The effects of borneol on the pharmacokinetics and brain distribution of tanshinone IIA,salvianolic acid B and ginsenoside Rg1 in Fufang Danshen preparation in rats

Fufang Danshen preparation(FDP)is consisted of Salviae Miltiorrhizar Radix et Rhizoma(Danshen),Notoginseng Radix et Rhizoma(Sanqi)and Borneolum Syntheticum(borneol).FDP is usually used to treat myocardial ischemia hypoxia,cerebral ischemia and alzheimer’s disease,*** the treatment of cerebrovascular diseases,borneol is usually used to promote the absorption and distribution of the bioactive components to proper organs,especially to the *** purpose of this study is investigating the effects of borneol on the pharmacokinetics and brain distribution of tanshinone IIA(TS IIA),salvianolic acid B(SAB)and ginsenoside Rg1 in *** healthy Sprague-Dawley(SD)rats were given Danshen extracts,Sanqi extracts(Panax notoginseng saponins)or simultaneously administered Danshen extracts,Sanqi extracts and *** and brain samples were collected at different points in *** concentration of TS IIA,SAB and Rg1 was determined by UPLC-MS/MS *** main pharmacokinetics parameters of plasma and brain tissue were calculated by using Phoenix WinNolin 6.1 *** comparison with Danshen and Sanqi alone,there were significant differences in pharmacokinetic parameters of TS IIA,SAB and Rg1,and the brain distribution of SAB and TS IIA when Danshen,Sanqi and borneol were administrated *** statistically significant shortened tmax of TS IIA,SAB and Rg1 in plasma and brain,increased the bioavaiability of Rg1,inhibited metabolism of Rg1 and enhanced the transport of TS IIA and SAB to *** results indicated that borneol could affect the multiple targets components and produce synergistic *** accelerating the intestinal absorption and brain distribution,borneol caused the effective ingredients of Danshen and Sanqi to play a quicker therapeutic role and improved the therapeutic effect.

Association between uveitis and psoriatic disease: a systematic review and Meta-analysis based on the evidence from cohort studies

AIM: To conduct a systematic review and Meta-analysis to examine the association between uveitis and psoriatic disease, and to evaluate whether one condition predisposes individuals to the other. METHODS: We performed a comprehensive search of Pub Med and EMBASE to identify cohort studies examining the association between uveitis and psoriatic disease [psoriasis and/or psoriatic arthritis(Ps A)]. We used a random-effects model to calculate the pooled relative risks(RRs) adjusted for confounders, along with the 95% confidence intervals(CIs). RESULTS: This Meta-analysis included a total of 6 studies and a maximum of 80 178 648 participants. Compared with non-psoriatic controls, uveitis risk was significantly elevated in patients with psoriasis(RR=1.49;95%CI: 1.08-2.07), and Ps A(RR=3.16;95%CI: 2.16-4.63). Furthermore, pre-existing uveitis was associated with a significantly increased risk of psoriasis(RR=1.62;95%CI: 1.44-1.83), and Ps A(RR=4.44;95%CI: 3.52-5.60).CONCLUSION: The results of this systematic review and Meta-analysis suggest an overall positive bidirectional association between uveitis and psoriatic disease(psoriasis and Ps A), warranting increased awareness among clinicians involved in the management of these two ***, there remains a need for more detailed studies of the possible common pathogenesis of psoriatic disease and uveitis.

Akt aberrant activation accelerated tumor development and metastasis,and our previous study identified silent information regulator 6(SIRT6)as a novel and critical tumor suppressor in colorectal cancer(CRC)downstream of Akt.1 However,the function of SIRT6 in cancer progression has not been fully *** the present study,we found that except for inducing apoptosis,SIRT6 could serve as a crucial regulator to initiate autophagy by directly interacting with and activating *** importantly,we also reported autophagy regulation in a SIRT6-dependent indirect manner,namely,SIRT6 competitively bound to PUMA,which led to the release of *** note,autophagy inhibition enhanced the pro-apoptotic effect of Midostaurin both in vitro and in ***,SIRT6 induced apoptosis accompanied by protective autophagy,and SIRT6 promoted autophagy by directly interacting with ULK1 and competitive binding to *** new insight identified the dual function of SIRT6 on apoptosis and autophagy,which has attractive clinical applications in cancer therapy.

Plasma levels of omentin-1 and visfatin in senile patients with coronary heart disease and heart failure

Objective:To investigate the alteration of plasma levels of omentin-1 and visfatin in elderly patients with coronary heart disease(CHD)and heart ***:Plasma omentin-1 and visfatin levels were measured in 90 subjects(29 stable angina pectoris(SAP)cases,30 unstable angina pectoris(UAP)cases and 31 age-and sex-matched healthy controls(age≥60 years)by enzyme-linked immunosorbent assay *** to the New York Heart Association classification.59 CHDs were divided into three groups:functional I class,11 cases;functional II/III class,36 cases;and functional IV class,12 ***:The plasma level of omentin-1in CHO patients was significantly lower than thai of the control ***-lin SAP group and UAP group were significantly lower compared to the control group(there was no statistical significance between UAP group and SAP group;P>0.05).The plasma level of visfatin in CHD patients was significandy higher than that of the control ***,visfatin in SAP group and UAP group were all significantly higher compared to the control group,while there was no statistical significance between UAP group,and SAP *** plasma omentin-1 level was negatively correlated with SBP(r=-0.264,P<0.05),positively correlated with HDL-c level(r=0.271,P<0.05);the plasma visfatin level was positively correlated with TC(r=0.292,P<0.05),negatively correlated with HDL-c level(r=-0.266,P<0.05).There was a negative correlation between plasma omentin-1 and visfatin levels(r=-0.280,P<0.05).Moreover,multiple linear stepwise regression analysis showed that omentin-1 and visfatin levels might be affected by HDL-c *** regression analysis showed that visfatin could be an independent risk factor of ***:Decreased levels of omentin-1 and increased levels of visfatin may be involved in the occurrence and development of ***-1 and visfatin,independently,may be protective and pro-inflammatory ***,both omentin-1 and visfatin may

Ototoxic destruction by co-administration of kanamycin and ethacrynic acid in rats

It is well known that ethacrynic acid (EA) can potentiate the ototoxicity of aminoglycoside antibiotics (AmAn) such as kanamycin (KM),if they were applied at the same ***,to create the model of EA-KMinduced cochlear lesion in rats,adult rats received a single injection of EA (75 mg/kg,intravenous injection),or followed immediately by KM (500 mg/kg,intramuscular injection).The hearing function was assessed by auditory brainstem response (ABR) measurement in response to click and/or tone bursts at 4,8,12,16,20,24,and 32 *** static microcirculation status in the stria vascularis after a single EA injection was evaluated with eosin *** pathological changes in cochlear and vestibular hair cells were also quantified after co-administration of EA and *** a single EA injection,blood flow in vessels supplying the stria vascularis rapidly ***,the blood supply to the cochlear lateral wall partially recovered 5 h after EA *** changes in ABR were basically parallel to the microcirculation changes in stria vascularis after single EA ***,disposable co-administration of EA and KM resulted in a permanent hearing loss and severe damage to the cochlear hair cells,but spared the vestibular hair *** the cochlear lateral wall is the important part of the blood-cochlea barrier,EA-induced anoxic damage to the epithelium of stria vascularis may enhance the entry of KM to the ***,experimental animal model of selective cochlear damage with normal vestibular systems can be reliably created through co-administration of EA and KM.

Predication of plasma concentration of remifentanil based on Elman neural network

Due to the nature of ultra-short-acting opioid remifentanil of high time-varying,complex compartment model and low-accuracy of plasma concentration prediction,the traditional estimation method of population pharmacokinetics parameters,nonlinear mixed effects model(NONMEM),has the abuses of tedious work and plenty of man-made jamming *** Elman feedback neural network was *** relationships between the patients’plasma concentration of remifentanil and time,patient’age,gender,lean body mass,height,body surface area,sampling time,total dose,and injection rate through network training were obtained to predict the plasma concentration of remifentanil,and after that,it was compared with the results of NONMEM *** conclusion,the average error of Elman network is 6.34%,while that of NONMEM is 18.99%.The absolute average error of Elman network is 27.07%,while that of NONMEM is 38.09%.The experimental results indicate that Elman neural network could predict the plasma concentration of remifentanil rapidly and stably,with high accuracy and low *** the characteristics of simple principle and fast computing speed,this method is suitable to data analysis of short-acting anesthesia drug population pharmacokinetic and pharmacodynamics.

Integrated analysis of micro RNA and m RNA expression profiles in HBx-expressing hepatic cells

AIM To identify the miRNA-mRNA regulatory network in hepatitis B virus X (HBx)-expressing hepatic cells. METHODS A stable HBx-expressing human liver cell line L02 was established. The mRNA and miRNA expression profiles of L02/HBx and L02/pcDNA liver cells were identified by RNA-sequencing analysis. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis was performed to investigate the function of candidate biomarkers, and the relationship between miRNA and mRNA was studied by network analysis. RESULTS Compared with L02/pcDNA cells, 742 unregulated genes and 501 downregulated genes were determined as differentially expressed in L02/HBx cells. Gene ontology analysis suggested that the differentially expressed genes were relevant to different processes. Concurrently, 22 differential miRNAs were also determined in L02/HBx cells. Furthermore, integrated analysis of miRNA and mRNA expression profiles identified a core miRNA-mRNA regulatory network that is correlated with the carcinogenic role of HBx. CONCLUSION Collectively, the miRNA-mRNA network-based analysis could be useful to elucidate the potential role of HBx in liver cell malignant transformation and shed light on the underlying molecular mechanism and novel therapy targets for hepatocellular carcinoma.
