Coronavirus disease 2019 (COVID-19), which is caused by severe acute respiratory syndrome coronavirus 2 (SARS- CoV-2), is a life-threatening disease, especially in elderly individuals and those with comorbidities. The predominant clinical manifestation of COVID-19 is respiratory dysfunction, while neurological presentations are increasingly being recognized. SARS-CoV-2 invades host cells primarily via attachment of the spike protein to the angiotensin-converting enzyme 2 (ACE2) receptor expressed on cell membranes. Patients with Alzheimer’s disease (AD) are more susceptible to SARS-CoV-2 infection and prone to severe clinical outcomes. Recent studies have revealed some common risk fac-tors for AD and COVID-19. An understanding of the association between COVID-19 and AD and the potential related mechanisms may lead to the development of novel approaches to treating both diseases. In the present review, we first summarize the mechanisms by which SARS-CoV-2 invades the central nervous system (CNS) and then discuss the associations and potential shared key factors between COVID-19 and AD, with a focus on the ACE2 receptor, apolipo-protein E (APOE) genotype, age, and neuroinflammation.

Deacetylation of TFEB promotes fibrillar Aβ degradation by upregulating lysosomal biogenesis in microglia

Microglia play a pivotal role in clearance of Aβ by degrading them in lysosomes, countering amyloid pla- que pathogenesis in Alzheimer＇s disease （AD）. Recent evidence suggests that lysosomal dysfunction leads to insufficient elimination of toxic protein aggregates. We tested whether enhancing lysosomal function with transcription factor EB （TFEB）, an essential regulator modulating lysosomal pathways, would promote Aβ clearance in microglia. Here we show that microglial expression of TFEB facilitates fibrillar Aβ （fAβ） degra- dation and reduces deposited amyloid plaques, which are further enhanced by deacetylation of TFEB. Using mass spectrometry analysis, we firstly confirmed acetylation as a previously unreported modification of TFEB and found that SIRT1 directly interacted with and deacetylated TFEB at lysine residue 116. Subsequently, SIRT1 overexpression enhanced lysosomal function and fAβ degradation by upregulating transcriptional levels of TFEB downstream targets, which could be inhibited when TFEB was knocked down. Furthermore, overexpression of deacetylated TFEB at K116R mutant in microglia accelerated intracellular fAβ degradation by stimulating lysosomal biogenesis and greatly reduced the deposited amyloid plaques in the brain slices of APPIPS1 transgenic mice. Our findings reveal that deacetylaUon of TFEB could regulate lysosomal biogenesis and fAβ degradation, making microglial activation of TFEB a possible strategy for attenuating amyloid plaque deposition in AD.

The effect of frequency and sample shape on fatigue behaviors of DZ125 superalloy are systematically *** results show that fatigue fracture still occurs above the cycle of 10~8 for tests carried out at the frequency of f=20 kHz and stress ratio R =-1,so the traditional fatigue limit at cycle of 10~7 is not appropriate for fatigue *** fracture surfaces are perpendicular to stress axis for cylindrical and plate specimens,and the fatigue cracks originate from the extra surface of the *** crack is apt to propagate from cutting direction to forward direction,which occurs mainly in the second propagation stage at higher stress amplitude. There is an obvious frequency effect for DZ125 *** higher the test frequency is,the more serious the effect of frequency on fatigue behaviors of the *** the frequency correction,the ultra-high cycle fatigue S-N curve well coincide with the traditional fatigue S-N curve.

Manufacture technique of bronze-iron bimetallic objects found in M27 of Liangdaicun Site, Hancheng, Shaanxi

Analysis of the fabrication technology of a bronze knife with an iron blade and a bronze Ge with an iron blade, two copper-iron bimetallic wares, unearthed in M27 of Liangdaicun Site, Hancheng of Shaanxi, is performed in this paper by using metallographic, EPMA and AMS-14C dating methods. The micro-structures of the two samples are typical wrought bloomery iron containing a substantial amount of carbon, which is also called carburized steel, made from bloomery iron by cementation in the solid state. The objects can be dated back to the early Spring and Autumn period. This study provides new evidence for understanding the beginning of iron smelting in China. Most of the early known iron wares of the period between the late Western Zhou Dynasty and the early Spring and Autumn were unearthed in the region at the junction of Henan, Shanxi and Shaanxi, at the middle reaches of the Yellow River, suggesting that this region may likely be one of the earliest centers of iron smelting technology in China and deserves further archaeological research. As early iron products were also discovered in the area of the upper reaches of the Yellow River and in Xinjiang, appropriate attention also should be paid to the relationship between these two areas in terms of the origin of iron smelting.

The age-dependent peculiarities of stimulation of free radical processes in subcellular fractions of skeletal muscle of rats subjected to long-term immobilization stress were studied in order to improve knowledge about changes of muscular tissue during ontogenesis. It is found that adult animals do not show accumulation of proteins carbonyls, TBA-reactive substances, and Schiff bases in subcellular fractions of the thigh muscle when immobUized. Long-term immobilization causes apparent manifestation of oxidative stress only in mitochondrial fraction in pubertal rats. Mitochondrial oxidative stress defense systems are sufficiently effective, however, direction of pathways of free radical oxidation carbonyl products catabolism alters in the cytoplasm of myocytes in old rats under long-term immobilization conditions.

Sphingosine-1-phosphate(S1P)is a biologically active *** homolog 2(Spns2)acts as an S1P transporter in zebrafish and mice regulating heart development and lymphocyte trafficking *** mechanism of action of Spns2 is still elusive in mammals especially considering the multiple roles of S1P ***,we demonstrate that Spns2-deficient mice rapidly lost auditory sensitivity and endocochlear potential(EP)from 2 to 3weeks ***2 is expressed in both the organ of Corti and lateral wall of the *** found progressive degeneration of sensory hair cells in the organ of Corti,but the earliest defect was a decline in the EP suggesting that dysfunction of the lateral wall was the primary *** the lateral wall of adult mutants,we observed structural changes of marginal cell boundaries and of strial capillaries,and reduced expression of several key proteins involved in the generation of EP(Kcnj10,Kcnq1,Gjb2 and Gjb6),but these changes were likely to be *** of the boundaries of the striavascularis and of the strial capillaries appeared *** inactivation of Spns2 in red blood cells,platelets,or lymphatic or vascular endothelial cells did not affect hearing,but targeted ablation of Spns2 in the cochlea using a Sox10-Cre allele produced a similar auditory phenotype to the original *** study suggests that local Spns2 expression is critical for hearing in *** findings indicate that Spns2 and S1P signalling are required for normal maintenance of the EP and hence for normal auditory function.
