traditional chinese medicine(TCM)has been successfully applied worldwide in the treatment of coronavirus disease 2019(COVID-19),which is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).However,the pharmacological mechanisms underlying this success remain unclear.Hence,the aim of this review is to combine pharmacological assays based on the theory of TCM in order to elucidate the potential signaling pathways,targets,active compounds,and formulas of herbs that are involved in the TCM treatment of COVID-19,which exhibits combatting viral infections,immune regulation,and amelioration of lung injury and fibrosis.Extensive reports on target screening are elucidated using virtual prediction via docking analysis or network pharmacology based on existing data.The results of these reports indicate that an intricate regulatory mechanism is involved in the pathogenesis of COVID-19.Therefore,more pharmacological research on the natural herbs used in TCM should be conducted in order to determine the association between TCM and COVID-19 and account for the observed therapeutic effects of TCM against COVID-19.

Dear Editor,The safety and modernization of traditional chinese medicine (TCM) are significant concerns for users’ lives and health. Recently, the adverse effects caused by TCM and traditional nontoxic TCM have been frequently reported (Joo et al., 2020;Liu et al., 2021). As the safety of TCM continues to attract widespread attention, many researchers have been investigating the toxicity of TCM in regard to the toxic ingredients, effects, mechanisms, and detoxification strategies in TCM (Figure S1 in Supporting Information). Although some TCMs cause toxic reactions, they also play key roles in treating certain unignorable diseases (Fang et al., 2022).Therefore, toxicity research is crucial and urgently needed to develop and safely use TCM.

Zuogui(左归丸)and Yougui pills(右归丸)improve perimenopausal syndrome via regulation of apoptosis in mice

OBJECTIVE:To examine,dissect,and understand the molecular mechanisms and combinatorial effects of Zuogui(左归丸,ZGP)and Yougui pills(右归丸,YGP)in 4-vinyl cyclohexene diepoxide(4-VCD)-induced Perimenopausal syndrome(PMS).METHODS:Using the 4-VCD-induced PMS mouse model,uterine and ovary index were measured,and serum sex steroidal hormone levels were evaluated after treatment with ZGP,YGP,ZGP+YGP,estradiol valerate(EV),and Gengnian An(更年安,GNA).Histopathological examinations,ingredient-target network predictions,western blotting,and real-time quantitative polymerase chain reaction(RT-q PCR)analyses were performed to ascertain the possible pharmacological effects and molecular mechanisms of ZYP and YGP.RESULTS:Treatment with ZGP and YGP remarkably improves estrous cyclicity and prevents pathological damage to the uterus.Also,altered sex hormones,including AMH,E2,FSH,LH,P,and T,were restored to normal levels after ZGP and YGP administration.Ingredient-target network analysis showed that the 5 ingredients common to the ZGP and YGP formula modulate 53 targets shared with PMS.Pathwayenrichment analysis further predicted that ZGY and YGP likely regulate of apoptosis and other essential pathways during PMS.In-vivo studies showed that ZGP and YGP suppress PMS via modulating apoptosis through decreasing Caspase-3 and Bcl-2-associated X(Bax)levels and increasing B-cell lymphoma-2(Bcl-2)/Bax and Bcl-2 levels.Importantly,ZGP+YGP treatment modulation effects were somewhat or significantly better compared to ZGP or YGP alone treatment.CONCLUSION:ZGP and YGP represent novel anti-PMS agents whose effects involve restoring altered hormonal levels,protecting the uterus,and regulating apoptosis.

Cancer has been an insurmountable problem in the history of medical science.The uncontrollable proliferation of cancer cells is one of cancers main characteristics,which is closely associated with abn ormal mitosis.Targeting mitosis is an effective method for cancer treatment.This review summarizes several natural products with anti-tumor effects related to mitosis,focusing on targeting microtubulin,inducing DNA damage,and modulating mitosis-associated kinases.Furthermore,the main disadvantages of several typical compounds,including drug resistance,toxicity to non-tumor tissues,and poor aqueous solubility and pharmacokinetic properties,are also discussed,together with strategies to address them.Improved understanding of cancer cell mitosis and natural products may pave the way to drug development for the treatment of cancer.

As a serious cardiovascular disease,atherosclerosis(AS)causes chronic inflammation and oxidative stress in the body and poses a threat to human health.Lipoprotein-associated phospholipase A2(Lp-PLA2)is a member of the phospholipase A2(PLA2)family,and its elevated levels have been shown to contribute to AS.Lp-PLA2 is closely related to a variety of lipoproteins,and its role in promoting inflammatory responses and oxidative stress in AS is mainly achieved by hydrolyzing oxidized phosphatidylcholine(ox PC)to produce lysophosphatidylcholine(lyso PC).Moreover,macrophage apoptosis within plaque is promoted by localized Lp-PLA2 which also promotes plaque instability.This paper reviews those researches of chinese medicine in treating AS via reducing Lp-PLA2 levels to guide future experimental studies and clinical applications related to AS.

Diabetes comprises a group of metabolic diseases characterized by hyperglycemia stemming from various factors.Current diabetes management primarily focuses on blood glucose control,yet it is inherently progressive,necessitating increased reliance on exogenous blood glucose control methods over time.Therefore,there is an urgent need to explore novel intervention strategies addressing both diabetes and its complications.The human intestinal microbiota,often referred to as the"second genome",exhibits significant diversity and plays a pivotal role in insulin resistance,glucose and lipid metabolism,and inflammatory response.Notably,Li and Guo have elucidated the involvement of intestinal flora in the pathogenesis of type 2 diabetes mellitus(T2DM)and proposed a novel therapeutic approach targeting intestinal microbes.This advancement enhances our comprehension of the multifaceted and multi-target regulation of T2DM by intestinal microflora,thereby offering fresh avenues for understanding its pathogenesis and clinical management.This letter briefly summarizes the role of intestinal flora in T2DM based on findings from animal experiments and clinical studies.Additionally,it discusses the potential clinical applications and challenges associated with targeting intestinal flora as therapeutic interventions.

Alzheimer’s disease(AD)is the most common neurodegenerative disease characterized by progressive cognitive impairment.The pathogenesis of AD is complex,and its susceptibility and development process are affected by age,genetic and epigenetic factors.Recent studies confirmed that gut microbiota(GM)might contribute to AD through a variety of pathways including hypothalamic pituitary adrenal axis and inflammatory and immune processes.CM formula,herbs,and monomer enjoy unique advantages to treat and prevent AD.Hence,the purpose of this review is to outline the roles of GM and its core metabolites in the pathogenesis of AD.Research progress of CMs regarding the mechanisms of how they regulate GM to improve cognitive impairment of AD is also reviewed.The authors tried to explore new therapeutic strategies to AD based on the regulation of GM using CM.

Vascular endothelial growth factor(VEGF)is the main regulator of physiological angiogenesis during embryonic development,bone growth,and reproductive function,and it also participates in a series of pathological changes.traditional chinese medicine(TCM),with a history of more than 2000 years,has been widely used in clinical practice,while the exploration of its mechanisms has only begun.This review summarizes the research of recent years on the influence of TCM on VEGF.It is found that many chinese medicines and recipes have a regulatory effect on VEGF,indicating that chinese medicine has broad prospects as a complementary and alternative therapy,providing new treatment ideas for clinical applications and the theoretical basis for research on the mechanisms of TCM.

Ineffectiveness of Skin Tests in Predicting Allergic Reactions Induced by chinese Herbal Injections

Objective:To evaluate whether skin tests are suitable to predict the allergy reactions induced by chinese herbal injections(CHIs).Methods:The skin tests including skin prick tests(SPT),intradermal tests(IDT)and provocation tests including subcutaneous tests and intravenous tests were administered to 249 healthy subjects and 180 allergic patients for 3 CHIs,including ginkgolide injection,diterpene ginkgolide meglumine injection and Salvianolate lyophilized injection.The results of the provocation tests were used as the"gold standard"to determine the sensitivity and specificity of the skin tests.Results:The results did not show any significant differences between the healthy and allergy groups in both skin tests and provocation tests(P>0.05).The specificities of SPT and IDT were 0.976 and 0.797,respectively,and the sensitivities of both SPT and IDT were 0.Conclusion:Skin tests are insufficient to predict the likelihood of allergic reactions resulting from CHIs.(ChiCTR-CPC-15006921)

Vaccination is a major achievement that has become an effective prevention strategy against infectious diseases and active control of emerging pathogens worldwide.In response to the coronavirus disease 2019(COVID-19)pandemic,several diverse vaccines against severe acute respiratory syndrome coronavirus 2 have been developed and deployed for use in a large number of individuals,and have been reported to protect against symptomatic COVID-19 cases and deaths.However,the application of vaccines has a series of limitations,including protective failure for variants of concern,unavailability of individuals due to immune deficiency,and the disappearance of immune protection for increasing infections in vaccinated individuals.These aspects raise the question of how to modulate the immune system that contributes to the COVID-19 vaccine protective effects.Herbal medicines are widely used for their immune regulatory abilities in clinics.More attractively,herbal medicines have been well accepted for their positive role in the COVID-19 prevention and suppression through regulation of the immune system.This review presents a brief overview of the strategy of COVID-19 vaccination and the response of the immune system to vaccines,the regulatory effects and mechanisms of herbal medicine in immune-related macrophages,natural killer cells,dendritic cells,and lymphocytes T and B cells,and how they help vaccines work.Later in the article,the potential role and application of herbal medicines in the most recent COVID-19 vaccination are discussed.This article provides new insights into herbal medicines as promising alternative supplements that may benefit from COVID-19 vaccination.
