The number of ureterorenoscopic procedures for upper urinary tract stone management has increased dramatically during recent years worldwide[1].Developments in flexible ureteroscope and laser technology have made it possible to successfully address larger and more complex stone scenarios retrogradely.On the other hand,this means that more and more patients are exposed to the potential adverse effects of ureteroscopy[2].In general,ureteroscopy is considered a safe procedure.However,serious complications and even deaths do occur,and these events are most likely underreported[3,4].

Animal manure consists of predominantly urine and feces,but also may contain bedding materials,dropped feed,scurf,and other farming wastes.Manure is typically applied to soils as fertilizer for agricultural production.The estimated amount of manure produced in 12 major livestock-producing countries is 9 × 109 Mg of manure annually.Manure is rich in plant nutrients.However,manure is also considered as an environmental pollutant when it is over-applied to cropland or following runoff into surface water.Manure can also in?uence global climate change via emissions of methane（CH4） and nitrous oxide（N2O）.Thus,increased and updated knowledge of applied and environmental chemistry of animal manure is needed to shed light on the research and development of animal manure utilization and minimization of its adverse environmental concerns.The advances in basic and applied studies of manure major components,organic matter,phosphorus,and nitrogen,primarily related to US livestock production are summarized in this review.Detailed focus was placed on three notable challenges for future manure research:1) soil application of animal manure,2)manure phytate phosphorus,and 3) manure nitrogen availability.This review may contribute to the global effort in sustainable and environmentally sound agriculture by stimulating new ideas and directions in animal manure research,and promoting application of knowledge and insight derived from manure research into improved manure management strategies.

Plant b-1,3-glucanases are members of the pathogenesis-related protein 2（PR-2） family,which is one of the 17 PR protein families and plays important roles in biotic and abiotic stress responses.One of the differentially expressed proteins（spot 842） identified in a recent proteomic comparison between five pairs of closely related maize（Zea mays L.） lines differing in aflatoxin resistance was further investigated in the present study.Here,the corresponding cDNA was cloned from maize and designated as ZmGns.ZmGns encodes a protein of338 amino acids containing a potential signal peptide.The expression of Zm Gns was detectible in all tissues studied with the highest level in silks.ZmGns was significantly induced by biotic stresses including three bacteria and the fungus Aspergillus flavus.ZmGns was also induced by most abiotic stresses tested and growth hormones including salicylic acid.In vivo,ZmGns showed a significant inhibitory activity against thebacterial pathogen Pseudomonas syringae pv.tomato DC3000 and fungal pathogen Botrytis cinerea when it overexpressed in Arabidopsis.Its high level of expression in the silk tissue and its induced expression by phytohormone treatment,as well as by bacterial and fungal infections,suggest it plays a complex role in maize growth,development,and defense.

Feasibility study of assessing cotton fiber maturity from near infrared hyperspectral imaging technique

Background Fiber maturity is a key cotton quality property,and its variability in a sample impacts fiber processing and dyeing performance.Currently,the maturity is determined by using established protocols in laboratories under a controlled environment.There is an increasing need to measure fiber maturity using low-cost(in general less than $20000)and small portable systems.In this study,a laboratory feasibility was performed to assess the ability of the shortwave infrared hyperspectral imaging(SWIR HSI)technique for determining the conditioned fiber maturity,and as a comparison,a bench-top commercial and expensive(in general greater than $60000)near infrared(NIR)instrument was used.Results Although SWIR HSI and NIR represent different measurement technologies,consistent spectral characteristics were observed between the two instruments when they were used to measure the maturity of the locule fiber samples in seed cotton and of the well-defined fiber samples,respectively.Partial least squares(PLS)models were established using different spectral preprocessing parameters to predict fiber maturity.The high prediction precision was observed by a lower root mean square error of prediction(RMSEP)(0.518),and greater percentage(97.0%)of samples within the 95% agreement range in the entire NIR region(1000-2500 nm)without the moisture band at 1940 nm.Conclusion SWIR HSI has a good potential for assessing cotton fiber maturity in a laboratory environment.

Improving nitrogen management for zero-tillage rice in China

Zero-tillage has become increasingly attractive in rice production in China.This study was conducted to determine the feasibility of two possible improved N management practices with fewer N applications in zero-tillage rice:(1)two split applications of urea at75 kg N ha^(-1)at mid-tillering and 45 kg N ha^(-1)at panicle initiation(U_(120–2)),and(2)a single application of cross-linked polyacrylamide-coated urea(a slow-release fertilizer)at midtillering at a rate of 150 kg N ha^(-1)(PCU_(150–1)).Three field experiments were conducted to compare grain yield and N-use efficiency among several N treatments:a zero-N control(CK),U_(120–2),PCU_(150–1),a single application of urea at mid-tillering at a rate of 150 kg N ha^(-1)(U_(150–1)),and a commonly recommended N management practice for conventional tillage rice(three split applications of urea with 75 kg N ha^(-1)as basal,30 kg N ha^(-1)at mid-tillering,and 45 kg N ha^(-1)at panicle initiation)(U_(150–3)).Treatments with N application(U_(120–2),PCU_(150–1),U_(150–1),and U_(150–3))produced 1.08–3.16 t ha^(-1)higher grain yields than CK.Grain yields under both U_(120–2)and PCU_(150–1)were comparable to that in U_(150–3).Recovery efficiency of N(RE_N),agronomic N-use efficiency(AE_N)and partial factor productivity of applied N(PFP_N)were increased under U_(120–2)and were similar under PCU_(150–1)to those under U_(150–3).U_(150–1)showed lower grain yield,RE_N,AE_N,and PFP_Nthan U_(150–3).These results suggest that U_(150–3)can be replaced with U_(120–2)to achieve both an increase in N-use efficiency and a reduction in number of N applications and or by PCU_(150–1)to achieve a maximum reduction in number of N applications in zero-tillage rice production in China.

This study was conducted to identify the factors associated with high grain yield in single seedling machine-transplanted hybrid rice under dense planting conditions. Field experiments were done in Yong＇an Town, Hunan Province, China in 2015 and 2016. Two hybrid rice cultivars were grown under single seedling machine transplanting （SMT） and conventional machine transplanting （CMT） at a high planting density in each year. Grain yield and yield attributes were compared between SMT and CMT. Averaged across cultivars and years, grain yield was 12％ higher under SMT than under CMT. Plant height, basal stem width, and shoot and root dry weights were higher in seedlings for SMT than those for CMT. SMT had less maximum tiller number per m2 and consequently less panicle number per m2 than did CMT. Branch number per panicle, especially the secondary branch number per panicle, and spikelet number per cm of panicle length were more under SMT than under CMT, which resulted in more spikelet number per panicle under SMT than under CMT. SMT had higher or equal spikelet filling percentage than did CMT. The difference in grain weight between SMT and CMT was relatively small and inconsistent cross years. SMT had higher or equal total biomass and harvest index than did CMT. Dry weight per stem under SMT was heavier than that under CMT. Larger leaf area per stem was partly responsible for the heavier dry weight per stem under SMT than under CMT. Our study suggests that improvement in seedling quality, panicle size, and dry weight per stem are critical to the high grain yield in single seedling machine-transplanted hybrid rice under dense planting conditions.

Editing of Rice Isoamylase Gene ISA1 Provides Insights into Its Function in Starch Formation

Isoamylase 1(ISA1) is an isoamylase-type debranching enzyme which plays a predominant role in amylopectin synthesis. In this study, the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated endonuclease 9(CRISPR/Cas9) system was used to edit ISA1 gene in rice via Agrobacterium-mediated transformation. We identified 36 genetic edited lines from 55 T_0 transgenic events, and classified the mutation forms into 7 types. Of those, two homozygous mutants, cr-isa1-1(type 1, with an adenine insertion) and cr-isa1-2(type 3, with a cytosine deletion) were selected for further analysis. Seed sizes of both cr-isa1-1 and cr-isa1-2 were affected, and the two mutants also displayed a shrunken endosperm with significantly lower grain weight. Electron microscopy analysis showed that abnormal starch granules and amyloplasts were found in cr-isa1-1 and cr-isa1-2 endosperm cells. The contents of total starch, amylose and amylopectin in the endosperm of the cr-isa1 mutants were significantly reduced, whereas sugar content and starch gel consistency were observably increased compared to the wild-type. The gelatinization temperature and starch chain length distributions of the cr-isa1 mutants were also altered. Moreover, transcript levels of most starch synthesis-related genes were significantly lower in cr-isa1 mutants. In conclusion, the results indicated that gene edition of ISA1 affected starch synthesis and endosperm development, and brought potential implications for rice quality breeding.

Identification of a soluble phosphatidate phosphohydrolase in the developing cotyledons of <i>Momordica</i><i>charantia</i>

Phosphatidate phosphatase (EC 3.***.3.4), PAP, catalyzes the dephosphorylation of phosphatidate (PtdOH) to form diacylglycerol. In eukaryotes, PAP driven reaction is the committed step in the synthesis of triacylglycerol. A Mg2+ independent PAP activity was identified in the soluble extract of Momordica charantia cotyledons undergoing maturation. While the microsomal fraction of the extract gave only 10% of the PAP activity, the remaining 90% of the activity was associated with the soluble fraction. At pH 3.0, the soluble PAP was bound to S column and eluted with glycine-HCl buffer containing high salt. The pH and temperature optima of the PAP activity were 6.0 and 53℃, respectively. Under optimum assay condition, the Vmax and Km for dioleoyl phosphatidic acid were 1.89 ηkat/mg of protein and 142 μM, respectively. For the synthetic substrate, ρ-nitrophenylphosphate, ρ- NPP, the Vmax and Km were 10.4 ηkat/mg of protein and 107 μM, respectively. The inclusion of Mg2+ and β-mercaptoethanol into the reaction mix did not change the enzyme activity nor did the addition of N-ethylmaleimide and phenylglyoxal, which indicates that cysteine and arginine are not involved in catalysis of PtdOH. The addition of Mg2+ up to 10 mM also did not change the level of PAP activity. Triton X-100, however, inhibited the activity. This is the first documented case of an in vitro PAP activity in the developing cotyledons of Momordica charantia. The PAP described here could serve as a model for lipin-1 or lipin-2 in humans. Mutations in these genes lead to acute myoglobinuria in human infants.

Amino Compounds in Poultry Litter,Litter-Amended Pasture Soils and Grass Shoots

Amino compounds(ACs),i.e.,amino acids and amino sugars,are the major forms of organic nitrogen(N)in animal manure and soil.To increase our understanding on the effect of long-term poultry litter(PL)application on soil AC pools and turnover,in this study,we determined the contents of 21 ACs in 23 PL samples,15 soil samples collected from 0–20,20–40,and 40–60 cm layers of five pasture plots with 0,5,10,15 and 20 years of PL applications,and 5 grass shoot samples grown on these pasture fields.The contents of 21 ACs were simultaneously determined by methanesulfonic acid hydrolysis/extraction and anion chromatography-pulsed amperometry.PL application increased soil total and individual AC contents with a distribution pattern similar to that of AC in PL.The highest AC-N concentrations were observed in the soils with 10-or 15-year PL applications,inconsistent with the order of annual application rates or cumulative applied PL amounts.Application of PL increased the AC contents in grass shoots whereby the highest increase of most ACs was with the shoots from the fields that received PL for 5 years.These observations suggested that both freshly applied and residual PL had contributions to the soil AC-N,and that PL application also accelerated AC-N transformation in soil.
