Dissecting the cell microenvironment of ovarian endometrioma through single-cell RNA sequencing

Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

Deciphering the mechanism of Liu Wei Di Huang Wan in treating premature ovarian failure:a comprehensive exploration through network pharmacology and molecular docking analysis

Premature ovarian failure(POF)is a prevalent gynecological disorder with significant implications for the physical and mental well-being of affected *** Wei Di Huang Wan(LWDHW),a Chinese herbal compound,has demonstrated efficacy in alleviating the effects of ***,the underlying mechanism of action of LWDHW remains *** study aimed to elucidate the potential molecular mechanism of LWDHW in treating POF using network pharmacology and molecular docking *** active ingredients of LWDHW were initially screened through the TCMsP *** the same time,the relevant target genes associated with POF were identified using databases such as Disgenet,TTD,Drugbank,Gene Cards,OMIM,and Pharm *** analysis was conducted using the R language,Cytoscape,and sTRING to construct and analyze the traditional Chinese medicine(TCM)regulatory network and protein-protein interaction(PPI)network ***,GO and KEGG enrichment analyses were performed using the R ***,molecular docking was carried out between the protein receptors of the core genes and the corresponding small-molecule *** study revealed 49 components and 189 predicted targets(after de-duplication)of LWDHW,along with 4524 targets(after de-duplication)associated with *** comparative analysis,163 potential genes were identified as common targets of LWDHW and POF,participating in biological processes such as response to chemical substances,molecular function regulation,and signaling receptor *** biological pathways implicated included the MAPK signaling pathway,IL-17 signaling pathway,and HIF-1 signaling pathway,among *** docking results demonstrated a robust binding ability between the core genes of LWDHW and their corresponding *** conclusion,this comprehensive analysis provided insights into the potential molecular mechanisms of LWDHW in treating *** identified common targets and associated pathways contributed to

The Trend and Cause of Mortality Burden in Infancy—China,1990–2019

Introduction:China has made remarkable achievements in reducing infant deaths over the past 30 ***,the trend and cause composition of infant deaths have significantly changed,and the high-level years of life lost(YLLs)presents challenges to public *** study analyzes the temporal trend and cause of infant mortality burden in China from 1990 to ***:Based on the updated estimates from the 2019 Global Burden of Disease study,we used death and YLLs to describe the infant mortality *** percentage changes were applied to illustrate the temporal variation over the period ***:In 2019,the estimated infant deaths and YLLs in China were 101.9(95%uncertainty interval:88.6-116.4)thousand and 9.0(7.9-10.3)million,respectively,which decreased by 89.9%compared with *** infant deaths occurred in the neonatal period,especially the first 7 days of ***’s infant mortality rates for all causes decreased from 1990 to 2019,whereas certain preventable causes such as preterm birth and congenital heart anomalies accounted for increasing proportions during this *** and Implications for Public health Practice:The present study reveals the major preventable causes of infant deaths in *** neonatal mortality and improving health status of children at early stage of life have great significance on public *** is also a solution to achieve the goal of“ending preventable child deaths”of the 2030 sustainable Development Agenda.
