Genomic surveillance of emerging SARS-CoV-2 Omicron variations in tianjin Municipality, China 2022

The coronavirus disease 2019(COvID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has severely impacted public *** 2022,the Omicron variant of SARS-CoV-2 rapidly became the dominant circulating variant in the local COVID-19 outbreaks in tianjin Municipality,*** gain a deeper understanding of the genetic variations of the Omicron variant in tianjin,specimens from indi-viduals who tested positive for SARS-CoV-2 between December 2021 and November 2022 were used for virus whole genome sequencing and phylogenetic analysis.A total of 1,674 high-quality Omicron sequences were obtained,consisting of 1,339 sequences from local cases belonging to 20 Phylogenetic Assignment of Named Global Outbreak(PANGO)lineages and 335 sequences from imported cases belonging to 70 *** experienced five waves of local outbreaks,accompanied by multiple substitutions among subvariants,ranging from the initial BA.1.1 lineage to the subsequent BA.2,BF.7,and BA.5.2 *** evolutionary rate of local strains,estimated to be 28.999 substitutions per year,and the evolutionary rate of imported strains,estimated to be 24.946 substitutions per year,were lower than that of the strains circulating *** addi-tional substitutions and deletions of local strains have been used to identify and disrupt the virus transmission *** subvariants such as BA.5.2.48,BA.5.2.49,BF.7.14,and XBB.1 circulating in the fifth epidemic wave presented criterial immune escape mutations including S:R346T,S:L452R and S:*** is essential to implement genomic surveillance strategies to investigate further the development of genomic mutation char-acteristics in the SARS-CoV-2 *** ongoing monitoring will contribute to a better understanding of the virus's genetic changes and aid in effective control measures.

Different Types of Bone Fractures Observed by Terahertz Time-Domain Spectroscopy

Microcracks are common in compact bone,but their continued propagation can lead to macroscopic *** microcracks cannot be visualized radiographically,necessitating alternative noninvasive methods to identify excessive microcracking and prevent *** this study,terahertz time-domain spectroscopy(THz-TDS)was used to examine bone interiors near cracks resulting from loading in bovine tibia *** loading configurations,such as impact,quasi-static loading,and fatigue loading,known to induce different types of micro-scale damage,were *** values of refractive index and absorption coefficient of the bone samples were then determined from the THz-TDS spectra acquired before loading and after *** study revealed that different loading configurations led to varying terahertz optical coefficients associated with various types of bone ***,the refractive index notably increased under fatigue loading but remained relatively stable during quasi-static *** absorption coefficient of bone decreased only under fatigue ***,samples were subjected to axial and radial impacts without sustaining *** indicated that in the undamaged state,the change in refractive index was smaller compared to after impact failure,while the change in absorption coefficient remained consistent after *** radial impact loading,changes in refractive index and absorption coefficient were significantly more pronounced than under axial *** to loading,the measured value of refractive index was 2.72±0.11,and the absorption coefficient was 6.33±0.09 mm^(−1)at 0.5 THz.

Mechanically lined pipe(MLP)is often used for offshore oil and gas transport because of its low cost and corrosion *** installation and operation,the pipe may undergo severe bending deformation,which causes the liner to separate from the outer pipe and buckles,affecting the stability of the whole *** this paper,the buckling response of MLP subjected to bending is investigated to clarify its bending characteristics by employing both experiments,numerical simulation,as theoretical *** types of MLPs were manufactured with GB 45 carbon steel(SLP)and Al 6061(ALP)used as the outer pipe material,*** hydraulic expansion and bending experiments of small-scale MLPs are *** addition to the ovalized shape of the cross-section for the SLP specimens,the copper liner was found to wrinkle on the compressive *** contrast,the liner of ALP remains intact without developing any wrinkling and collapse *** addition,a dedicated numerical framework and theoretical models were also *** was found both the manufacturing and bending responses of the MLP can be well reproduced,and the predicted maximum moment and critical curvatures are in good agreement with the experimental results.

Exosomes derived from microglia overexpressing miR-124-3p alleviate neuronal endoplasmic reticulum stress damage after repetitive mild traumatic brain injury

We previously reported that miR-124-3p is markedly upregulated in microglia-derived exosomes following repetitive mild traumatic brain ***,its impact on neuronal endoplasmic reticulum stress following repetitive mild traumatic brain injury remains *** this study,we first used an HT22 scratch injury model to mimic traumatic brain injury,then co-cultured the HT22 cells with BV2 microglia expressing high levels of *** found that exosomes containing high levels of miR-124-3p attenuated apoptosis and endoplasmic reticulum ***,luciferase reporter assay analysis confirmed that miR-124-3p bound specifically to the endoplasmic reticulum stress-related protein IRE1α,while an IRE1αfunctional salvage experiment confirmed that miR-124-3p targeted IRE1αand reduced its expression,thereby inhibiting endoplasmic reticulum stress in injured ***,we delivered microglia-derived exosomes containing miR-124-3p intranasally to a mouse model of repetitive mild traumatic brain injury and found that endoplasmic reticulum stress and apoptosis levels in hippocampal neurons were significantly *** findings suggest that,after repetitive mild traumatic brain injury,miR-124-3 can be transferred from microglia-derived exosomes to injured neurons,where it exerts a neuroprotective effect by inhibiting endoplasmic reticulum ***,microglia-derived exosomes containing miR-124-3p may represent a novel therapeutic strategy for repetitive mild traumatic brain injury.

Evolution of Biofilm and Its Effect on Microstructure of Mortar Surfaces in Simulated Seawater

To explore the role of biofilm formation on the corrosion of marine concrete structures, we investigated the attachment of biofilm on mortar surfaces in simulated seawater and the influence of biofilm on the microstructure of mortar surfaces. The results show that the evolution of biofilm on mortar surfaces in simulated seawater is closely related to the corrosion suffered by the mortar, and the process of biofilm attachment and shedding is continuous and cyclical. It is found that the specimens in the absence of biofilm attachment are more severely eroded internally by the corrosive medium in simulated seawater than those in the presence of biofilm attachment. For the specimens without biofilm attachment, after 60 days, gypsum forms,and after 120 days, the number of pores in the mortar is reduced. In contrast, for the specimens in the presence of biofilm attachment, gypsum could only be detected after 90 days, and fewer pores are filled. Therefore, the formation of biofilm could delay the invasion of the corrosive medium into the interior of mortar during the evolution of biofilm on mortar surfaces, mitigating the corrosion of mortars in seawater.

Performance and Application of Double-layered Microcapsule Corrosion Inhibitors

Double-layered microcapsule corrosion inhibitors were developed by sodium monofluorophosphate as the core material,polymethyl methacrylate as the inner wall material,and polyvinyl alcohol as the outer wall material combining the solvent evaporation method and spray drying *** protection by the outer capsule wall was used to prolong the service life of the corrosion *** dispersion,encapsulation,thermal stability of microcapsules,and the degradation rate of capsule wall in concrete pore solution were analyzed by ultra-deep field microscopy,scanning electron microscopy,thermal analyzer,and sodium ion release rate *** microcapsules were incorporated into mortar samples containing steel reinforcement,and the effects of double-layered microcapsule corrosion inhibitors on the performance of the cement matrix and the actual corrosion-inhibiting effect were *** experimental results show that the double-layered microcapsules have a moderate particle size and uniform distribution,and the capsules were completely *** microcapsules as a whole have good thermal stability below 230 ℃.The monolayer membrane structure microcapsules completely broke within 1 day in the simulated concrete pore solution,and the double-layer membrane structure prolonged the service life of the microcapsules to 80 days in the simulated concrete pore solution before the core material was completely *** mortar samples containing steel reinforcement incorporated with the double-layered microcapsule corrosion inhibitors still maintained a higher corrosion potential than the monolayer microcapsule corrosion inhibitors control group at 60 *** incorporation of double-layered microcapsules into the cement matrix has no significant adverse effect on the setting time and early strength.

Influence of Incomplete Soil Plugs on Bearing Capacities of Bucket Foundations in Clay

Due to the uneven seabed and heaving of soil during pumping,incomplete soil plugs may occur during the installation of bucket foundations,and the impacts on the bearing capacities of bucket foundations need to be *** this paper,the contact ratio(the ratio of the top diameter of the soil plug to the diameter of the bucket)and the soil plug ratio(the ratio of the soil heave height to the skirt height)are defined to describe the shape and size of the incomplete soil ***,finite element models are established to investigate the bearing capacities of bucket foundations with incomplete soil plugs and the influences of the contact ratios,and the soil plug ratios on the bearing capacities are *** results show that the vertical bearing capacity of bucket foundations in homogeneous soil continuously improves with the increase of the contact ***,in normally consolidated soil,the vertical bearing capacity barely changes when the contact ratio is smaller than 0.75,while the bearing capacity suddenly increases when the contact ratio increases to 1 due to the change of failure *** contact ratio hardly affects the horizontal bearing capacity of bucket ***,the moment bearing capacity improves with the increase of the contact ratio for small aspect ratios,but hardly varies with increasing contact ratio for aspect ratios larger than ***,the reduction coefficient method is proposed based on this analysis to calculate the bearing capacities of bucket foundations considering the influence of incomplete soil *** comparison results show that the proposed reduction coefficient method can be used to evaluate the influences of incomplete soil plug on the bearing capacities of bucket foundations.
