Weak SARS-CoV-2-specific responses of TIGIT-expressing CD8^(+)T cells in people living with HIV after a third dose of a SARS-CoV-2 inactivated vaccine

Background:T-cell immunoreceptor with immunoglobulin and immunoreceptor tyrosine-based inhibition motif domains(TIGIT),an inhibitory receptor expressed on T cells,plays a dysfunctional role in antiviral infection and antitumor activity.However,it is unknown whether TIGIT expression on T cells influences the immunological effects of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)inactivated vaccines.Methods:Forty-five people living with HIV(PLWH)on antiretroviral therapy(ART)for more than two years and 31 healthy controls(HCs),all received a third dose of a SARS-CoV-2 inactivated vaccine,were enrolled in this study.The amounts,activation,proportion of cell subsets,and magnitude of the SARS-CoV-2-specific immune response of TIGIT^(+)CD4^(+)and TIGIT^(+)CD8^(+)T cells were investigated before the third dose but 6 months after the second vaccine dose(0W),4 weeks(4W)and 12 weeks(12W)after the third dose.Results:Compared to that in HCs,the frequency of TIGIT^(+)CD8^(+)T cells in the peripheral blood of PLWH increased at 12W after the third dose of the inactivated vaccine,and the immune activation of TIGIT^(+)CD8^(+)T cells also increased.A decrease in the ratio of both T naïve(T_(N))and central memory(T_(CM))cells among TIGIT^(+)CD8^(+)T cells and an increase in the ratio of the effector memory(T_(EM))subpopulation were observed at 12W in PLWH.Interestingly,particularly at 12W,a higher proportion of TIGIT^(+)CD8^(+)T cells expressing CD137 and CD69 simultaneously was observed in HCs than in PLWH based on the activation-induced marker assay.Compared with 0W,SARS-CoV-2-specific TIGIT^(+)CD8^(+)T-cell responses in PLWH were not enhanced at 12W but were enhanced in HCs.Additionally,at all time points,the SARS-CoV-2-specific responses of TIGIT^(+)CD8^(+)T cells in PLWH were significantly weaker than those of TIGIT-CD8^(+)T cells.However,in HCs,the difference in the SARS-CoV-2-specific responses induced between TIGIT^(+)CD8^(+)T cells and TIGIT-CD8^(+)T cells was insignificant at 4W and 12W,except at 0W.Conclusio

Background:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine can induce a potent cellular and humoral immune response to protect against SARS-CoV-2 infection.However,it was unknown whether SARS-CoV-2 vaccination can induce effective natural killer(NK)cell response in people living with human immunodeficiency virus(PLWH)and healthy individuals.Methods:Forty-seven PLWH and thirty healthy controls(HCs)inoculated with SARS-CoV-2 inactivated vaccine were enrolled from Beijing Youan Hospital in this study.The effect of SARS-CoV-2 vaccine on NK cell frequency,phenotype,and function in PLWH and HCs was evaluated by flow cytometry,and the response of NK cells to SARS-CoV-2 Omicron Spike(SARS-2-OS)protein stimulation was also evaluated.Results:SARS-CoV-2 vaccine inoculation elicited activation and degranulation of NK cells in PLWH,which peaked at 2 weeks and then decreased to a minimum at 12 weeks after the third dose of vaccine.However,in vitro stimulation of the corresponding peripheral blood monocular cells from PLWH with SARS-2-OS protein did not upregulate the expression of the aforementioned markers.Additionally,the frequencies of NK cells expressing the activation markers CD25 and CD69 in PLWH were significantly lower than those in HCs at 0,4 and 12 weeks,but the percentage of CD16^(+)NK cells in PLWH was significantly higher than that in HCs at 2,4 and 12 weeks after the third dose of vaccine.Interestingly,the frequency of CD16^(+)NK cells was significantly negatively correlated with the proportion of CD107a^(+)NK cells in PLWH at each time point after the third dose.Similarly,this phenomenon was also observed in HCs at 0,2,and 4 weeks after the third dose.Finally,regardless of whether NK cells were stimulated with SARS-2-OS or not,we did not observe any differences in the expression of NK cell degranulation markers between PLWH and HCs.Conclusions:SARS-CoV-2 vaccine elicited activation and degranulation of NK cells,indicating that the inoculation of SARS-CoV-2 vaccine enhances NK cell immune respo

AIM:To investigate the expression and methylation status of the secreted frizzled-related protein 2(SFRP2) in esophageal squamous cell carcinoma(ESCC) and explore its role in ESCC carcinogenesis.METHODS:Seven ESCC cell lines(KYSE 30,KYSE150,KYSE410,KYSE510,EC109,EC9706 and TE-1) and one immortalized human esophageal epithelial cell line(Het1A),20 ESCC tissue samples and 20 paired adjacent non-tumor esophageal epithelial tissues were analyzed in this study.Reverse-transcription polymerase chain reaction(RT-PCR) was employed to investigate the expression of SFRP2 in cell lines,primary ESCC tumor tissue,and paired adjacent normal tissue.Methylation status was evaluated by methylation-specific PCR and bisulfite sequencing.The correlation between expression and promoter methylation of the SFRP2 gene was confirmed with treatment of 5-aza-2'-deoxycytidine.To assess the potential role of SFRP2 in ESCC,we es-tablished stable SFRP2-transfected cells and examined them with regard to cell proliferation,colony formation,apoptosis and cell cycle in vivo and in vitro.RESULTS:SFRP2 mRNA was expressed in the immortalized normal esophageal epithelial cell line but not in seven ESCC cell lines.By methylation-specific PCR,complete methylation was detected in three cell lines with silenced SFRP2 expression,and extensive methylation was observed in the other four ESCC cell lines.5-aza-2'-deoxycytidine could restore the expression of SFRP2 mRNA in the three ESCC cell lines lacking SFRP2 expression.SFRP2 mRNA expression was obviously lower in primary ESCC tissue than in adjacent normal tissue(0.939 ± 0.398 vs 1.51 ± 0.399,P < 0.01).SFRP2 methylation was higher in tumor tissue than in paired normal tissue(95% vs 65%,P < 0.05).The DNA methylation status of the SFRP2 correlated inversely with the SFRP2 expression.To assess the potential role of SFRP2 in ESCC,we established stable SFRP2 transfectants and control counterparts by introducing pcDNA3.1/v5 hisA-SFRP2 or pcDNA3.1/v5 hisA-empty vector into KYSE30 cells lacking SFRP2 expression.Aft

Avanafil, a potent new selective phosphodiesterase type 5 （PDE5） inhibitor, has been developed for the treatment of erectile dysfunction （ED）. We carried out a systematic review and meta-analysis to assess the efficacy and safety of this drug for the treatment of ED. A literature review was performed to identify all published randomized, double-blind, placebo-controlled trials of avanafil for the treatment of ED. The search included the following databases： MEDLINE, EMBASE and the Cochrane Controlled Trials Register. The reference lists of the retrieved studies were also investigated. Four publications, involving a total of 1381 patients, were used in the analysis, including four randomized controlled trials （RCTs） that compared avanafU with a placebo. Among the co-primary efficacy end points indicating that avanafil 100 mg was more effective than a placebo were successful vaginal penetration （SEP2）（the odds ratio （OR） =5.06, 95% confidence interval （CI） =3.29-7.78, P〈 0.00001） and successful intercourse （SEP3）（OR = 3.99, 95% CI = 2.80-5.67, P 〈 0.00001）. Men randomized to receive avanafil were less likely than those receiving the placebo to drop out due to an adverse event （AE）（OR = 1.48, 95% CI = 0.54-4.08, P = 0.44）. Specific AEs with avanafil included headache and flushing, which were significantly less likely to occur with placebo. This meta-analysis indicates that avanafil 100 or 200 mg is an effective and well-tolerated treatment for ED. Compared with avanafil 100 mg, patients who take avanafil 200 mg are more likely to experience headaches.

Previous animal studies of cauda equina injury have primarily used rat models, which display significant differences from humans. Furthermore, most studies have focused on electrophysio- logical examination. To better mimic the outcome after surgical repair of cauda equina injury, a novel animal model was established in the goat. Electrophysiological, histological and magnetic resonance imaging methods were used to evaluate the morphological and functional outcome after cauda equina injury and end-to-end suture. Our results demonstrate successful establish- ment of the goat experimental model of cauda equina injury. This novel model can provide detailed information on the nerve regenerative process following surgical repair of cauda equina injury.

In resonant grounding systems,most single-phaseto-ground faults evolve from IAFs(Intermittent Arc Faults).Earlier detection of IAFs can facilitate fault avoidance.This work proposes a novel method based on machine learning for detecting IAFs in three steps.First,the feature of zero-sequence current is automatically extracted and selected by a newlydesigned FINET(“For IAFs,Neuron Elaboration Net”),instead of traditional feature selection based on time-frequency decomposition.Moreover,data of the zero-sequence current divided by different time windows are successively input into the trained FINET.A proposed PSF(principal-subordinate factor)analyses the results obtained from FINET to improve anti-interference in the mentioned IAF detection algorithm.Experiments using PSCAD/EMTDC software simulation data show the proposed method is feasible and highly adaptable.In addition,the detection result of on-site recorded data demonstrates the effectiveness of the proposed method in practical resonant grounding systems.

Background: Arteriovenous malformation(AVM) have long-term "blood stealing" characteristics, which result in complicated hemodynamic features. To analyze the application of intraoperative indocyanine green angiography with FLOW 800 software in AVM surgeries.Methods: Data on 17 patients undergoing surgery with ICG fluorescence were collected in Beijing tiantan Hospital. To analyze the hemodynamic features of AVM and the influence on the peripheral cortex of AVM resection, we assessed the following hemodynamic parameters: maximum intensity, slope of rise, time to half-maximal fluorescence, and transit time from arteries to veins.Results: In the 17 superficial AVMs studied, the time-delay color mode of the FLOW 800 software was superior to the traditional playback mode for identifying feeding arteries, draining veins, and their relation to normal cortical vessels. The maximum fluorescence intensity and slope of the ICG fluorescence curve of feeder arteries and draining veins were higher than those of normal peripheral vessels (P 0.05).Conclusions: Hemodynamic parameter analysis provided quality guidance for the resection of AVMs and could also be used in estimating changes in blood flow in the local cortex to identify abnormal hyperperfusion and residual nidus.
