Zinc finger protein 831 promotes apoptosis and enhances chemosensitivity in breast cancer by acting as a novel transcriptional repressor targeting the STAT3/Bcl2 signaling pathway

Emerging evidence suggested that zinc finger protein 831(ZNF831)was associated with immune activity and stem cell regulation in breast cancer.Whereas,the roles and molec-ular mechanisms of ZNF831 in oncogenesis remain unclear.ZNF831 expression was significantly diminished in breast cancer which was associated with promoter CpG methylation but not mu-tation.Ectopic over-expression of ZNF831 suppressed breast cancer cell proliferation and col-ony formation and promoted apoptosis in vitro,while knockdown of ZNF831 resulted in an opposite phenotype.Anti-proliferation effect of ZNF831 was verified in vivo.Bioinformatic analysis of public databases and transcriptome sequencing both showed that ZNF831 could enhance apoptosis through transcriptional regulation of the JAK/STAT pathway.ChiP and luciferase report assays demonstrated that ZNF831 could directly bind to one specific region of STAT3 promoter and induce the transcriptional inhibition of STAT3.As a result,the attenuation of STAT3 led to a restraint of the transcription of Bcl2 and thus accelerated the apoptotic progression.Augmentation of STAT3 diminished the apoptosis-promoting effect of ZNF831 in breast cancer cell lines.Furthermore,ZNF831 could ameliorate the anti-proliferation effect of capecitabine and gemcitabine in breast cancer cell lines.Our findings demonstrate for the first time that ZNF831 is a novel transcriptional suppressor through inhibiting the expression of STAT3/Bcl2 and promoting the apoptosis process in breast cancer,suggesting ZNF831 as a novel biomarker and potential therapeutic target for breast cancer patients.

Geology and Geochemistry of K-feldspar Veins in Lamprophyre at the Zhenyuan Gold Deposit,Yunnan,Southwest China:Implications for Gold Mineralization

Lamprophyres typically appear in hydrothermal gold deposits.The relationship between lamprophyres and gold deposits is investigated widely.Some researchers suggest that the emplacement of lamprophyres triggers gold mineralization,whereas others hypothesize that the formation of lamprophyres increases the fertility of mantle sources and ore-forming fluids.K-feldspar veins,with ages between those of lamprophyres and gold deposits,appear in lamprophyres in Zhenyuan.Therefore,K-feldspar veins are ideal for investigating the relationship between lamprophyres and gold deposits.Phlogopite in K-feldspar veins has lower Mg#,Ni,and Cr contents and higher TiO2,Li,Ba,Sr,Sc,Zr,Nb,and Cs contents than phlogopite in lamprophyres.The in-situ Sr isotopic values of apatites(0.7063–0.7066)in K-feldspar veins are within the range for apatites(0.7064–0.7078)from lamprophyres.High large-ion lithophile element concentrations and low Nb and Ta concentrations in phlogopite from lamprophyres,in addition to high(87Sr/86Sr)i values of apatite(0.7064–0.7078),indicate that the magma parental to these phlogopite and apatite crystals is derived from an enriched mantle.Kfeldspar veins are genetically correlated with lamprophyres,whereas sulfide mineral assemblage and trace element compositions of pyrite in K-feldspar veins suggest that K-feldspar veins in lamprophyres are not directly related to gold mineralization of the Zhenyuan deposit.

Sea cucumber derived sulfated sterols alter glucose metabolism by promoting gluconeogenesis and reducing glycogenesis in healthy mice

Sea cucumber derived sulfated sterols (SS) significantly ameliorated insulin resistance and decreased lipid accumulation compared to plant sterols. Interestingly， our recent study found that intervention with sea cucumber sulfated sterols could significantly increase blood glucose levels of healthy mice in the presence of glucose， while cholesterol sulfate， as one of sulfated sterols， did not have the same effect. However， the exact mechanism of SS on glucose metabolism is still unknown. In the present study， we investigated the potential mechanism by which SS influenced blood glucose homeostasis in healthy mice. Results showed that intervention with sea cucumber sulfated sterols did not affect the levels of hormones related to glucose metabolism， while led to a significant decrease in the synthesis of liver glycogen and muscle glycogen. Besides， the expression of proteins associated with the promotion of gluconeogenesis dramatically increased in the mice intervened with sea cucumber sulfated sterols. These findings suggested that sea cucumber sulfated sterols might change blood glucose metabolism in healthy mice by reducing glycogenesis and promoting gluconeogenesis.

Hyperspectral remote sensing identification of marine oil emulsions based on the fusion of spatial and spectral features

Marine oil spill emulsions are difficult to recover,and the damage to the environment is not easy to eliminate.The use of remote sensing to accurately identify oil spill emulsions is highly important for the protection of marine environments.However,the spectrum of oil emulsions changes due to different water content.Hyperspectral remote sensing and deep learning can use spectral and spatial information to identify different types of oil emulsions.Nonetheless,hyperspectral data can also cause information redundancy,reducing classification accuracy and efficiency,and even overfitting in machine learning models.To address these problems,an oil emulsion deep-learning identification model with spatial-spectral feature fusion is established,and feature bands that can distinguish between crude oil,seawater,water-in-oil emulsion(WO),and oil-in-water emulsion(OW)are filtered based on a standard deviation threshold–mutual information method.Using oil spill airborne hyperspectral data,we conducted identification experiments on oil emulsions in different background waters and under different spatial and temporal conditions,analyzed the transferability of the model,and explored the effects of feature band selection and spectral resolution on the identification of oil emulsions.The results show the following.(1)The standard deviation–mutual information feature selection method is able to effectively extract feature bands that can distinguish between WO,OW,oil slick,and seawater.The number of bands was reduced from 224 to 134 after feature selection on the Airborne Visible Infrared Imaging Spectrometer(AVIRIS)data and from 126 to 100 on the S185 data.(2)With feature selection,the overall accuracy and Kappa of the identification results for the training area are 91.80%and 0.86,respectively,improved by 2.62%and 0.04,and the overall accuracy and Kappa of the identification results for the migration area are 86.53%and 0.80,respectively,improved by 3.45%and 0.05.(3)The oil emulsion identification model has a certain degree of trans

Tobacco smoking is a prevalent and detrimental habit practiced worldwide,increasing the risk of various diseases,including chronic obstructive pulmonary disease(COPD),cardiovascular disease,liver disease,and cancer.Although previous research has explored the detrimental health effects of tobacco smoking,recent studies suggest that gut microbiota dysbiosis may play a critical role in these outcomes.Numerous tobacco smoke components,such as nicotine,are found in the gastrointestinal tract and interact with gut microbiota,leading to lasting impacts on host health and diseases.This review delves into the ways tobacco smoking and its various constituents influence gut microbiota composition and functionality.We also summarize recent advancements in understanding how tobacco smoking-induced gut microbiota dysbiosis affects host health.Furthermore,this review introduces a novel perspective on how changes in gut microbiota following smoking cessation may contribute to withdrawal syndrome and the degree of health improvements in smokers.

The coronavirus disease 2019(COVID-19)pandemic,caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has been significantly alleviated.However,long-term health effects and prevention strategy remain unresolved.Thus,it is essential to explore the pathophysiological mechanisms and intervention for SARS-CoV-2 infection.Emerging research indicates a link between COVID-19 and bile acids,traditionally known for facilitating dietary fat absorption.The bile acid ursodeoxycholic acid potentially protects against SARS-CoV-2 infection by inhibiting the farnesoid X receptor,a bile acid nuclear receptor.The activation of G-protein-coupled bile acid receptor,another membrane receptor for bile acids,has also been found to regulate the expression of angiotensin-converting enzyme 2,the receptor through which the virus enters human cells.Here,we review the latest basic and clinical evidence linking bile acids to SARS-CoV-2,and reveal their complicated pathophysiological mechanisms.

Geochronological and geochemical constraints on the petrogenesis and geodynamic process of Hemler,Vlinder,and Il'ichev seamount lavas in NW Pacific

Oceanic intraplate volcanoes with linear age progressions are usually accepted to be derived from melting of an upwelling mantle plume.Several seamount groups in NW Pacific,however,show complex age-distance relationships that are difficult to explain using the classic“mantle plume hypothesis”,and thus their origins are controversial.In this study,we present ^(40)Ar-^(39)Ar age,geochemical,and Sr-Nd-Pb-Hf isotopic data of lavas from Hemler,Vlinder,and Il’ichev seamounts in NW Pacific,to elucidate their petrogenesis and geodynamic process.The lavas from Hemler,Vlinder,and Il’ichev seamounts are classified as alkali basalt,basanite/nephelinite,and trachyte.Lavas with Mg O>8 wt.%exhibit high contents of CaO,FeO^(T),and TiO_(2),similar to the composition of melts formed from reaction between carbonated eclogite-derived melts and fertile peridotite.These lavas have elevated Zr/Hf ratios(40.6-45.2)and negative Zr and Hf anomalies,indicating the presence of a carbonate component in the mantle source.They are enriched in incompatible trace elements and have enriched mantle 1(EM1)-like SrNd-Pb-Hf isotopic compositions.The isotopic compositions of Vlinder,Il’ichev basanite,and Hemler lavas in this study are similar to the Rarotonga hotspot.Although occurring at the same seamount,the Il’ichev alkali basalts display more depleted SrNd-Hf isotopic compositions compared to Il’ichev basanite.According to plate tectonic reconstruction results,the ages of Hemler(100.1 Ma),Vlinder pre-(100.2 Ma)and post-shield(87.5 Ma),and Il’ichev(56.4 Ma)lavas clearly deviate from the Macdonald,Arago,Rarotonga,and Samoa hotspot tracks,indicating that they cannot directly originate from mantle plumes.We propose that in the mid-Cretaceous,when the Pacific plate passed over Rarotonga hotspot,melting of Rarotonga plume formed the Vlinder(main-shield stage),Pako,and Ioah seamounts.The Rarotonga(and possibly Samoa)plume materials would have been dispersed into the surrounding asthenosphere by mantle convection.These diffuse plume materials would underg

Physical mechanism of the rapid increase in intense and long-lived extreme heatwaves in the Northern Hemisphere since 1980

Since 1980,both the intensity and duration of summer heatwaves in the middle and high latitudes of the Northern Hemisphere have significantly increased,leading this region to become a critical area for a significant increase in the frequency of intense and long-lived extreme heatwaves.We found that stronger and more persistent high-pressure systems and lower soil moisture before the events were the main drivers of intense and long-lived extreme heatwaves in western Europe and the middle and high latitudes of North America.However,in eastern Europe and Siberia,lower cloud cover before events is also a main driver of this type of extreme heatwave,in addition to the above drivers.These factors are coupled with each other and can change heatwave intensity and duration by influencing surface radiation processes during events.Using the self-organizing map classification method,we found that 6 weather patterns with increased frequency,intensity,and duration were the main dynamic reasons leading to the increase in intense and long-lived extreme heatwaves after 1980.In addition,the decrease in summer average soil moisture in most areas of the mid-high latitudes of the Northern Hemisphere and the decrease in average cloud cover in eastern Europe and Siberia are found to be the main thermodynamic reasons leading to the increase in these extreme heatwaves.

Objective YAP1 plays a dual role as an oncogene and tumor suppressor gene in several tumors;differentiating between these roles may depend on the YAP1 phosphorylation pattern.The specific function of YAP1 in B cell acute lymphoblastic leukemia(B-ALL),however,is currently unclear.Thus,in the present study,the role of YAP1 in B-ALL was investigated using relevant cell lines and patient datasets.Methods The effects of shRNA-mediated knockdown on YAP1 and LATS1 levels in the NALM6 and MOLT-4 cell lines were examined using Western blotting,quantitative real-time polymerase chain reaction,flow cytometry,immunostaining,and nude mouse subcutaneous tumorigenesis experiments.Gene expression levels of Hippo pathway-related molecules before and after verteporfin(VP)treatment were compared using RNA-Seq to identify significant Hippo pathway-related genes in NALM6 cells.Results Patients with ALL showing high YAP1 expression and low YAP1-Ser127 phosphorylation levels had worse prognoses than those with low YAP1 protein expression and high YAP1-Ser127 phosphorylation levels.YAP1-Ser127 phosphorylation levels were lower in NALM6 cells than in MOLT-4 and control cells;YAP1 was distributed in the nuclei in NALM6 cells.Knockdown of YAP1 inhibited MOLT-4 and NALM6 cell proliferation and arrested the NALM6 cell cycle in the G0/G1 phase.Before and after VP treatment,the expression of the upstream gene LATS1 was upregulated;its overexpression promoted YAP1-Ser127 phosphorylation.Further,YAP1 was distributed in the plasma.Conclusion LATS1 may downregulate YAP1-Ser127 phosphorylation and maintain B-ALL cell function;thus,VP,which targets this axis,may serve as a new therapeutic method for improving the outcomes for B-ALL patients.
