and tumor necrosis factorα(TNF-α)were assessed by enzyme linked immunosorbent assay(ELISA).Western blot was used to ascertain the protein expressions of signal transducer and activator of transcription 3(STAT3),nuclear factor kappa B(NF-κB)p65,phospho-STAT3(p-STAT3,Tyr705),inhibitor of NF-κB(IκB)α,and phospho-IκBα(p-IκBα,Ser32),and confocal imaging was used to identify the nuclear translocation of NF-κB p65 and p-STAT3(Tyr705).Additionally,the yolk sacs of zebrafish(3 days post fertilization)were injected with 2 nL LPS(0.5 mg/mL)to induce an inflammation model in *** analysis,hematoxylin-eosin(HE)staining,observation of neutrophil migration,and quantitative real-time polymerase chain reaction(qR T-PCR)were used to further study the anti-inflammatory effects of ethyl lithospermate and its probable mechanisms in ***:The non-toxic concentrations of ethyl lithospermate have been found to range from 12.5 to 100μmol/*** lithospermate inhibited the release of IL-6 and TNF-α(P<0.05 or P<0.01),decreased IκBαdegradation and phosphorylation(P<0.05)as well as the nuclear translocation of NF-κB p65 and p-STAT3(Tyr705)in LPS-induced RAW 264.7 cells(P<0.01).Ethyl lithospermate also decreased inflammatory cells infiltration and neutrophil migration while increasing the survival rate of LPS-stimulated zebrafish(P<0.05 or P<0.01).In addition,ethyl lithospermate also inhibited the mR NA expression levels of of IL
