Quantitative Analysis of Myocaridal Perfusion in Rabbits by Tansthoracic Real-time Myocardial Contrast Echocardiography

To evaluate the feasibility of real-time myocardial contrast echocardiography （RTMCE） by quantitative analysis of myocardial perfusion in rabbits, transthoracic RTMCE was performed in 10 healthy rabbits by using continuous infusion of SonoVue into the auricular vein. The short axis view at the papillary muscle level was obtained. The duration of the time that the contrast took to appear in right heart, left heart and myocardium was recorded. The regional myocardial signal intensity （SI） versus re-filling time plots were fitted to an exponential function： y（t） =A（1–e–β（t–t0））＋ C, where y is SI at any given time, A is the SI plateau that reflects myocardial blood volume, and β is the slope of the refilling curve that reflects myocardial microbubble velocity. The A, β and A×β values at different infusion rate of SonoVue were analyzed and the A, β and A×β values in each segment in the short axis view at the papillary muscle level were compared. All the animal experiments were successful and high-quality im-ages were obtained. The best intravenous infusion rate for SonoVue was 30 mL/h. The contrast appeared in right heart, left heart and myocardium at 7.5±2.2 s, 9.1±2.4 s and 12.2±1.6 s respectively. After 16.6±2.3s, myocardial opacification reached a steady state. The mean A, β and A×β value in the short axis view at the papillary muscle level were 9.8±3.0 dB, 1.4±0.5 s-1 and 13.5±3.6 dB×s-1 respectively. A, β and A×β values showed no significant differences among 6 segments. It was suggested that RTMCE was feasible for quantitative analysis of myocardial perfusion in rabbits. It provides a non-invasive method to evaluate the myocardial perfusion in rabbit disease models.

Sensitive DNA Mutation Detection at Physiological Temperature with Endonuclease IV by Inhibiting Its Side Activity

Endonuclease IV(Endo IV)has the side activity to cleave the apurinic/apyrimidinic site in the single-stranded DNA probe(AP-ssDNA),which adversely affects its performance in single-nucleotide variation(SNV)*** this work,we developed a simple strategy to inhibit Endo IV's side activity by introducing an assistant strand into the detection *** assistant strand effectively inhibited the side-activity of Endo IV by conf ini ng the freedom of AP-ssDNA through rigid double-stranded DNA formati on,and the inhibition efficiency could reach 98%.About 20 times enhancement of the discrimination factor(DF)was obtained compared to the detection system without the assistant *** optimization,a mean DF value was calculated to be 738 for different mutation types,and the sample with 0.005%allele frequency was discriminated from the wild-type *** advantage of using an assists nt stra nd was that the SNV detection could be executed at physiological temperature without precise temperature optimization as in other Endo IV-based SNV detection ***,EGFR T790M mutant in the synthesized and clinical samples was detected by our *** results showed satisfactory sensitivity and accuracy by comparison with Sanger ***,this strategy has the potential to be applied in the field of precision medicine.

Therapeutic effect and mechanism of breviscapine on cisplatin-induced nephrotoxicity in mice

Volatile anesthetics inhibit the activity of calmodulin by interacting with its hydrophobic site

Background Volatile anesthetics （VAs） may affect varied and complex physiology processes by manipulating Ca2＋-calmodulin （CAM）. However, the detailed mechanism about the action of VAs on CaM has not been elucidated. This study was undertaken to examine the effects of VAs on the conformational change, hydrophobic site, and downstream signaling pathway of CaM, to explore the possible mechanism of anesthetic action of VAs. Methods Real-time second-harmonic generation （SHG） was performed to monitor the conformational change of CaM in the presence of VAs, each plus 100 μmol/L Ca2＋. A hydrophobic fluorescence indicator, 8-anilinonaphthalene-l-sulfonate （ANS）, was utilized to define whether the VAs would interact with CaM at the hydrophobic site or not. High-performance liquid chromatography （HPLC） was carried out to analyze the activity of CaM-dependent phosphodiesterase （PDE1） in the presence of VAs. The VAs studied were ether, enflurane, isoflurane, and sevoflurane, with their aqueous concentrations 7.6, 9.5, 11.4 mmol/L; 0.42, 0.52, 0.62 mmol/L; 0.25, 0.31, 0.37 mmol/L and 0.47, 0.59, 0.71 mmol/L respectively, each were equivalent to their 0.8, 1.0 and 1.2 concentration for 50% of maximal effect （EC50） for general anesthesia. Results The second-harmonic radiation of CaM in the presence of Ca2＋ was largely inhibited by the VAs. The fluorescence intensity of ANS, generated by binding of Ca2＋ to CaM, was reversed by the VAs. HPLC results also showed that AMP, the product of the hydrolysis of cAMP by CaM-dependent PDE1, was reduced by the VAs. Conclusions Our findings demonstrate that the above VAs interact with the hydrophobic core of Ca2＋-CaM and the interaction results in the inhibition of the conformational change and activity of CaM. This in vitro study may provide us insight into the possible mechanism of anesthetic action of VAs in vivo.

Prolonged Alzheimer-like Tau Hyperphosphorylation Induced by Simultaneous Inhibition of Phosphoinositol-3 Kinase and Protein Kinase C in N2a cells

Co-injection of wortmannin（inhibitor of phosphatidylinositol-3 kinase,PI3K）and GF109203X（inhibitor of protein kinase C,PKC）into the rat brain was found to induce spatial memory deficiency andenhance tau hyperphosphorylation in the hippocampus of rat *** establish a cell model with durativeAlzheimer-like tau hyperphosphorylation in this study,we treated N2a neuroblastoma cells with wortmanninand GF109203X separately and simultaneously,and measured the glycogen synthase kinase 3（GSK-3）activity by γ-（32）P-labeling and the level of tau phosphorylation by Western *** was found that theapplication of wortmannin alone only transitorily increased the activity of GSK-3（about 1 h）and the level oftau hyperphosphorylation at Ser396/Ser404 and Ser199/Ser202 sites（no longer than 3 h）;however,a prolongedand intense activation of GSK-3（over 12 h）and enhanced tan hyperphosphorylation（about 24 h）wereobserved when these two selective kinase inhibitors were applied *** conclude that the simultaneousinhibition of PI3K and PKC can induce GSK-3 overactivation,and further strengthen and prolong the Alzheimer-like tau hyperphosphorylation in N2a cells,suggesting the establishment of a cell model with early pathologicalevents of Alzheimer’s disease.

Glioblastoma(GBM)is one of the malignant brain tumors with high mortality and no curative *** elevated vascular endothelial growth factor(VEGF)in GBM seriously disrupts the blood brain barrier(BBB)with an increased permeability,resulting in poor outcome and *** interference has shown strong potential to inhibit VEGF expression,thus it is necessary to development an effective and safe gene delivery system possessing the ability to cross the BBB and target GBM *** study aims to explore the anti-GBM effect of angiopep-2(Ap)peptide modified reactive oxygen species(ROS)cleavable thioketal(TK)linked glycolipid-like nanocarrier(CSTKSA)delivering anti-VEGF siRNA(R),termed as Ap-CSTKSA/R *** functionalized modification produced an enhanced cellular uptake and a stronger bio-distribution of Ap-CSTKSA/R complexes in U87 MG cells and brain tumor tissues,***-CSTKSA/R complexes exhibited great superiority in GBM growth inhibition and finally translated into the longest survival period mainly via receptor-mediated targeting delivery,VEGF gene silencing accompanied with remarkable angiogenesis inhibition,and suppressed expression of caveolin-1 which is involved in BBB functional regulation in the occurrence and treatment of *** study indicated that Ap functionalization on ROS-responsive glycolipid-like copolymer exhibits a promising and effective gene delivery platform for GBM targeted treatment.

Application of ^(18) F-FDG PET/CT Imaging in Diagnosing Bladder Tumor Metastasis Lesions

Bladder tumor is the most common malignant tumor in urinary system and always com- panied with lymph node metastasis. The accurate staging plays a significant role in treatment for bladder tumor and prognostic evaluation, and the distant metastasis predicts worse prognosis. The objective of this study was to assess the clinical significance of 18F-FDG PET/CT imaging in diagnosing bladder tumor metastasis lesions. A retrospective analysis of 60 patients with bladder tumor from October 2008 to May 2010 was done. The patients were stratified based on the imaging technique. Among all 60 cases, besides the primary lesion, 81 suspected lesions were spotted and 73 confirmed as metastasis, including 50 lymph node metastases, 22 distant metastases, and 1 bone metastasis. For PET/CT imaging, its sensitivity was 94.5%, specificity 87.5%, positive predictive value 98.6%, negative predictive value 63.6% and accuracy 93.8% respectively. For CT, its sensitivity was 82.2%, specificity 50%, positive predictive value 93.8%, negative predictive value 23.5% and accuracy 79% respectively. PET/CT im- aging was superior to CT in sensitivity, specificity and accuracy. In conclusion, 18F-FDG PET/CT imaging is more significant in diagnosing bladder tumor metastasis lesions.

APPLICATION OF TWO-DIMENSIONAL ECHOCARDIOGRAPHY IN LOCATION OF BALLOON OF THE SWAN-GANZ CATHETER

Two-dimensional echocardiogrnphy was used to guide and locate the balloon of Swan-Ganz catheter during catheterization in 43 patients. The echoes of balloon appeared as a series of strong light spots, with a rate of detection of 100%. Under monitoring of two-dimensional echocardiography, the site of balloon and its moving route could be determined. The coincidence of echographic findings and X-ray findings showed that it can be used instead of X-ray monitoring to lessen irradiation during catheterization. The depth of catheter inserted was measured. The differential points between the echoes of balloon and tubing, the terminology of the balloon site in echocardiography and fluoroscopy, its clinical value and limitation are discussed.
