Immunotherapy represents a promising strategy for cancer treatment that utilizes immune cells or drugs to activate the patient's own immune system and eliminate cancer *** of the most exciting advances within this field is the targeting of neoantigens,which are peptides derived from non-synonymous somatic mutations that are found exclusively within cancer cells and absent in normal *** neoantigen-based therapeutic vaccines have not received approval for standard cancer treatment,early clinical trials have yielded encouraging outcomes as standalone monotherapy or when combined with checkpoint *** made in high-throughput sequencing and bioinformatics have greatly facilitated the precise and efficient identification of ***,personalized neoantigen-based vaccines tailored to each patient have been developed that are capable of eliciting a robust and long-lasting immune response which effectively eliminates tumors and prevents *** review provides a concise overview consolidating the latest clinical advances in neoantigen-based therapeutic vaccines,and also discusses challenges and future perspectives for this innovative approach,particularly emphasizing the potential of neoantigen-based therapeutic vaccines to enhance clinical efficacy against advanced solid tumors.

Early clinical outcome with lens position adjustment following implantable collamer lens surgery

AIM:To observe early clinical outcome with lens position adjustment following the implantable collamer lens(ICL)***:Sixty patients were selected for this retrospective *** eye from each patient received Toric ICL for astigmatism correction,and the other received non-astigmatic ICL surgery using horizontal *** with higher postoperative arch height were selected,and their non-astigmatic eye clinical outcome were observed after ICL surgery at 1wk,1,and *** clinical measurements included uncorrected visual acuity(UCVA),intraocular pressure(IOP),refractive state,corneal endothelium cell count,and arch *** months later,the ICL in each patient’s non-astigmatic eye was adjusted to the vertical from the horizontal *** results were compared before and 1wk,1,and 3mo after ***:UCVA and IOP were significantly reduced 1wk after position adjustment compared to 1wk after ICL implantation(P0.05).CONCLUSION:Early positioning adjustment postphakic ICL implantation can benefit patients with adjusted arch height or higher *** the good clinical effects,the doctors should pay attention to the potential for adverse effects on UCVA and corneal endothelium cells following early position adjustment after posterior chamber phakic ICL implantation.

In vitro comparison of the hemocompatibility of diamond-like carbon and carbon nitride coatings with different atomic percentages of N

Carbon nitride （CN,） and diamond-like carbon （DLC） coatings were prepared by dc magnetron sputtering at room temperature. Different partial pressures of N2 were used to synthesize CNx to evaluate the relationship between the atomic percentage of ni- trogen and hemocompatibility. Auger electron spectroscopy and atomic force microscopy indicated atomic percentages of N of 0.12 and 0.22 and that the CNx coatings were smooth. An in vitro study of the hemocompatibility of the coatings revealed that both CNx coatings had better anticoagulant properties and lower platelet adhesion than DLC. Compared with CN0.1〉 the CN0.22 coating showed longer dynamic clotting time （about 42 min）, static clotting time （23.6 min） and recalcification time （45.6 s）, as well as lower platelet adhesion （102 cells μm-2）, aggregation, and activation. The presence of nitrogen in the CNx coatings in- duced their enhanced hemocompatibility compared with DLC.

Satisfactory surgical outcome of T2 gastric cancer after modified D2 lymphadenectomy

Objective： Though D2 lymphadenectomy has been increasingly regarded as standard surgical procedure for advanced gastric cancer （GC）, the modified D2 （D 1＋7, 8a and 9） lymphadenectomy may be more suitable than D2 dissection for T2 stage GC. The purpose of this study is to elucidate whether the surgical outcome of modified D2 lymphadenectomy was comparable to that of standard D2 dissection in T2 stage GC patients. Methods： A retrospective cohort study with 77 cases and 77 controls matched for baseline characteristics was conducted. Patients were categorized into two groups according to the extent of lymphadenectomy： the modified D2 group （roD2） and the standard D2 group （D2）. Surgical outcome and recurrence date were compared between the two groups. Results： The 5-year overall survival （OS） rate was 71.4% for patients accepted mD2 lymphadenectomy and 70.1% for those accepted standard D2, respectively, and the difference was not statistically significant. Multivariate survival analysis revealed that curability, tumor size, TNM stage and postoperative complications were independently prognostic factors for T2 stage GC patients. Patients in the mD2 group tended to have less intraoperative blood loss （P=0.001） and shorter operation time （P〈0.001） than those in the D2 group. While there were no significant differences in recurrence rate and types, especially lymph node recurrence, between the two groups. Conclusions： The surgical outcome of mD2 lymphadenectomy was equal to that of standard D2, and the use of mD2 instead of standard D2 can be a better option for T2 stage GC.

Background:Atrial septal defect(ASD)is one of the common congenital heart *** MYH6 gene has a critical role in cardiac development but the role of MYH6 promoter variants in patients with ASD has not been ***:In 613 subjects including 320 ASD patients,we investigated the MYH6 gene promoter variants and verified the effect on gene expression by using cellular functional experiments and bioinformatics ***:Eleven variants were identified in the MYH6 gene promoter,of which four variants were found only in ASD patients,and two variants(g.3434G>C and g.4524C>T)were identified for the first *** functional experiments indicated that all four variants reduced the transcriptional activity of the MYH6 gene promoter(p<0.05).Subsequent analysis through the JASPAR(A database of transcription factor binding profiles)suggests that these variants may alter transcription factor binding sites,which may in turn lead to changes in myocardin subunit expression and ASD ***:Our study for the first time focuses on variants in the promoter region of the MYH6 gene in Chinese patients with ASD and the discovered variants have functional *** study provides new insights in the role of the MYH6 gene promoter region to better understand the genetic basis of ASD formation and facilitates clinical diagnosis.

An injectable curcumin-releasing organohydrogel with non-drying property and high mechanical stability at low-temperature for expe dite d skin wound care

Although hydrogels have demonstrated great potential as new wound dressing materials,their instability in shape and/or mechanical characteristics due to water loss or freezing remains a shortcoming for wound care ***,a novel injectable organohydrogel(IOH)of physically crosslinked polyvinyl alco-hol/glycerol that possesses non-drying capability and high stability at low temperature was developed for wound *** has a skin-like stiffness(G’,∼280 Pa),high injectability,self-healing capability,high water-vapor transmission rate,and bacterial inhibitory *** exhibits high shape and mechanical stabilities after curing at 37°C and 50%relative humidity for 7 days or after curing at-20°C for 1 *** addition,glycerol in IOH enabled an efficient loading and release of water-insoluble curcumin,a well-known anti-bacterial and anti-inflammation *** curcumin-releasing IOH(Cur-IOH)demonstrated significantly enhanced anti-bacterial performance compared to IOH or curcumin-loading polyvinyl alco-hol *** importantly,Cur-IOH could accelerate wound healing in a murine full-thickness skin defect wound model,revealing improved wound contraction,collagen deposition,angiogenesis,and epi-dermis *** study demonstrates the great potential of organohydrogel for the reparation of severe wounds and Cur-IOH as a new type of injectable wound healing material.

Negative impact of hepatitis B surface seroclearance on prognosis of hepatitis B-related primary liver cancer

AIM To assess the impact of hepatitis B surface(HBs Ag) seroclearance on survival outcomes in hepatitis B-related primary liver *** Information from patients with hepatitis B-related liver cancer admitted in our hospital from 2008-2017 was retrieved. Cases diagnosed with HBs Ag(-) and HBc Ab(+) liver cancer were included in the HBs Ag seroclearance(SC) group. HBs Ag(+) liver cancer patients strictly matched for liver cancer stage(AJCC staging system, 8 th edition), Child-Pugh score, and first diagnosis/treatment method(surgery, ablation and TACE) were assigned to the HBsA g non-seroclearance(NSC) group. Then, clinical, pathological and survival data in both groups were *** The SC and NSC groups comprised of 72 and 216 patients, respectively. Patient age(P surgery had more intrahepatic cholangiocarcinoma(ICC) and combined HCC-CC(CHC) cases than the NSC group, but no significant differences in tumor cell differentiation and history of liver cirrhosis were found between the two groups. The numbers of interventional treatments were similar in both groups(4.57 vs 5.07, P > 0.05). Overall survival was lower in the SC group than the NSC group(P = 0.019), with 1-,3-, and 5-year survival rates of 82.1% vs 85.1%, 43.2%vs 56.8%, and 27.0% vs 45.2%, respectively. Survival of patients with AJCC stage Ⅰ disease in the SC group was lower than that of the NSC group(P = 0.029).CONCLUSION Seroclearance in patients with hepatitis B-related primary liver cancer has protective effects with respect to tumorigenesis, cirrhosis, and portal hypertension but confers worse prognosis, which may be due to the frequent occurrence of highly malignant ICC and CHC.
