AIM:To determine the feasibility and safety of high dose rate intraluminal brachytherapy(HDR-ILBT) boost during preoperative chemoradiation for rectal ***:Between 2008 and 2009,thirty-six patients with locally advanced rectal cancer(≥ T3 or N+),were treated initially with concurrent capecitabine(825 mg/m2 oral twice daily) and pelvic external beam radiotherapy(EBRT)(45 Gy in 25 fractions),then were randomized to group A;HDR-ILBT group(n = 17) to receive 5.5-7 Gy×2 to gross tumor volume(GTV) and group B;EBRT group(n = 19) to receive 5.4 Gy×3 fractions to GTV with *** patients underwent total mesorectal ***:Grade 3 acute toxicities were registered in 12 patients(70.6%) in group A and in 8(42.1%) in group *** pathologic response of T stage(ypT0) in group A was registered in 10 patients(58.8%) and in group B,3 patients(15.8%) had ypT0(P < 0.0001).Sphincter preservation was reported in 6/9 patients(66.7%) in group A and in 5/10 patients(50%) in group B(P < 0.01).Overall radiological response was 68.15% and 66.04% in Group A and B,*** a median follow up of 18 mo,late grade 1 and 2 sequelae were registered in 3 patients(17.6%) and 4 patients(21.1%) in the groups A and B,***:HDR-ILBT was found to be effective dose escalation technique in preoperative chemoradiation for rectal cancers,with higher response rates,downstaging and with manageable acute toxicities.

Prostate mucinous adenocarcinoma with signet ring cells (MC-SRC) is a rare morphologic variant of prostate cancer, with only 12 cases reported to date. Diagnosis of this carcinoma requires that at least 25% of the tumor tissue should consist of an extracellular mucin pool. In this report, we present a case of prostate prostate mucinous adenocarcinoma with signet ring cells.

Owing to the novel physicochemical properties of graphene, materials such as graphene oxide （GO） are being developed for applications in various fields such as biomedicine. Nonetheless, considerable knowledge gaps still exist regarding the impact of GOs on environmental health and safety （EHS）. Thus far, its secondary toxicity, synergistic effects, and mal-adaption have not been focused much upon. Here, we show that at low concentrations （that did not directly result in significant cytotoxicity）, GO could greatly enhance metal toxicity in macrophages by altering their cellular priming state. Specificall）~ GO caused impairments to the cellular morphology and membrane integrity of macrophages, and remarkably enhanced the cellular uptake of Cd and other non-essential metal ions （such as Hg and Gd）. Furthermore, upon low-dose GO pre-treatment, the uptake of Cd at a non-toxic concentration brought about a remarkable amount of oxidative stress in macrophages, and ultimately resulted in increased cell death. Mechanistic investigations illustrated that GO pre-treatment triggered cell death through apoptosis because of Cd exposure. Overall this study＇s results reveal a new path for understanding the impact of GOs on EHS through the perspective of its synergistic and secondary effects, previously unidentified mechanisms via which nanomaterials might pose detrimental effects on organisms.

Objective:To study the protective effect of telmisartan on rats with renal failure and its ***:60 Wistar rats were chosen as study objective,and were divided into4 groups randomly:15 in group A(sham operation group).15 in group B(model group),15 in group C(telmisartan group) and 15 in group D(telmisartan+GW9962 group).The difference of survival rate,blood-urine biochemical indexes,renal pathological change,and the expression level of PPAR γ and nNOS were ***:After 12 weeks,the survival rate of group A was 93.33% (14/15),that of group B was 46.67% (7/15),that of group C was86.67% (13/15),that of group D was 60.00% (9/15),and the difference among 4 groups had statistical significance(P0.05);after 3 weeks,6 weeks and 12 weeks,these difference was statistical significant(P<0.05).The difference of blood-urine biochemical indexes,that of renal pathological change,and that of the expression level of PPAR γ and nNOS was statistical significant(P<0.05).Conclusions:Telmisartan has protective effect on renal failure caused by 5/6 nephrectomy,which might be relative to the expression level of PPAR γ and nNOS.

The Biological Effect of Hepsin on the Proliferation and Invasion of PC-3 Prostate Cancer Cells

OBJECTIVE Recent studies have shown that hepsin, a type of transmembrane serine protease, is highly upregulated in prostate cancer, but, little is known about its role in progression and invasion of this cancer. We constructed a hepsin-expressing plasmid and transfected it into PC-3 cells to investigate the effect of the hepsin gene on the biological behavior of the PC-3 cells. METHODS Plasmid pHepsin-IRES2 was transfected into prostate cancer PC-3 cells using Fugene6, and the cells with stable hepsin expression were screened and selected with Zeocin (600 mg/L). The hepsin mRNA level was measured by real-time PCR and the growth curve of the PC-3-transfected cells assessed using MTT and BrdU assays. A Boyden chamber was used to examine the difference in invasion and metastases between transfected and non-transfected cells. RESULTS The hepsin mRNA level in pHepsin-IRES2 transfected -PC-3 cells was significantly higher than that found in the control PC -3 cells. While the growth curve of the hepsin gene transfected PC -3 cells showed that there was no significant effect on proliferation, the invasive ability of the pHepsin-IRES2 transfected PC-3 cells, as compared with control cells, was significantly increased (P<0.05). CONCLUSION The results suggest that even though hepsin has no effect on the proliferation of prostate cancer PC-3 cells, it does promote cellular invasion and *** hepsin may have a role in the development of prostate cancer.

IntroductionChina is a country with a low morbidity of prostate carcinoma. The incidence of prostate carcinoma in China is 1.6/100,000, which is much lower than the rate in the United States, i.e., 119.9/100,000. Due to changes of lifestyle and improved measurement of serum prostate specific antigen （PSA） over the past decades, the incidence of prostate carcinoma in China also showed a yearly increase. However, the misdiagnosis and mistreatment of this disease commonly occur. The aim of this study is to report the case of a patient with prostate carcinoma presenting asymptomatic but with left supraclavicular lymphadenopathy, and analyze the reasons of misdiagnosis and mistreatment.
